    Nutrition-induced Changes of Growth from Birth to First Calving and Its Impact on Mammary Development and First-lactation Milk Yield in Dairy Heifers: A Review

    This review focuses on the nutritional effects from birth until age at first calving on growth, mammary developmental changes, and first-lactation milk yield in heifer calves. The advancement in the genetic potential and the nutritional requirements of the animals has hastened the growth rate. Genetic selection for high milk yield has suggested higher growth capacity and hence increasing nutritional inputs are required. Rapid rearing by feeding high energy or high concentrate diets not only reduces the age of sexual maturity but also lowers the time period of attaining the age of first calving. However, high energy diets may cause undesirable fat deposition thereby affecting future milk yield potential. Discrepancies exist whether overfed or overweight heifers at puberty can influence the mammary development and future milk yield potential and performance. The data on post-pubertal nutritional management suggested that body weight at calving and post-pubertal growth rate is important in first lactation milk yield. There is a continuous research need for strategic feeding that accelerates growth of dairy heifers without reduction in subsequent production. Nutritional management from birth, across puberty and during pregnancy is critical for mammary growth and for producing a successful cow. This review will mostly highlight studies carried out on dairy breeds and possible available opportunities to manipulate nutritional status from birth until age at first calving

    Situational Analysis of the Upper Bhima sub-basin in the context of the Water-Food-Biodiversity Nexus

    This report was prepared as part of a research project titled “Soft systems analysis: Streamlining participatory approaches and agent-based models to explore ideas of fairness at the food-water-biodiversity (F-W-B) nexus (fairSTREAM)”. This project is funded by the International Institute for Applied System Analysis (IIASA) and is implemented in collaboration with two partners from India: Society for Promoting Participative Ecosystem Management (SOPPECOM) and Indian Institute of Science Education and Research Pune (IISER Pune). The project started in September 2021, and it will run till December 2024. The main goal of fairSTREAM is to develop and demonstrate a co-production methodology for including equity and justice (fairness) alongside efficiency in developing sustainable policy options across the Water-Food-Biodiversity nexus (WFB nexus). The demonstration component is placed in the Upper Bhima basin, and the specific objectives here are to design and test a systems-informed stakeholder knowledge co-production process with the purpose of developing fair and sustainable policy options for the WFB nexus. The co-production process involves several steps, namely: the assessment phase (including preparatory work, problem framing, and exploration of options) and the action planning phase. An evaluation process is also embedded across the different steps and phases. This report summarizes the findings of the preparatory and problem-framing phase, to contextualize the WFB nexus in the Upper Bhima, and what specific challenges it raises, both from a sustainability, equity, and fairness perspective. Other than serving as a reference document for primary stakeholders - farmers, fishers, and forest-dependent communities to contextualize the WFB nexus in the Upper Bhima, this situational analysis is intended to inform future phases, including the development and co-production of options using a combination of modeling and soft approaches

    Comparison of Aviary, Barn and Conventional Cage Raising of Chickens on Laying Performance and Egg Quality

    This study intended to compare the productive performance of three different layer raising systems; conventional cage (CC), barn (BR) and aviary (AV). The AV is welfare bestowed housing that allows free locomotion for birds within the BR. The BR allows bird’s free locomotion inside BR but without multilevel structures. Both pullets and cockerels were housed together in both AV and BR, but only pullets in CC. Seventeen weeks old Lohmann Brown Lite (n = 800) pullets were housed in AV during this study. The same age layer pullets were simultaneously assigned to either at CC or BR to compare egg production performance with AV. The duration of experiment was 40 weeks (from 21st to 60th week). There were no remarkable differences in egg production, hen day egg production (HDEP) and average egg weight among three rearing systems. First 20 weeks (phase-1) average HDEP (%) of AV, CC, and BR were 85.9, 88.8, 87.1 and average egg weights (g) were 57.5, 59.9, and 56.9 respectively. Those of the remaining 20 weeks (phase-2) were 87.1, 87.9, 85.5 and 64.2, 63.0 62.1, respectively. Daily feed intakes (122 g, 110 g, 125 g); feed conversion ratio (2.4, 2.1, 2.5) and daily egg mass (53.9 g, 54.4 g, 52.8 g) data from AV, CC, and BR were not influenced significantly by the respective raising systems. Daily feed intake of layers in both AV (124 g) and BR (127 g) tended to be higher than that in CC (113 g) during phase-2. Overall, exterior egg quality (dirty and cracked eggs) in both phases was superior in BR compared with AV and CC, whereas CC generated intermediate results. This study indicated that the HDEP per se in AV and BR were not significantly different from that in CC. The study implied that the facility depreciation cost for AV and cost for increased feed intake in AV compared to CC are believed to be critical to evaluate the cost effectiveness of egg production in AV

    Effects of Feeding Levels of Starter on Weaning Age, Performance, Nutrient Digestibility and Health Parameters in Holstein Dairy Calves

    To evaluate the effects of feeding four different levels of starter in male Holstein dairy calves, a completely randomized study was conducted, using 28 calves with initial body weight of 40.5±2.4 kg. The animals were fed iso-nitrogenous starter and were weaned when they consumed 350, 500, 650 and 800 g/d of starter for 3 d consecutively. Starter and water were available ad-libitum throughout the experiment. Body weight at pre-weaning (less than 5 wk) and post-weaning (8 wk) was lower in calves that received 350 g/d of starter than in the other treatments (p0.05). Dry matter, organic matter and crude protein digestibilities were lower in calves that received 350 g/d of starter compared with other treatments (p0.05).Treatments had no significant effect on time of starting rumination, respiratory score, and days of drug administration for pneumonia. There were no meaningful differences in feces, fecal odor scores, body temperature, and days of drug administration for diarrhea among all treatments (p>0.05). Total dry matter intake at the end of experiment showed no significant difference among calves fed 600 and 800 g/d of starter, but calves fed 350 and 500 g/d of starter showed more dry matter (DM) intake than calves in the 600 and 800 g/d groups (p<0.05)

    The hypothesis of the study was that inhibition of PPARβ/δ increases glucose uptake and lactose synthesis in bovine mammary epithelial cells by reducing the expression of the glucose transporter mRNA destabiliser calreticulin. Three experiments were conducted to test the hypothesis using immortalised bovine mammary alveolar (MACT) and primary bovine mammary (PBMC) cells. In Experiment 1, the most effective dose to inhibit PPARβ/δ activity among two synthetic antagonists (GSK-3787 and PT-s58) was assessed using a gene reporter assay. In Experiment 2, the effect on glucose uptake and lactose synthesis was evaluated by measuring glucose and lactose in the media and expression of related key genes upon modulation of PPARβ/δ using GSK-3787, the synthetic PPARβ/δ agonist GW-501516, or a combination of the two in cells cultivated in plastic. In Experiment 3, the same treatments were applied to cells cultivated in Matrigel and glucose and lactose in media were measured. In Experiment 1 it was determined that a significant inhibition of PPARβ/δ in the presence or absence of fetal bovine serum was achieved with ≥ 1000 nm GSK-3787 but no significant inhibition was observed with PT-s58. In Experiment 2, inhibition of PPARβ/δ had no effect on glucose uptake and lactose synthesis but they were both increased by GW-501516 in PBMC. The mRNA abundance of PPARβ/δ target gene pyruvate dehydrogenase kinase 4 was increased but transcription of calreticulin was decreased (only in MACT cells) by GW-501516. Treatment with GSK-3787 did not affect the transcription of measured genes. No effects on glucose uptake or lactose synthesis were detected by modulation of PPARβ/δ activity on cells cultivated in Matrigel. The above data do not provide support for the original hypothesis and suggest that PPARβ/δ does not play a major role in glucose uptake and lactose synthesis in bovine mammary epithelial cells

    Replacement of Soybean Meal with Silkworm Meal In The Diets of White Leghorn Layers and Effects on Performance, Apparent Total Tract Digestibility, Blood Profile and Egg Quality

    This study elucidated the effects of replacing soybean meal with silkworm meal on production performance, apparent total tract digestibility of nutrients, blood profile, and egg quality traits in white leghorn layers. A total of 150 white leghorn laying birds (52 weeks of age) were randomly divided into fifteen replicate groups (n=10 per replicate), and reared on five experimental diets having three replicates allocated to each treatment group for a period of six weeks. Five iso-nitrogenous and iso-caloric diets (D) were utilized as; 0 (D1), 25 (D2), 50 (D3), 75 (D4) and 100% (D5) as replacement of soybean meal with silkworm meal in commercial layer rations. The weight of the bird, daily feed intake (g/day/bird), hen day production (%), average egg weight (g), and feed conversion ratio did not differ significantly (P>0.05) among the dietary groups. Apparent total tract digestibility of nutrients was not different in the control group compared to treatment groups. Blood profile and egg quality parameters also showed no significant (P>0.05) differences among dietary groups. The egg weight, albumen height, yolk weight and shell thickness were not affected (P>0.05) by dietary treatments. Based on our results, it could be concluded that silkworm meal can be effectively used as an alternative protein sourceto soybean meal without any adverse effects on the layers

    2,4-Thiazolidinedione in well-fed lactating dairy goats: II. Response to intra-mammary infection.

    In a prior experiment, treatment of goats with the putative PPARγ agonist 2,4-thiazolidinedione (2,4-TZD) ameliorated the response to intramammary infection without evidence of PPARγ activation. The lack of PPARγ activation was possibly due to deficiency of vitamin A and/or a poor body condition of the animals. Therefore, the present study hypothesized that activation of PPARγ by 2,4-TZD in goats supplemented with adequate amounts of vitamin A can improve the response to sub-clinical mastitis. Lactating goats receiving a diet that met National Research Council requirements, including vitamin A, were injected with 8 mg/kg BW of 2,4-TZD (n = 6) or saline (n = 6; control (CTR)) daily. Two weeks into treatment, all goats received Streptococcus uberis (IMI) in the right mammary gland. Blood biomarkers of metabolism, inflammation, and oxidative status plus leukocytes phagocytosis were measured. Mammary epithelial cells (MEC) and macrophages were isolated from milk and liver tissue collected for gene expression analysis. Milk fat was maintained by treatment with 2,4-TZD, but decreased in CTR, after IMI. Haptoglobin was increased after IMI only in 2,4-TZD without any effect on negative acute phase proteins, indicating an improved liver function. 2,4-TZD vs. CTR had a greater amount of globulin. The expression of inflammation-related genes was increased by IMI in both macrophages and MEC. Except for decreasing expression of SCD1 in MEC, 2,4-TZD did not affect the expression of measured genes. Results confirmed the successful induction of sub-clinical mastitis but did not confirm the positive effect of 2,4-TZD on the response to IMI in well-fed goats
