    Assessing attitudes and high-risk practices to HIV/AIDS infection among the people of Lake Chad basin, Borno State, Nigeria

    A cross sectional quantitative survey that obtained information on high risk behaviours which expose individuals to sexually transmitted infections (STIs) especially HIV was carried out in Lake Chad Basin of Borno among 1,790 volunteers. A well-structured questionnaire was developed and used to collect the desired baseline sociodemographic characteristics by direct interview of respondents in their houses among the populations living in the ordinary households, the mobile population and vulnerable population group (VPG) not living in the households were interviewed in their places of abode with discretion. The mean age range of the participants was 32.5 years (SD + 2.21) ranging from 15 to 49 years. 1,234 (68.9%) of the respondents were males while 556 (31.1%) were females. The prison inmates had the highest prevalence of STIs, 37% of the respondents didn’t know that they were at risk of contracting HIV. The knowledge of HIV status was higher among the males (80.2%) than the females (19.3%). The use of male and female condom was higher among the rural (19.2%) than the urban (10.8%) respondents. This study revealed high risk practices among the respondents even though there is tremendous HIV test intake across both the general and vulnerable populations, there is need for more awareness as rate of risk perception is very low

    Knowledge of awareness about HIV/AIDS among the people of Lake Chad basin of Borno State, Nigeria

    A cross sectional quantitative survey that obtained information on the behavioural characteristics of the general population and the vulnerable groups with respect to knowledge of awareness of HIV/AIDS in Lake Chad Basin of Borno State was carried out among 1,790 volunteers. The respondents, who had ever heard about HIV/AIDS, were disaggregated by target groups. 78.7% of prison inmates, 76.8% of commercial sex workers, 75.2% transporters, 64.5% of uniform service men/women and 58.7% of Island population have heard about HIV/AIDS, while the percentage among traders and Household stood at 56.4% and 45.5% each. On disaggregation by gender, 63.9% of males and 49.8% of the females have heard about HIV/AIDS. 60.4% of the urban and 59.3% of the rural communities reported to have heard about HIV/AIDS. On the knowledge of transmission of the HIV/AIDS, there was 45% awareness on unprotected sexual intercourse, 40% on blood and blood products, 6% on mother to child transmission, 1% on sharp objects while 8% does not know how the virus is transmitted. On HIV/AIDS prevention tools, 34% were aware of abstinence, 32% use of condoms, 13% faithfulness, 8% reported other methods while 13% did not know. The knowledge of awareness about HIV/AIDS though cut across both the general and vulnerable populations, there is need for more awareness to curb this dreaded disease

    T1 Mapping Quantifies Spinal Cord Compression in Patients With Various Degrees of Cervical Spinal Canal Stenosis

    Age-related degeneration of the cervical spinal column is the most common cause of spinal cord lesions. T1 mapping has been shown to indicate the grade and site of spinal cord compression in low grade spinal canal stenosis (SCS). Aim of our study was to further investigate the diagnostic potential of a novel T1 mapping method at 0.75 mm resolution and 4 s acquisition time in 31 patients with various grades of degenerative cervical SCS. T1 mapping was performed in axial sections of the stenosis as well as above and below. Included subjects received standard T2-weighted MRI of the cervical spine (including SCS-grading 0-III), electrophysiological, and clinical examination. We found that patients with cervical SCS showed a significant difference in T1 relaxation times within the stenosis (727 ± 66 ms, mean ± standard deviation) in comparison to non-stenotic segments above (854 ± 104 ms, p < 0.001) and below (893 ± 137 ms, p < 0.001). There was no difference in mean T1 in non-stenotic segments in patients (p = 0.232) or between segments in controls (p = 0.272). Mean difference of the T1 relaxation times was significantly higher in grade III stenosis (234 ± 45) vs. in grade II stenosis (176 ± 45, p = 0.037) vs. in grade I stenosis (90 ± 87 ms, p = 0.010). A higher difference in T1 relaxation time was associated with a central efferent conduction deficit. In conclusion, T1 mapping may be useful as a tool for SCS quantification in all grades of SCS, including high-grade stenosis with myelopathy signal in conventional T2-weighted imaging

    Identification of regeneration-associated genes after central and peripheral nerve injury in the adult rat

    Background: It is well known that neurons of the peripheral nervous system have the capacity to regenerate a severed axon leading to functional recovery, whereas neurons of the central nervous system do not regenerate successfully after injury. The underlying molecular programs initiated by axotomized peripheral and central nervous system neurons are not yet fully understood.Results: To gain insight into the molecular mechanisms underlying the process of regeneration in the nervous system, differential display polymerase chain reaction has been used to identify differentially expressed genes following axotomy of peripheral and central nerve fibers. For this purpose, axotomy induced changes of regenerating facial nucleus neurons, and non-regenerating red nucleus and Clarke's nucleus neurons have been analyzed in an intra-animal side-to-side comparison. One hundred and thirty five gene fragments have been isolated, of which 69 correspond to known genes encoding for a number of different functional classes of proteins such as transcription factors, signaling molecules, homeobox-genes, receptors and proteins involved in metabolism. Sixty gene fragments correspond to genomic mouse sequences without known function. In situ-hybridization has been used to confirm differential expression and to analyze the cellular localization of these gene fragments. Twenty one genes (similar to 15%) have been demonstrated to be differentially expressed.Conclusions: The detailed analysis of differentially expressed genes in different lesion paradigms provides new insights into the molecular mechanisms underlying the process of regeneration and may lead to the identification of genes which play key roles in functional repair of central nervous tissues

    High periventricular T1 relaxation times predict gait improvement after spinal tap in patients with idiopathic normal pressure hydrocephalus

    Purpose:The diagnosis of idiopathic normal pressure hydrocephalus (iNPH) can be challenging. Aim of this study was to use a novel T1 mapping method to enrich the diagnostic work-up of patients with suspected iNPH.Methods:Using 3T magnetic resonance imaging (MRI) we prospectively evaluated rapid high-resolution T1 mapping at 0.5 mm resolution and 4 s acquisition time in 15 patients with suspected iNPH and 8 age-matched, healthy controls.T1 mapping in axial sections of the cerebrum, clinical and neuropsychological testing were performed prior to and after cerebrospinal fluid tap test (CSF-TT). T1 relaxation times were measured in 5 predefined periventricular regions.Results:All 15 patients with suspected iNPH showed gait impairment, 13 (86.6%) showed signs of cognitive impairment and 8 (53.3%) patients had urinary incontinence. Gait improvement was noted in 12 patients (80%) after CSF-TT. T1 relaxation times in all periventricular regions were elevated in patients with iNPH compared to controls with the most pronounced differences in the anterior (1006 ± 93 ms vs. 911 ± 77 ms; p = 0.023) and posterior horns (983 ± 103 ms vs. 893 ± 68 ms; p = 0.037) of the lateral ventricles.     Dual Function of the pUL7-pUL51 Tegument Protein Complex in Herpes Simplex Virus 1 Infection

    The tegument of herpesviruses is a highly complex structural layer between the nucleocapsid and the envelope of virions. Tegument proteins play both structural and regulatory functions during replication and spread, but the interactions and functions of many of these proteins are poorly understood. Here we focus on two tegument proteins from herpes simplex virus 1 (HSV-1), pUL7 and pUL51, which have homologues in all other herpesviruses. We have now identified that HSV-1 pUL7 and pUL51 form a stable and direct protein-protein interaction, their expression levels rely on the presence of each other, and they function as a complex in infected cells. We demonstrate that expression of the pUL7-pUL51 complex is important for efficient HSV-1 assembly and plaque formation. Furthermore, we also discovered that the pUL7-pUL51 complex localizes to focal adhesions at the plasma membrane in both infected cells and in the absence of other viral proteins. The expression of pUL7-pUL51 is important to stabilize focal adhesions and maintain cell morphology in infected cells and cells infected with viruses lacking pUL7 and/or pUL51 round up more rapidly than cells infected with wild-type HSV-1. Our data suggest that, in addition to the previously reported functions in virus assembly and spread for pUL51, the pUL7-pUL51 complex is important for maintaining the attachment of infected cells to their surroundings through modulating the activity of focal adhesion complexes. IMPORTANCE\textbf{IMPORTANCE}: The Herpesviridae \textit{Herpesviridae } is a large family of highly successful human and animal pathogens. Virions of these viruses are composed of many different proteins, most of which are contained within the tegument, a complex structural layer between the nucleocapsid and the envelope within virus particles. Tegument proteins have important roles in assembling virus particles as well as modifying host cells to promote virus replication and spread. However, little is known about the function of many tegument proteins during virus replication. Our study focuses on two tegument proteins from herpes simplex virus 1 that are conserved in all herpesviruses: pUL7 and pUL51. We demonstrate that these proteins directly interact and form a functional complex that is important for both virus assembly and modulation of host cell morphology.
