    Remparts ou menaces ? Trajectoires politiques de l'endiguement en France, aux Pays Bas et aux Etats Unis

    International audienceIn the 1990s, public policies of flood risk prevention in France have changed in nature. Construction of protective infrastructure that prevailed since the nineteenth century was challenged. The flood was then on characterized as inevitable phenomenon in town that was beneficial in rural areas. Criticisms raised against the natural disasters guarantee system adopted in the 1980s. This paper replaces this evolution in a broader context by comparing the French model to the United States and the Netherlands, where corps of engineers have played an important role. We show how these national policies have built territories both privileged and very dependent on the state. We trace the influence of environmental movements in the production of new models at the international level. The French evolution can thus be understood as both an endogenous dynamic and the result of international policy diffusion.Dans les années 1990, les politiques publiques de prévention du risque inondation en France ont changé de nature. Les aménagements de protection qui prévalaient depuis le XIXe siècle ont été remis en cause et l'inondation a été requalifiée comme un phénomène inévitable en ville pour les crues extrêmes et bénéfique en zone rurale. Des critiques se sont levées contre le système de garantie contre les catastrophes naturelles adopté dans les années 1980. Cet article replace cette évolution dans un contexte plus large en comparant le modèle français à ceux développés aux États-Unis et aux Pays-Bas, deux autres pays où les corps d'ingénieurs ont joué un rôle important. Nous montrons comment ces politiques nationales ont construit des territoires à la fois privilégiés et très dépendants de l'État. Puis nous retraçons l'influence des mouvements environnementalistes dans la production de nouveaux modèles au niveau international. L'évolution française peut ainsi être comprise à la fois comme une dynamique endogène et le résultat d'opportunités ouvertes par le niveau international

    The dual hit hypothesis of schizophrenia:evidence from animal models

    Schizophrenia is a heterogeneous psychiatric disorder, which can severely impact social and professional functioning. Epidemiological and clinical studies show that schizophrenia has a multifactorial aetiology comprising genetic and environmental risk factors. Although several risk factors have been identified, it is still not clear how they result in schizophrenia. This knowledge gap, however, can be investigated in animal studies. In this review, we summarise animal studies regarding molecular and cellular mechanisms through which genetic and environmental factors may affect brain development, ultimately causing schizophrenia. Preclinical studies suggest that early environmental risk factors can affect the immune, GABAergic, glutamatergic, or dopaminergic system and thus increase the susceptibility to another risk factor later in life. A second insult, like social isolation, stress, or drug abuse, can further disrupt these systems and the interactions between them, leading to behavioural abnormalities. Surprisingly, first insults like maternal infection and early maternal separation can also have protective effects. Single gene mutations associated with schizophrenia did not have a major impact on the susceptibility to subsequent environmental hits

    Water Supply Basins of São Paulo Metropolitan Region: Hydro-Climatic Characteristics of the 2013–2015 Water Crisis

    São Paulo metropolitan region experienced an acute water crisis between 2013 and 2015. According to the Brazilian and international press, it was due to climatic, anthropogenic, and water management factors. This paper assesses the hydro-climatic characteristics of the crisis by focusing on the Alto-Tietê basin and the headwater of the Piracicaba-Capivari-Jundiaí (PCJ) basin that supply 70% of the water consumed in the metropolis. Based on 16 rain-gauge stations, 5 runoff-gauge stations, and several statistical analyses carried over the 1951–2015 period, this assessment shows that the 2013–2015 hydro-climatic crisis resulted from a large number of days without rain in the north of the Alto-Tietê basin and to less intense precipitation events in the headwaters of the PCJ basin during the austral spring 2013 and the year 2014. It also defines a return period of 98 years for observed river flows in 2014. Despite the rare nature of this hydrological drought, the need for efficient water saving policies is brought forward

    Maternal infection during pregnancy aggravates the behavioral response to an immune challenge during adolescence in female rats

    Prenatal and early postnatal infection have been associated with changes in microglial activity and the development of psychiatric disorders. Here, we investigated the effect of prenatal immune activation and postnatal immune challenge, alone and combined, on behavior and microglial cell density in female Wistar rats. Pregnant rats were injected with poly I:C to induce a maternal immune activation (MIA). Their female offspring were subsequently exposed to a lipopolysaccharide (LPS) immune challenge during adolescence. Anhedonia, social behavior, anxiety, locomotion, and working memory were measured with the sucrose preference, social interaction, open field, elevated-plus maze, and Y-maze test, respectively. Microglia cell density was quantified by counting the number of Iba-1 positive cells in the brain cortex. Female MIA offspring were more susceptible to the LPS immune challenge during adolescence than control offspring as demonstrated by a more pronounced reduction in sucrose preference and body weight on the days following the LPS immune challenge. Furthermore, only the rats exposed to both MIA and LPS showed long-lasting changes in social behavior and locomotion. Conversely, the combination MIA and LPS prevented the anxiety induced by MIA alone during adulthood. MIA, LPS, or their combination did not change microglial cell density in the parietal and frontal cortex of adult rats. The results of our study suggest that the maternal immune activation during pregnancy aggravates the response to an immune challenge during adolescence in female rats.</p

    A single dose of ketamine cannot prevent protracted stress-induced anhedonia and neuroinflammation in rats

    Worldwide, millions of people suffer from treatment-resistant depression. Ketamine, a glutamatergic receptor antagonist, can have a rapid antidepressant effect even in treatment-resistant patients. A proposed mechanism for the antidepressant effect of ketamine is the reduction of neuroinflammation. To further explore this hypothesis, we investigated whether a single dose of ketamine can modulate protracted neuroinflammation in a repeated social defeat (RSD) stress rat model, which resembles features of depression. To this end, male animals exposed to RSD were injected with ketamine (20 mg/kg) or vehicle. A combination of behavioral analyses and PET scans of the inflammatory marker TSPO in the brain were performed. Rats submitted to RSD showed anhedonia-like behavior in the sucrose preference test, decreased weight gain, and increased TSPO levels in the insular and entorhinal cortices, as observed by [11C]-PK11195 PET. Whole brain TSPO levels correlated with corticosterone levels in several brain regions of RSD exposed animals, but not in controls. Ketamine injection 1 day after RSD disrupted the correlation between TSPO levels and serum corticosterone levels, but had no effect on depressive-like symptoms, weight gain or the protracted RSD-induced increase in TSPO expression in male rats. These results suggest that ketamine does not exert its effect on the hypothalamic-pituitary-adrenal axis by modulation of neuroinflammation

    The Ubiquitous Dermokine Delta Activates Rab5 Function in the Early Endocytic Pathway

    The expression of the recently identified dermokine (Dmkn) gene leads to four families of proteins with as yet unknown functions. The secreted α, β and γ isoforms share an epidermis-restricted expression pattern, whereas the δ isoform is intracellular and ubiquitous. To get an insight into Dmknδ function, we performed yeast two-hybrid screening and identified the small GTPases Rab5 as partners for Dmknδ. The Rab5 proteins are known to regulate membrane docking and fusion in the early endocytic pathway. GST pull-down assays confirmed the direct interaction between Rab5 and Dmknδ. Transient expression of Dmknδ in HeLa cells led to the formation of punctate structures colocalized with endogenous Rab5 and clathrin, indicating Dmknδ involvement in the early steps of endocytosis. Dmknδ indeed colocalized with transferrin at early stages of endocytosis, but did not modulate its endocytosis or recycling kinetics. We also showed that Dmknδ was able to bind both inactive (GDP-bound) and active (GTP-bound) forms of Rab5 in vitro but preferentially targeted GDP-bound form in HeLa cells. Interestingly, Dmknδ expression rescued the Rab5S34N-mediated inhibition of endosome fusion. Moreover, Dmknδ caused the enlargement of vesicles positive for Rab5 by promoting GTP loading onto the small GTPase. Together our data reveal that Dmknδ activates Rab5 function and thus is involved in the early endosomal trafficking
