Non-parametric comparison of histogrammed two-dimensional data distributions using the Energy Test

When monitoring complex experiments, comparison is often made between regularly acquired histograms of data and reference histograms which represent the ideal state of the equipment. With the larger HEP experiments now ramping up, there is a need for automation of this task since the volume of comparisons could overwhelm human operators. However, the two-dimensional histogram comparison tools available in ROOT have been noted in the past to exhibit shortcomings. We discuss a newer comparison test for two-dimensional histograms, based on the Energy Test of Aslan and Zech, which provides more conclusive discrimination between histograms of data coming from different distributions than methods provided in a recent ROOT release. The Science and Technology Facilities Council, U

G2A Attenuates Propionibacterium acnes Induction of Inflammatory Cytokines in Human Monocytes.

Background:Acne vulgaris is a disease of the pilosebaceous unit characterized by increased sebum production, hyperkeratinization, and immune responses to Propionibacterium acnes (PA). Here, we explore a possible mechanism by which a lipid receptor, G2A, regulates immune responses to a commensal bacterium. Objective:To elucidate the inflammatory properties of G2A in monocytes in response to PA stimulation. Furthermore, our study sought to investigate pathways by which lipids modulate immune responses in response to PA. Methods:Our studies focused on monocytes collected from human peripheral blood mononuclear cells, the monocytic cell line THP-1, and a lab strain of PA. Our studies involved the use of enzyme-linked immunosorbent, Western blot, reverse transcription polymerase chain reaction, small interfering RNA (siRNA), and microarray analysis of human acne lesions in the measurements of inflammatory markers. Results:G2A gene expression is higher in acne lesions compared to normal skin and is inducible by the acne therapeutic, 13-cis-retinoic acid. In vitro, PA induces both the Toll-like receptor 2-dependent expression of G2A as well as the production of the G2A ligand, 9-hydroxyoctadecadienoic acid, from human monocytes. G2A gene knockdown through siRNA enhances PA stimulation of interleukin (IL)-6, IL-8, and IL-1β possibly through increased activation of the ERK1/2 MAP kinase and nuclear factor kappa B p65 pathways. Conclusion:G2A may play a role in quelling inflammatory cytokine response to PA, revealing G2A as a potential attenuator of inflammatory response in a disease associated with a commensal bacterium

p21-activated kinase (PAK) regulates cytoskeletal reorganization and directional migration in human neutrophils

Neutrophils serve as a first line of defense in innate immunity owing in part to their ability to rapidly migrate towards chemotactic factors derived from invading pathogens. As a migratory function, neutrophil chemotaxis is regulated by the Rho family of small GTPases. However, the mechanisms by which Rho GTPases orchestrate cytoskeletal dynamics in migrating neutrophils remain ill-defined. In this study, we characterized the role of p21-activated kinase (PAK) downstream of Rho GTPases in cytoskeletal remodeling and chemotactic processes of human neutrophils. We found that PAK activation occurred upon stimulation of neutrophils with f-Met-Leu-Phe (fMLP), and PAK accumulated at the actin-rich leading edge of stimulated neutrophils, suggesting a role for PAK in Rac-dependent actin remodeling. Treatment with the pharmacological PAK inhibitor, PF3758309, abrogated the integrity of RhoA-mediated actomyosin contractility and surface adhesion. Moreover, inhibition of PAK activity impaired neutrophil morphological polarization and directional migration under a gradient of fMLP, and was associated with dysregulated Ca2+ signaling. These results suggest that PAK serves as an important effector of Rho-family GTPases in neutrophil cytoskeletal reorganization, and plays a key role in driving efficient directional migration of human neutrophils

How good are your fits? Unbinned multivariate goodness-of-fit tests in high energy physics

Multivariate analyses play an important role in high energy physics. Such analyses often involve performing an unbinned maximum likelihood fit of a probability density function (p.d.f.) to the data. This paper explores a variety of unbinned methods for determining the goodness of fit of the p.d.f. to the data. The application and performance of each method is discussed in the context of a real-life high energy physics analysis (a Dalitz-plot analysis). Several of the methods presented in this paper can also be used for the non-parametric determination of whether two samples originate from the same parent p.d.f. This can be used, e.g., to determine the quality of a detector Monte Carlo simulation without the need for a parametric expression of the efficiency.Comment: 32 pages, 12 figure

A model for shock wave chaos

We propose the following model equation: ut+1/2(u2uus)x=f(x,us),u_{t}+1/2(u^{2}-uu_{s})_{x}=f(x,u_{s}), that predicts chaotic shock waves. It is given on the half-line x<0x<0 and the shock is located at x=0x=0 for any t0t\ge0. Here us(t)u_{s}(t) is the shock state and the source term ff is assumed to satisfy certain integrability constraints as explained in the main text. We demonstrate that this simple equation reproduces many of the properties of detonations in gaseous mixtures, which one finds by solving the reactive Euler equations: existence of steady traveling-wave solutions and their instability, a cascade of period-doubling bifurcations, onset of chaos, and shock formation in the reaction zone.Comment: 4 pages, 4 figure

Mathematical Content Knowledge for Teaching Elementary Mathematics: A Focus on Geometry and Measurement

    Get PDF
This paper summarizes the extant peer-‐reviewed research on PTs’ understanding of geometry and measurement, focusing on a wide variety of topics within these content domains. When looking across the 26 studies reviewed, findings span a variety of content topics, providing little depth in either the geometry or measurement content domain. However, collective findings do indicate PTs’ overall conceptions in geometry and measurement to be limited and weak, with PTs relying on memorized procedural processes. Some evidence indicates that cognitive development, along with spatial visualization skills, plays a greater role in learning geometry than memory skills. In addition, the van Hiele levels of geometric learning provide a helpful framework to think about the development of geometric ideas. Direction of future research is elaborated to address ways to develop PTs’ understanding of geometry and measurement. Gaps that still exist in the research literature regarding PTs’ mathematical content knowledge in geometry and measurement are identified
