    Evoluția și etica eugeniei

    În acest articol Ăźncerc să argumentez opinia că, așa cum este definită eugenia, este foarte dificil de făcut o diferențiere clară Ăźntre știință (medicină, ingineria genetică) și eugenie. Și de stabilit o linie peste care ingineria genetică nu ar trebui să treacă, conform unor norme morale, juridice și religioase. AtĂąta timp cĂąt acceptăm ajutorul geneticii Ăźn găsirea unor modalități de combatere a cancerului, diabetului sau HIV, acceptăm Ăźn mod implicit și eugenia pozitivă, conform definiției actuale. Și atĂąta timp cĂąt acceptăm screening-ul genetic, și intervenții asupra fătului nenăscut, sau avortul, acceptăm Ăźn mod implicit și eugenia negativă. În plus, la nivel de guverne, deși oficial eugenia este repudiată, ea a fost legalizată Ăźn foarte multe țări pĂąnă de curĂąnd, și Ăźncă mai este acceptată și legalizată, chiar dacă Ăźn forme mai subtile, și Ăźn prezent. În Introducere definesc termenul și modurile de clasificare. Urmează Istoria eugeniei pornind din perioada antică, introducerea eugeniei de Francis Galton, practica eugeniei ca politică de stat Ăźn diverse țări, și eugenia actuală (eugenia liberală). Analizez apoi diverse probleme ridicate de Etica eugeniei liberale, și am dezvoltat o secțiune aparte pentru Viitorul eugeniei, cu accent pe proiectul genomului uman.     Crystal and Electronic Structure of Bismuth Thiophosphate, BiPS4:An Earth Abundant Solar Absorber

    The optoelectronic properties of crystalline BiPS4 have been described for the first time for solar energy conversion. Detailed structural analysis is extracted from XRD refinement of powders synthesized by the solid-state method. BiPS4 exhibits a rather unusual 3-dimensional orthorhombic structure with two distinctive Bi sites with octahedral coordination distorted by 6s2 lone pairs. High-resolution TEM imaging clearly shows the two Bi–Bi interatomic distances in close agreement with the XRD analysis. BiPS4 displays a complex Raman spectrum under near-resonant conditions which is rationalized by density functional perturbation theory. Hybrid-functional-DFT calculations show significant spin–orbit coupling effects in Bi 6p bands, not only affecting the band dispersion but also lowering the conduction band minimum by approximately 0.5 eV. The optical properties of BiPS4 powders are dominated by a direct transition at 1.72 eV, closely matching the calculated band gap. Electrochemical experiments revealed n-type conductivity with a flat band potential located at 0.16 V vs RHE. We also show a remarkable agreement between the position of the band edges estimated from first-principles calculations and electrochemical measurements. The time-resolved photoluminescence transient revealed a carrier lifetime of approximately 1 ns, manifesting as strong potential- and wavelength-dependent photocurrent responses. Finally, the nature of the structural defects responsible for the relatively short lifetime is briefly discussed

    Évaluation des compĂ©tences en laparoscopie : comparaison de la fiabilitĂ© des outils d’évaluation globale et des outils d’évaluation de la confiance

    Background: Competence by design (CBD) residency programs increasingly depend on tools that provide reliable assessments, require minimal rater training, and measure progression through the CBD milestones. To assess intraoperative skills, global rating scales and entrustability ratings are commonly used but may require extensive training.  The Competency Continuum (CC) is a CBD framework that may be used as an assessment tool to assess laparoscopic skills. The study aimed to compare the CC to two other assessment tools: the Global Operative Assessment of Laparoscopic Skills (GOALS) and the Zwisch scale. Methods: Four expert surgeons rated thirty laparoscopic cholecystectomy videos. Two raters used the GOALS scale while the remaining two raters used both the Zwisch scale and CC. Each rater received scale-specific training. Descriptive statistics, inter-rater reliabilities (IRR), and Pearson’s correlations were calculated for each scale. Results: Significant positive correlations between GOALS and Zwisch (r = 0.75, p < 0.001), CC and GOALS (r = 0.79, p < 0.001), and CC and Zwisch (r = 0.90, p < 0.001) were found. The CC had an inter-rater reliability of 0.74 whereas the GOALS and Zwisch scales had inter-rater reliabilities of 0.44 and 0.43, respectively. Compared to GOALS and Zwisch scales, the CC had the highest inter-rater reliability and required minimal rater training to achieve reliable scores. Conclusion: The CC may be a reliable tool to assess intraoperative laparoscopic skills and provide trainees with formative feedback relevant to the CBD milestones. Further research should collect further validity evidence for the use of the CC as an independent assessment tool.Contexte : Les programmes de rĂ©sidence structurĂ©s autour de la compĂ©tence par conception (CPC) dĂ©pendent de plus en plus d’outils qui fournissent des Ă©valuations fiables, nĂ©cessitent une formation minimale des Ă©valuateurs et mesurent la progression dans les Ă©tapes de la CPC. Pour Ă©valuer les compĂ©tences peropĂ©ratoires, les Ă©chelles d’évaluation globale et de confiance sont couramment utilisĂ©es mais peuvent nĂ©cessiter une formation approfondie. Le Continuum des compĂ©tences (CC) est un cadre de la CPC qui peut ĂȘtre utilisĂ© comme outil d’évaluation des compĂ©tences laparoscopiques. L’étude visait Ă  comparer le CC Ă  deux autres outils d’évaluation : l’évaluation globale opĂ©ratoire des compĂ©tences laparoscopiques (GOALS) et l’échelle de Zwisch. MĂ©thodes : Quatre chirurgiens experts ont Ă©valuĂ© trente vidĂ©os de cholĂ©cystectomie laparoscopique. Deux Ă©valuateurs ont utilisĂ© l’échelle GOALS tandis que les deux autres ont utilisĂ© l’échelle Zwisch et le CC. Chacun d’eux avait reçu une formation spĂ©cifique Ă  l’échelle utilisĂ©e. Des statistiques descriptives, la fiabilitĂ© inter-Ă©valuateurs (FIÉ) et des corrĂ©lations de Pearson ont Ă©tĂ© calculĂ©es pour chaque Ă©chelle. RĂ©sultats : Des corrĂ©lations positives significatives ont Ă©tĂ© trouvĂ©es entre les Ă©chelles GOALS et Zwisch (r=0.75, p<0.001), CC et GOALS (r=0.79, p<0.001), et CC et Zwisch (r=0.90, p<0.001). Le CC avait une fiabilitĂ© inter-Ă©valuateurs de 0,74 tandis que les Ă©chelles GOALS et Zwisch avaient des fiabilitĂ©s inter-Ă©valuateurs de 0,44 et 0,43, respectivement.     Super-resolution Imaging of the T cell Central Supramolecular Signaling Cluster Using Stimulated Emission Depletion Microscopy

    Supramolecular signaling assemblies are of interest for their unique signaling properties. A ”m scale signaling assembly, the central supramolecular signaling cluster (cSMAC), forms at the center interface of T cells activated by antigen presenting cells (APC). The adaptor protein linker for activation of T cells (LAT) is a key cSMAC component. The cSMAC has widely been studied using total internal reflection fluorescence microscopy of CD4(+) T cells activated by planar APC substitutes. Here we provide a protocol to image the cSMAC in its cellular context at the interface between a T cell and an APC. Super resolution stimulated emission depletion microscopy (STED) was utilized to determine the localization of LAT, that of its active, phosphorylated form and its entire pool. Agonist peptide-loaded APCs were incubated with TCR transgenic CD4(+) T cells for 4.5 min before fixation and antibody staining. Fixed cell couples were imaged using a 100x 1.4 NA objective on a Leica SP8 AOBS confocal laser scanning microscope. LAT clustered in multiple supramolecular complexes and their number and size distributions were determined. Using this protocol, cSMAC properties in its cellular context at the interface between a T cell and an APC could be quantified

    SR9009 administered for one day after myocardial ischemia-reperfusion prevents heart failure in mice by targeting the cardiac inflammasome

    Reperfusion of patients after myocardial infarction (heart attack) triggers cardiac inflammation that leads to infarct expansion and heart failure (HF). We previously showed that the circadian mechanism is a critical regulator of reperfusion injury. However, whether pharmacological targeting using circadian medicine limits reperfusion injury and protects against HF is unknown. Here, we show that short-term targeting of the circadian driver REV-ERB with SR9009 benefits long-term cardiac repair post-myocardial ischemia reperfusion in mice. Gain and loss of function studies demonstrate specificity of targeting REV-ERB in mice. Treatment for just one day abates the cardiac NLRP3 inflammasome, decreasing immunocyte recruitment, and thereby allowing the vulnerable infarct to heal. Therapy is given in vivo, after reperfusion, and promotes efficient repair. This study presents downregulation of the cardiac inflammasome in fibroblasts as a cellular target of SR9009, inviting more targeted therapeutic investigations in the future

    TFG Promotes Organization of Transitional ER and Efficient Collagen Secretion

    Collagen is the most abundant protein in the animal kingdom. It is of fundamental importance during development for cell differentiation and tissue morphogenesis as well as in pathological processes such as fibrosis and cancer cell migration. However, our understanding of the mechanisms of procollagen secretion remains limited. Here, we show that TFG organizes transitional ER (tER) and ER exit sites (ERESs) into larger structures. Depletion of TFG results in dispersion of tER elements that remain associated with individual ER-Golgi intermediate compartments (ERGICs) as largely functional ERESs. We show that TFG is not required for the transport and packaging of small soluble cargoes but is necessary for the export of procollagen from the ER.     Discovery of os cordis in the cardiac skeleton of chimpanzees (Pan troglodytes)

    © 2020, The Author(s). Cardiovascular diseases, especially idiopathic myocardial fibrosis, is one of the most significant causes of morbidity and mortality in captive great apes. This study compared the structure and morphology of 16 hearts from chimpanzees (Pan troglodytes) which were either healthy or affected by myocardial fibrosis using X-ray microtomography. In four hearts, a single, hyperdense structure was detected within the right fibrous trigone of the cardiac skeleton. High resolution scans and histopathology revealed trabecular bones in two cases, hyaline cartilage in another case and a focus of mineralised fibro-cartilaginous metaplasia with endochondral ossification in the last case. Four other animals presented with multiple foci of ectopic calcification within the walls of the great vessels. All hearts affected by marked myocardial fibrosis presented with bone or cartilage formation, and increased collagen levels in tissues adjacent to the bone/cartilage, while unaffected hearts did not present with os cordis or cartilago cordis. The presence of an os cordis has been described in some ruminants, camelids, and otters, but never in great apes. This novel research indicates that an os cordis and cartilago cordis is present in some chimpanzees, particularly those affected by myocardial fibrosis, and could influence the risk of cardiac arrhythmias and sudden death

    Morphology, adipocyte size, and fatty acid analysis of dairy cattle digital cushions, and the effect of body condition score and age

    The digital cushion is an essential part of maintaining a healthy foot, working to dissipate foot strike and body weight forces and lameness from claw horn disruption lesions. Despite the importance of the digital cushion, little is known about the basic anatomy, adipocyte morphology, and fatty acid composition in relation to age, limb position, and body condition score. In total, 60 claws (from 17 cows) were selected and collected from a herd, ensuring that body condition score data and computed micro-tomography were known for each animal. Digital cushion tissue underwent histological staining combined with stereology, systematic random sampling, and cell morphology analysis, in addition to lipid extraction followed by fatty acid analysis. The results describe digital cushion architecture and adipocyte sizes. Adipocyte size was similar across all 4 claws (distal left lateral and medial and distal right lateral and medial) and across the ages (aged 2–7 yr); however, animals with body condition score of 3.00 or more at slaughter had a significantly increased cell size in comparison to those with a score of less than 2.50. Of 37 fatty acid methyl esters identified, 5 differed between either the body condition score or different age groups. C10:0 capric acid, C14:0 myristic acid, C15:0 pentadecanoic acid, and C20:0 arachidic acid percentages were all lesser in lower body condition score cows, whereas C22:1n-9 erucic acid measurements were lesser in younger cows. Saturated fatty acid, monounsaturated fatty acid, and polyunsaturated fatty acid percentages were not altered in the different claws, ages, or body condition score groups. Triglyceride quantities did not differ for claw position or age but had decreased quantities in lower body condition score animals. Digital cushion anatomy, cellular morphology, and fatty acid composition have been described in general and also in animals with differing ages, body condition scores, and in the differing claws. Understanding fat deposition, mobilization, and composition are essential in not only understanding the roles that the digital cushion plays but also in preventing disorders and maintaining cattle health and welfare

    Narratives of ethnic identity among practitioners in community settings in the northeast of England

    The increasing ethnic diversity of the UK has been mirrored by growing public awareness of multicultural issues, alongside developments in academic and government thinking. This paper explores the contested meanings around ethnic identity/ies in community settings, drawing on semi-structured interviews with staff from Children’s Centres and allied agencies conducted for a research project that examined the relationship between identity and the participation of parents/carers in services in northeast England. The research found that respondents were unclear about, especially, white ethnic identities, and commonly referred to other social categorizations, such as age, nationality, and circumstances such as mobility, when discussing service users. While in some cases this may have reflected legitimate attempts to resist overethnicizing non-ethnic phenomena, such constructions coexisted with assumptions about ethnic difference and how it might translate into service needs. These findings raise important considerations for policy and practice

    An International Expert Delphi Consensus to Develop Dedicated Geriatric Radiation Oncology Curriculum Learning Outcomes.

    PURPOSE: The management of older adults with cancer is rapidly becoming a significant challenge in radiation oncology (RO) practice. The education of future radiation oncologists in geriatric oncology is fundamental to ensuring that older adults receive high-quality care. Currently RO trainees receive little training and education in geriatric oncology. The objective of this study was to define core geriatric RO curriculum learning outcomes relevant to RO trainees worldwide. METHODS AND MATERIALS: A 2-stage modified Delphi consensus was conducted. Stage 1 involved the formation of an expert reference panel (ERP) of multiprofessional experts in geriatric oncology and/or RO and the compilation of a potential geriatric RO learning outcomes set. Stage 2 involved 3 iterative rounds: round 1 and round 2 (both online surveys), and an intervening ERP round. These aimed at identifying and refining ideal geriatric RO learning outcomes. Invited participants for round 1 and 2 included oncology health care professionals with expertise across RO, geriatric oncology, and/or education and consumers. Predefined Delphi consensus definitions were applied to the results of rounds 1 and 2. RESULTS: An ERP of 11 experts in geriatric oncology and/or RO was formed. Seventy potential knowledge- and skill-based learning outcomes were identified. In round 1, 103 of 179 invited eligible Delphi participants completed the survey (58% response rate). The ERP round was conducted, resulting in the exclusion of 28 learning outcomes. In round 2, 54 of 103 completed the survey (52% response rate). This identified a final total of 33 geriatric RO learning outcomes. CONCLUSIONS: The geriatric RO learning outcomes described in this study form an international consensus that can inform RO training bodies worldwide. This represents the first fundamental step in developing a global educational framework aimed at improving RO trainee knowledge and skills in geriatric oncology
