Exploitation of the Ugi 5-Center-4-Component Reaction (U-5C-4CR) for the Generation of Diverse Libraries of Polycyclic (Spiro)Compounds
An Ugi multicomponent reaction with chiral cyclic amino acids, benzyl isocyanide and cyclic ketones (or acetone) has been exploited as key step for the generation of peptidomimetics. After a straightforward set of elaborations, the peptidomimetics were converted into polycyclic scaffolds displaying two orthogonally protected secondary amines. Libraries of compounds were obtained decorating the molecules through acylation/reductive amination reactions on these functional groups
Identification and optimization of ligands for the PDZ domains of the proteins Shank3 and Dishevelled 3
Protein-Protein-Interaktionen (PPIs) sind fĂŒr die Bildung von
Proteinnetzwerken von entscheidender Bedeutung, beispielsweise in der
Signaltransduktion oder der Apoptose. Die Interaktionen der Proteine werden
durch PPI-DomÀnen realisiert. Inhibitoren von PPIs werden wie z.B. Paclitaxel
bereits klinisch fĂŒr die Krebstherapie eingesetzt. AuĂerdem sind verschiedene
Krankheiten mit Fehlfunktionen im Auf- bzw. Abbau von Proteinnetzwerken
assoziiert, so dass Inhibitoren fĂŒr die entsprechenden PPI-DomĂ€nen einen
entscheidenden Beitrag zur AufklÀrung dieser Fehlfunktion bzw. zur Therapie
leisten können. Eine der am weitest verbreiteten PPI-DomÀnen ist die
Postsynaptic density 95/Discs large/Zonula occludentes-1 (PDZ)-DomÀne. In
dieser Arbeit wurde die PDZ-DomÀne des Proteins Shank3, dem zentralen
GerĂŒstprotein in der exzitatorischen Synapse, nĂ€her untersucht. Besonders die
PDZ-DomĂ€ne ist fĂŒr die Lokalisation des Proteins essenziell. Mutationen des
Shank-Gens wurden im Zusammenhang mit Autismus beobachtet. Eine signifikant
geringere Menge des Shank-Proteins wurde bei Alzheimerpatienten nachgewiesen.
Selektive Liganden dieser PDZ-DomÀne können wesentlich zur AufklÀrung der
Mechanismen dieser Krankheiten beitragen Um Liganden fĂŒr die Shank3-PDZ-DomĂ€ne
zu finden, wurde eine Fluoreszenzpolarisations-Assay etabliert und fĂŒr das
Screening der ChemBioNet-Bibliothek verwendet. Tetrahydrochinolinderivate
wurden als aktive Verbindungen identifiziert. Die Synthese und Untersuchung
einer fokussierten Bibliothek von Chinolinderivate ergab insgesamt vier aktive
Verbindungen (KI = 10 â 70 ”M). Die Ergebnisse der Struktur-AktivitĂ€ts-
Beziehungs-Studie konnten durch die Kristallstruktur der Shank3-PDZ-DomÀne im
Komplex mit einem der Tetrahydrochinolinderivate erklÀrt werden. Die
SelektivitĂ€tsuntersuchung, durchgefĂŒhrt an den PDZ-DomĂ€nen von DVL3, PSD-95
und Syntrophin, zeigte eine breite AktivitÀt der Chinolinderivate. Um
selektivere Inhibitoren zu erhalten, wurden zusÀtzlich diverse Fragmente und
ein Peptidmimetikum untersucht. Das Peptidmimetikum, in dem das C-terminale
Carboxylat durch ein Tetrazol substituiert war, zeigte wie die Fragmente eine
geringe AffinitÀt (KI ℠300 ”M). Das zweite untersuchte Protein, das DVL-
Protein, ist ein essenzieller Teil des kanonischen Wnt-Signalwegs. Eine
permanente Aktivierung dieser Signalkaskade wurde bei diversen
Krebserkrankungen beobachtet. AuĂerdem ist bekannt, dass das Fehlen der PDZ-
DomÀne des DVL-Proteins den Wnt-Signalweg herunterreguliert. Inhibitoren
dieser PDZ-DomÀne sollten folglich den gleichen Effekt erzeugen. Aus
Vorarbeiten waren AnthranilsĂ€urederivate als Inhibitoren fĂŒr die DVL3-PDZ-
DomÀne bekannt. Es wurden weitere Derivate synthetisiert, die KD-Werte von bis
zu 7 ”M aufwiesen. FĂŒr einen dieser Liganden konnte eine selektive Bindung an
die DVL3-PDZ-DomĂ€ne gegenĂŒber der DVL1-PDZ-DomĂ€ne nachgewiesen werden. Durch
Kristallstrukturen von zwei Liganden jeweils im Komplex mit der DVL3-PDZ-
DomĂ€ne konnte der Bindungsmodus aufgeklĂ€rt und damit eine ErklĂ€rung fĂŒr die
SelektivitĂ€t geben werden. AbschlieĂend ist festzuhalten, dass die Interaktion
mit der Carboxylatbindungsstelle der PDZ-DomĂ€ne fĂŒr die Bindung unabdingbar
ist. Gelingt es zusÀtzliche Interaktionen, beispielsweise entlang der
Peptidbindungsspalte, zu generieren, sollte eine Erhöhung der AffinitÀt bis in
den nanomolaren Bereich möglich sein.Protein-protein interactions (PPIs) are important for the formation of protein
networks, e.g. in signal transduction or apoptosis. The interactions of
proteins are mediated by PPI domains. Inhibitors of PPIs, e.g. Paclitaxel are
already in use for cancer therapy. Furthermore, problems concerning the
formation or degradation of protein networks are interconnected to several
diseases. Inhibitors for these PPI domains are useful as therapeutics as well
as tool compounds to elucidate the mechanism of such diseases. One of the most
widespread PPI domains is the Postsynaptic density 95/Discs large/Zonula
occludentes-1 (PDZ) domain. In the present work the PDZ domain of the protein
Shank3 was the subject of investigation. The Shank protein is the central
scaffold protein of excitatory synapses. Especially the Shank PDZ domain is
crucial for the localization of the protein within the synapse. Mutations in
the Shank gene are linked to autism spectrum disorders. A significant
reduction of Shank contents in the synapse is reported for Alzheimer patients.
Selective inhibitors of the Shank PDZ domain could be used as tool to clarify
the mechanisms of these disorders. In order to screen the ChemBioNet library a
homogeneous binding assay based on fluorescence polarization was developed.
The screening revealed tetrahydroquinoline derivatives as ligands for the
Shank3 PDZ domain. After synthesizing and analyzing a focused library of
quinoline derivatives four compounds showed activity (KI values between 10 and
70 ”M) in regard to the Shank3 PDZ domain. The result of a structure activity
relationship study was completely confirmed by the crystal structure of the
Shank3 PDZ domain in complex with a tetrahydroquinoline derivative of the
focused library. Binding studies of the active compounds using the PDZ domains
of PSD-95, Dishevelled 3 (DVL3) and syntrophin were performed. All tested
compounds showed bindings to several PDZ domains. To obtain selective ligands
several fragments derived from a virtual screening and a screening of a
fragment library were tested. Additionally a peptide analogue with a
C-terminal tetrazole as a known isoster of a carboxylate was synthesized and
tested. It showed as well as the fragments weak binding to the PDZ domain
(peptide analogue: KI ℠300 ”M). The DVL3 protein was the second one
investigated in this work. DVL is a crucial part of the canonical Wnt
signaling. A permanent activation of this signaling cascade was observed in
cancer. It is known that the absence of the PDZ domain in DVL results in a
down-regulation of Wnt signaling. Inhibitors of the PDZ domain should cause
the same effect. From previous work, it is known that anthranilic acid
derivatives are ligands for the DVL3 PDZ domain. The synthesis of derivatives
revealed compounds showing KD-values up to 7 ”M. One of these compounds showed
selectivity towards the DVL1 PDZ domain. The crystal structures of two ligands
in complex with the DVL3 PDZ domain revealed the binding mode and explained
the selectivity. It can be summarized that an interaction with the carboxylate
binding loop of the PDZ domain is crucial for binding of a ligand. It should
be possible to generate a nanomolar affinity, if additional interactions could
be produced
Small-molecule inhibitors of the PDZ domain of Dishevelled proteins interrupt Wnt signalling
Dishevelled (Dvl) proteins are important regulators of the Wnt signalling pathway, interacting through their PDZ domains with the Wnt receptor Frizzled. Blocking the Dvl PDZâFrizzled interaction represents a potential approach for cancer treatment, which stimulated the identification of small-molecule inhibitors, among them the anti-inflammatory drug Sulindac and Ky-02327. Aiming to develop tighter binding compounds without side effects, we investigated structureâactivity relationships of sulfonamides. X-ray crystallography showed high complementarity of anthranilic acid derivatives in the GLGF loop cavity and space for ligand growth towards the PDZ surface. Our best binding compound inhibits Wnt signalling in a dose-dependent manner as demonstrated by TOP-GFP assays (IC50âŒ50â”M) and Western blotting of ÎČ-catenin levels. Real-time PCR showed reduction in the expression of Wnt-specific genes. Our compound interacted with Dvl-1 PDZ (KD=2.4â”M) stronger than Ky-02327 and may be developed into a lead compound interfering with the Wnt pathway
