    Comparative analysis of two glyceraldehyde-3-phosphate dehydrogenases from a thermoacidophilic archaeon, Sulfolobus tokodaii

    AbstractSulfolobus tokodaii, a thermoacidophilic archaeon, possesses two structurally and functionally different enzymes that catalyze the oxidation of glyceraldehyde-3-phosphate (GAP): non-phosphorylating GAP dehydrogenase (St-GAPN) and phosphorylating GAP dehydrogenase (St-GAPDH). In contrast to previously characterized GAPN from Sulfolobus solfataricus, which exhibits V-type allosterism, St-GAPN showed K-type allosterism in which the positive cooperativity was abolished with concomitant activation by glucose 1-phosphate (G1P).     Misleading presentations in functional food trials led by contract research organizations were frequently observed in Japan : meta-epidemiological study

Research focusing on CRO-managed trials and the communication of trial outcomes to the consumer market remains underexplored. This metaepidemiological study aims to evaluate the quality of randomized controlled trials (RCTs) facilitated by prominent CROs in Japan and to examine the quality of the representations used to convey their results to consumers. [Study Design and Setting] This study focused on the food trials that were registered in the University Hospital Medical Information Network Clinical Trial Registry or the International Clinical Trials Registry Platform by the top 5 CROs. Press releases of study results or advertisements of food products based on the study results were identified by conducting a Google search. The risk of bias in the RCT publications was independently assessed by 2 reviewers, who also evaluated the presence of “spin” in the abstracts and full texts. An assessment of “spin” in press releases/advertisements was undertaken. [Results] A total of 76 RCT registrations, 32 RCT publications, and 11 press releases/advertisements were included. Approximately 72% of the RCT publications exhibited a high risk of bias due to selective outcome reporting. “Spin” was present in the results of the abstract (72%), abstract conclusion (81%), full-text results (44%), and full-text conclusion (84%). “Spin” appeared in 73% of press releases/advertisements due to the selective outcome reporting. [Conclusion] Functional food presentations in Japan frequently contained “spin.” The Japanese government should more rigorously check whether food manufacturers report outcomes selectively

    Peripheral platelet phagocytosis in an extremely low birth weight infant: a case report

    A 768 g female neonate, born at 25 weeks' gestation, developed sepsis due to methicillin-resistant Staphylococcus epidermidis on day 14. Severe thrombocytopenia was observed, and hemophagocytic macrophages were identified in her peripheral blood smear. Cytokine profiles at the time of onset suggested that an inflammatory cytokine storm had activated lymphocytes and macrophages, leading to platelet phagocytosis. After administration of vancomycin for 14 days and immunoglobulin therapy, she improved without any complications. Considering the results of cytokine profiles, early intervention for infection may have prevented progression to hemophagocytic lymphohistiocytosis and reduced the severity of clinical symptoms

    Investigation of biomarkers in a rare case of fulminant necrotizing enterocolitis in a preterm infant

    We encountered a very rare case of fulminant necrotizing enterocolitis (F-NEC) in a preterm male baby. The course of NEC and sepsis in this case was clearly different from the usual course. After onset at 14 days of life, catheter-related bloodstream infection was first assumed, and antibiotics and γ-globulin administration were started. However, 12 hours after onset, the baby's abdominal distension increased remarkably, and his entire abdominal wall turned red to purple. Escherichia coli were isolated from the blood culture, but the catheter tip culture was negative. Exchange transfusion was performed 32 hours after onset, but no significant changes were observed in the baby's general condition, and he died 46 hours after onset. The acute phase reactants of CRP and α1-acid glycoprotein increased, but haptoglobin did not. Although IL-1β and TNFα increased as expected with sepsis, IL-6, IL-8, IL-10, and G-CSF however increased to a greater extent than expected. From the above, we diagnosed the development of intestinal necrosis as a result of widespread intestinal ischemia, and that sepsis was associated with this poor condition

    Constraints on the Intracluster Dust Emission in the Coma Cluster of Galaxies

    We have undertaken a search for the infrared emission from the intracluster dust in the Coma cluster of galaxies by the Multiband Imaging Photometer for Spitzer. Our observations yield the deepest mid and far-infrared images of a galaxy cluster ever achieved. In each of the three bands, we have not detected a signature of the central excess component in contrast to the previous report on the detection by Infrared Space Observatory (ISO). We still find that the brightness ratio between 70 and 160 microns shows a marginal sign of the central excess, in qualitative agreement with the ISO result. Our analysis suggests that the excess ratio is more likely due to faint infrared sources lying on fluctuating cirrus foreground. Our observations yield the 2 sigma upper limits on the excess emission within 100 kpc of the cluster center as 5 x 10^-3 MJy/sr, 6 x 10^-2 MJy/sr, and 7 x 10^-2 MJy/sr, at 24, 70, and 160 microns, respectively. These values are in agreement with those found in other galaxy clusters and suggest that dust is deficient near the cluster center by more than 3 orders of magnitude compared to the interstellar medium.Comment: 10 pages, 9 figures, minor changes to match version published in Ap
