    Advanced Java-based application to process thermoluminescence digital color images

    Dielectric minerals that interact with ionizing radiation emit thermoluminescence when heated. The color of the emitted thermoluminescence varies among different samples, even for the same minerals. TL color analysis can be used to study mineral provenance. Thermoluminescence color images (TLCI) can be obtained using a digital camera. Inagaki et al. (2010) created a Java application to handle RGB (red-green-blue) information of each pixel. This Java application is able to visually represent thermoluminescence color on a CIE (Commission Internationale de l’Eclairage) chromaticity diagram. Here, we report an improved Java application that enables the numerical treatment of TLCI of various image sizes and resolutions. This application allows for the handling of many digital TLCI in a short time and it is useful for statistical color analyses

    Improved clonality detection in B-cell lymphoma using a semi-nested modification of the BIOMED-2 PCR assay for IGH rearrangement: A paraffin-embedded tissue study

    The BIOMED-2 PCR protocol for targeting the IGH gene is widely employed for detecting clonality in B-cell malignancies. Unfortunately, the detection of clonality with this method is not very sensitive when paraffin sections are used as a DNA source. To increase the sensitivity, we devised a semi-nested modification of a JH consensus primer. The clonality detection rates of three assays were compared: the standard BIOMED-2, BIOMED-2 assay followed by BIOMED-2 re-amplification, and BIOMED-2 assay followed by semi-nested BIOMED-2. We tested more than 100 cases using paraffin-embedded tissues of various B-cell lymphomas, and found that the clonality detection rates with the above three assays were 63.9%, 79.6%, and 88.0%, respectively. While BIOMED-2 re-amplification was significantly more sensitive than the standard BIOMED-2, the semi-nested BIOMED-2 was significantly more sensitive than both the standard BIOMED-2 and BIOMED-2 re-amplification. An increase in sensitivity was observed in all lymphoma subtypes examined. In conclusion, tumor clonality may be detected in nearly 90% of B-cell lymphoma cases with semi-nested BIOMED-2. This ancillary assay may be useful when the standard BIOMED-2 fails to detect clonality in histopathologically suspected B-cell lymphomas

    Paradoxical Regulation of Human FGF21 by Both Fasting and Feeding Signals: Is FGF21 a Nutritional Adaptation Factor?

    Fibroblast growth factor 21 (FGF21) has recently emerged as a metabolic hormone involved in regulating glucose and lipid metabolism in mouse, but the regulatory mechanisms and actions of FGF21 in humans remain unclear. Here we have investigated the regulatory mechanisms of the human FGF21 gene at the transcriptional level. A deletion study of the human FGF21 promoter (βˆ’1672 to +230 bp) revealed two fasting signals, including peroxisome proliferator-activated receptor Ξ± (PPARΞ±) and glucagon signals, that independently induced human FGF21 gene transcription in mouse primary hepatocytes. In addition, two feeding signals, glucose and xylitol, also dose-dependently induced human FGF21 gene transcription and mRNA expression in both human HepG2 cells and mouse primary hepatocytes. FGF21 protein expression and secretion were also induced by high glucose stimulation. The human FGF21 promoter (βˆ’1672 to +230 bp) was found to have a carbohydrate-responsive element at βˆ’380 to βˆ’366 bp, which is distinct from the PPAR response element (PPRE). Knock-down of the carbohydrate response element binding protein by RNAi diminished glucose-induced human FGF21 transcription. Moreover, we found that a region from βˆ’555 to βˆ’443 bp of the human FGF21 promoter region exerts an important role in the activation of basic transcription. In conclusion, human FGF21 gene expression is paradoxically and independently regulated by both fasting and feeding signals. These regulatory mechanisms suggest that human FGF21 is increased with nutritional crisis, including starvation and overfeeding

    Involvement of SIK3 in Glucose and Lipid Homeostasis in Mice

    Salt-inducible kinase 3 (SIK3), an AMP-activated protein kinase-related kinase, is induced in the murine liver after the consumption of a diet rich in fat, sucrose, and cholesterol. To examine whether SIK3 can modulate glucose and lipid metabolism in the liver, we analyzed phenotypes of SIK3-deficent mice. Sik3βˆ’/βˆ’ mice have a malnourished the phenotype (i.e., lipodystrophy, hypolipidemia, hypoglycemia, and hyper-insulin sensitivity) accompanied by cholestasis and cholelithiasis. The hypoglycemic and hyper-insulin-sensitive phenotypes may be due to reduced energy storage, which is represented by the low expression levels of mRNA for components of the fatty acid synthesis pathways in the liver. The biliary disorders in Sik3βˆ’/βˆ’ mice are associated with the dysregulation of gene expression programs that respond to nutritional stresses and are probably regulated by nuclear receptors. Retinoic acid plays a role in cholesterol and bile acid homeostasis, wheras ALDH1a which produces retinoic acid, is expressed at low levels in Sik3βˆ’/βˆ’ mice. Lipid metabolism disorders in Sik3βˆ’/βˆ’ mice are ameliorated by the treatment with 9-cis-retinoic acid. In conclusion, SIK3 is a novel energy regulator that modulates cholesterol and bile acid metabolism by coupling with retinoid metabolism, and may alter the size of energy storage in mice

    Nature of Streaky Structures Observed with a Doppler Lidar

    Observations using a three-dimensional scanning coherent Doppler lidar in an urban area revealed the characteristics of streaky structures above a rough, inhomogeneous surface for a high-Reynolds-number flow. The study focused on two points: (1) the frequency of occurrence and conditions required for the presence of streaky structures, and (2) the universal scaling of the spacing of streaky structures (. The horizontal snapshots of the radial velocity were visually classified into six groups: Streak, Mixed, Fishnet, No streak, Front, and Others. The Streak category accounted for more than 50% of all possible flows and occurred when the horizontal wind speed was large and the atmospheric stratification was near-neutral. The spacing ( was estimated from the power spectral density of the streamwise velocity fluctuations along the spanwise direction. The spacing decreased with an increase in the local velocity gradient. Furthermore, it was revealed that the local velocity gradient normalized by the friction velocity and the boundary-layer height ( comprehensively predicts under various experimental and environmental conditions, in terms of the scale of motion (i.e., indoor and outdoor scales), thermal stratification (i.e., from weakly unstable to stable stratification), and surface roughness (i.e., from flat to very rough surfaces)
