    Kraft lignin degradation by autochtonous streptomyces strains isolated from a tropical lagoon ecosystem

    Kraft lignin contributes to the toxicity of the pulping plant effluent and is known to resist microbial treatment.The lignin component must be removed from lignocellulose biomass to enhance the release of fermentable sugars for the production of biofuel and other value-added end products. Lignin-degrading bacteria provide an advantage due to their ease of isolation,wider tolerance of environmental conditions and genetic manipulations compared with their fungal counterparts. There is no documented evidence on the degradation of kraft lignin by bacteria in the tropical estuarine ecological niche in Nigeria. Bacterial growth and assessment of kraft-lignin degradation in submerged fermentation was carried out for a period of 10 days using Streptomyces spp isolated from a tropical lagoon as the inocula. The organisms utilized 23 to 99 % kraft-lignin at the rate of 2.3×10-5 to 9.9×10-5 g.d-1cm-3 with specific growth rates of 0.020 - 0.084 h-1and doubling times of 8.3 - 35.1 h. Maximum values obtained for laccase and peroxidase activities were 9.5x10-2 and 400 μ mol mg -1min -1 respectively. The aim of this study was to obtain evidences for Kraft lignin degradation by indigenous tropical estuarine Streptomyces species from Lagos, Nigeria. The Autochthonous bacterial species of the Lagos lagoon utilize kraft lignin as a sole carbon source and may be good candidates for biotechnological purposes. The outcome of this study has bridged an information gap in the tropical environment and will complement existing global data because the information on the degradation of kraft lignin by marine Streptomyces is not common

    Characterization of lignocellulolytic bacterial strains associated with decomposing wood residues in the Lagos lagoon, Nigeria

    Aims: The presence of lignocelluloses, especially sawdust in the Lagos lagoon and the attendant ecological problems warranted studies on their degradation. This study aimed to isolate and identify the indigenous bacterial strains capable of utilizing lignocellulosic wastes under the prevalent tropical estuarine conditions. Methodology and results: Nine bacterial species were obtained by elective culture from decomposing wood residues in the lagoon. They were identified on the basis of morphology, biochemical characteristics and analysis of their 16S rRNA gene sequences as Streptomyces, Bacillus and Paenibacillus species. They were cultured on various ligninrelated lignocellulosic substrates over a period of 7 to 12 days. All the isolates showed moderate to very good growth on sugarcane baggase. Streptomyces albogriseolus strain AOB and Paenibacillus sp. ROB showed good growth on grass while on sawdust, only Streptomyces AOB, and Bacillus megaterium strain NOB showed good growth. High performance liquid chromatographic analysis showed that the Streptomyces species completely utilized coniferyl alcohol, B. megaterium strain NOB utilized 90-100% of all the lignin- related aromatic compounds. All the bacterial species utilized less than 40% of sinapyl alcohol, Bacillus sp. OOB and Paenibacillus sp. strain ROB failed to utilize vanillic acid. Conclusion, significance and impact of study: The isolates degraded lignocellulosic wastes and lignin-related compounds. The role of fungi in the breakdown of lignocellulose in the Lagos lagoon had been the subject of previous research considerations whilst the role of bacteria spp was unreported. Autochthonous bacterial species may equally play a role in the bio-rehabilitation of the sawdust-polluted water of the Lagos lagoon

    Adequacy of Technical Facilities for Maternal Health Care Delivery in Public Secondary and Tertiary Facilities in Osun State, Nigeria

    Nigeria has one of the highest maternal mortalities and morbidities in the world, and the need to address these challenges continues to engage the attention of relevant stakeholders but very little attention has focused on technological resources within the maternal health research agenda. This study aims to identify and determine the adequacy of the key equipment for emergency obstetric care (EmOC) services in public-sector secondary and tertiary health facilities in Osun State, Southwest, Nigeria. Using a two-stage probability sampling procedure, 7 health facilities based in 6 randomly selected Local government areas (LGAs) were involved in the study. Data were obtained through the use of primary and secondary sources. For primary sources, a questionnaire was administered to 112 health workers and the study also utilized a checklist for equipment inventory. For secondary sources we reviewed existing hospital records on technology acquisition and maintenance. Relevant descriptive and inferential statistics were used for the data analysis. Almost three-fifths (58.1%) of the health workers considered the technical facilities for the delivery of EmOC in their facilities as inadequate, the proportion of health workers with such opinion was significantly higher in secondary compare to tertiary facilities (60.0% vs 40.0%, p=0.003). The inventory review and use of the checklist also showed varying degree of inadequate technical facilities; none of the facilities had all the 26 equipment considered as essential for comprehensive EmOC in functional state. We concluded that the public sector facilities in Osun State have inadequate technical facilities for EmOC; addressing this technological challenge will be critical to improving maternal health outcomes and statistics in Nigeria

    REPRODUCTIVE PERFORMANCE, FEED INTAKE AND EFFICIENCY OF INDIGENOUS AND CROSSBRED TURKEYS

    This study assessed the reproductive performance of indigenous and crossbred parent stocks and the influence of sire genotype on the growth and efficiency of feed utilization by their progenies. A total of 300 poults, 150 for each genotype were generated from two crosses (Nicholas white x Indigenous and Indigenous x Indigenous turkeys). Data were collected on the reproductive performance of the parents while body weight and feed intake from day old to the 20th week of age were also collected from the progenies generated. The experiment was a complete randomized design with data generated subjected to two way analysis of variance using SAS. The result of the study showed that sire genotype had significant effect (p<0.05) on semen colour and semen volume while semen pH, motility, morphology and live dead ratio were not significantly affected. Crossbred tom had higher semen volume (0.39 ml) compared to 0.18 ml recorded in indigenous turkey. Genotype had no significant effect (p>0.05) on all female reproductive traits such as fertility, hatchability, dead in-germ, weak in-shell and dead in-shell. However, the indigenous turkey had higher fertility percentage (85%) while 80% hatchability was observed in crossbred turkey. Crossbred turkey significantly (p<0.05) had higher body weight (3330g) at 20 week of age compared to the indigenous turkey (2869g). Sexual dimorphism in favour of the male turkey was also observed throughout the period of the experiment. Although the crossbred turkey consumed more feed, the efficiency of feed utilization was better in the indigenous turkey. This findings suggested that the indigenous turkeys can be successfully improved with the introgression of exotic genetic material while crossbred turkeys can be further selected for improved productive and adaptive traits. &nbsp

    Performance Evaluation of A Developed Dewatered Cassava Mash Sifter

    A cassava pulp sifter for grated and dewatered cassava was developed and evaluated. The machine was evaluated at three operating speeds of 260rpm, 350rpm and 530rpm and masses of 1kg, 2kg and 3kg.  It was observed that increase in the speed of operation increased both sieving capacity and sieving efficiency of the machine. Increase in mass, increased sieving capacity and decreased sieving efficiency. The average maximum sifting efficiency and capacity were 95.32% and 613.16kg/hr respectively. All the materials used for the fabrication were obtained locally and the machine has low labour requirements which can be adopted in gari processing industries.     Genotoxicity Evaluation of Landfill Solid Waste Leachates from a Waste Dumpsite in Lagos State, Nigeria using the Allium cepa Assay

    The management of municipal landfill’s leachate is considered one of the most significant environmental issues. Potential mutagenic and genotoxic effects of solid waste leachates (SWL) were investigated using the Allium cepa root meristematic cells assay technique. In Allium root growth inhibition test, experimental onion bulbs were cultivated in various concentrations of the SWL and distilled water was used as a negative control. The root tips from the treated bulb were processedby orcein squash technique after 72 h. The mean lengths of root bundles were obtained andeffective concentration (EC) values calculated. Some of the physicochemical properties of the WL were also determined. The A. cepa assay was carried out at concentrations of 0.05, 0.1, 2.0, 5, 10, 20, 50 and 100% of the WL. There was a statistically different (p < 0.05), concentrationdependent inhibition of onion root growth and mitotic index, and induction of chromosomal aberrations in the onion test. The results of the physicochemical analysis revealed that the concentrations of some parameters were above the maximum permissible limit set by the world health organization (WHO) and could partly be correlated with the toxicity of wastewater. The landfill leachates pose a risk to human health and the environment in general. Hence, ecotoxicity/genotoxicity assays would be useful in leachate risk assessment when coupled with physicochemical analysis

    Coniferyl alcohol is one of the major precursors of lignin; the most abundant aromatic compound and a natural resource currently receiving attention because of the value-added metabolites resulting from its degradation. Growth study of Streptomyces albogriseolus KF977548 (strain AOB) isolated from decaying wood residues in a tropical estuarine ecosystem was carried out using coniferyl alcohol as a sole carbon source. Cell growth and metabolite production were monitored at 24 h interval by dry weight measurements and HPLC, LC–MS-DAD analyses. Biochemical and PCR assays were carried out to detect the major catabolic enzymes of interest. Strain AOB utilized coniferyl alcohol completely within 72 h ( = 0.204 h−1, Td = 3.4 h). Laccase and peroxidase were released into the growth medium up to 0.099 and 98 mol/mL respectively. Protocatechuate 3, 4-dioxygenase and demethylase were detected in the genome whilst ortho-adipate pathway was clearly indicated. Growth on coniferyl alcohol or caffeic acid as mono substrates resulted in the production of secondary metabolites identified by HPLC–MS as 1- caffeoylquinic and 3,4,5-tricaffeoylquinic acids, known as chlorogenic acids, in the culture medium. The microbial production of chlorogenic acids from a lignin-related substrate base by strain AOB could arouse a plausible biotechnological process

    SELECTED WELFARE PARAMETERS OF BROILER CHICKENS ON DIFFERENT FEED QUANTITY AND TIME

    This study was conducted to determine welfare of broiler chickens fed ad libitum and restricted feed during early (08.00hour) and late (16:00hour) of the day. One hundred and eighty (21d old) broiler chickens were randomly allocated to 4 treatments in a 2x2 factorial arrangement comprising of ad libitum and restricted feeding (75% of ad libitum) quantity at 8 and 16 hours feeding time in three replicates. Data were collected on leg problem, dust bathing, body temperature, mortality and haematology. Result revealed that neither feed quantity nor time had significant (P&gt;0.05) effect on leg problem, dust bathing and mortality of broiler chickens. Broiler chickens under ad lib feed quantity and 16:00hours feeding time had (P&lt;0.05) highest body temperature (40.93oC) while those under ad libitum morning (08:00hours) recorded the least (40.75°C). Pack Cell Volume had similar (P&gt;0.05) increase from 26.75% at the beginning (21d) to 31.25 (%) at 56d with broiler chickens under restricted feeding and fed at 16:00hours feeding time. Basophil count reduced from a range of 0.5-2% (P&lt;0.05) to 1% (P&gt;0.05) with restricted feeding. Also eosinophil reduced from 2-4% (P&lt;0.05) range to 0-2.5% (P&gt;0.05) with restricted feeding while birds under ad lib increased from 0-1% (P&lt;0.05) at 21d to 2.75% (P&gt;0.05) at 56d. Better welfare (good body temperature) of broiler chicken could be achieved with early feeding time irrespective of quantity. However, restricted feeding could be explored in broiler production where reduced basophil, eosinophil and stress are vital for enhanced welfare and performance. &nbsp; &nbsp

    Effect of Coat Colour on Water Intake and Feed Utilization of Intensively Reared West African Dwarf Sheep in the Humid Tropics

    The effect of coat colour variation on water intake and feed utilization needs to be fully investigated, determined and exploited to help West African Dwarf (WAD) sheep breeders in genetic improvement programmes. This study examined the effect of coat colour on water intake and feed utilization of intensively reared WAD sheep in the humid tropics. Sixty (60) WAD sheep of different coat colours were used for the experiment which lasted for 6 months. Prominent coat colour types in the region are black, brown and badgerface.  Animals were grouped into sexes. Daily free water intake (DFWI), DFWI/metabolic weight, daily total water intake (DTWI), DTWI/metabolic weight, daily feed intake (DFI), DFI/metabolic weight, DFWI/DFI, daily weight gain (DWG) and feed efficiency (FE) were measured. Analysis of data revealed that coat colour and sex had significant (P&lt;0.05) effect on parameters measured except DTWI and DTWI/metabolic weight. Brown sheep had the highest value for the parameters measured except DFWI/DFI and FE. There was no significant (P&gt;0.05) difference in DWG of black sheep (96.64±3.42 g) and brown sheep (95.24±2.77 g). Best feed efficiency (0.17±0.01) was observed in black sheep. Rams had the highest value for the parameters measured except DFWI/DFI and FE. Based on our results, we concluded that brown WAD sheep and rams consumed more feed and water, while black sheep consumed less feed and they are better feed utilizers.
