    Statistics of amplitude and fluid velocity of large and rare waves in the ocean

The certified thesis is available in the Institute Archives and Special Collections.Includes bibliographical references (leaves 97-99).The understanding of large and rare waves in the ocean is becoming more important as these rare events are turning into more common observances. In order to design a marine structure or vehicle to withstand such a potentially devastating phenomenon, the designer must have knowledge of extreme waves with return periods of 50 and 100 years. Based on satellite radar altimeter data, researchers have successfully predicted extreme significant wave heights with the return periods of 50 and 100 years. This thesis extends their research further by estimating the most probable extreme wave heights and other wave statistics based on spectral analysis. The same technique used for extreme significant wave height prediction is applied to extrapolation of corresponding mean wave periods, and they are used to construct two parameter spectra representing storm sea conditions. The prediction of the most probable extreme wave heights as well as other statistical data is based on linear theory and short term order statistics. There exists sufficient knowledge of second order effects on wave generation, and it could be applied to a logical progression of the simulation approach in this thesis.(cont.) However, because this greatly increases computation time, and the kinematics of deep sea spilling breakers are not yet fully understood for which substantial new research is required, the nonlinear effects are not included in this thesis. Spectral analysis can provide valuable statistical information in addition to extreme wave height data, and preliminary results show good agreement with other prediction methods including wave simulation based on the Pierson-Moskowitz spectrum.by Il Ho Suh.Nav.E.and S.M.in Mechanical Engineerin

    Overexpression of peroxiredoxin I and thioredoxin1 in human breast carcinoma

    <p>Abstract</p> <p>Background</p> <p>Peroxiredoxins (Prxs) are a novel group of peroxidases containing high antioxidant efficiency. The mammalian Prx family has six distinct members (Prx I-VI) in various subcellular locations, including peroxisomes and mitochondria, places where oxidative stress is most evident. The function of Prx I in particular has been implicated in regulating cell proliferation, differentiation, and apoptosis. Since thioredoxin1 (Trx1) as an electron donor is functionally associated with Prx I, we investigated levels of expression of both Prx I and Trx1.</p> <p>Methods</p> <p>We investigated levels of expression of both Prx I and Trx1 in breast cancer by real-time polymerase chain reaction (RT-PCR) and Western blot.</p> <p>Results</p> <p>Levels of messenger RNA (mRNA) for both Prx I and Trx1 in normal human breast tissue were very low compared to other major human tissues, whereas their levels in breast cancer exceeded that in other solid cancers (colon, kidney, liver, lung, ovary, prostate, and thyroid). Among members of the Prx family (Prx I-VI) and Trx family (Trx1, Trx2), Prx I and Trx1 were preferentially induced in breast cancer. Moreover, the expression of each was associated with progress of breast cancer and correlated with each other. Western blot analysis of different and paired breast tissues revealed consistent and preferential expression of Prx I and Trx1 protein in breast cancer tissue.</p> <p>Conclusion</p> <p>Prx I and Trx1 are overexpressed in human breast carcinoma and the expression levels are associated with tumor grade. The striking induction of Prx I and Trx1 in breast cancer may enable their use as breast cancer markers.</p

    Numerical analysis for 137CS on seabed sediment including atmospheric deposition near the Fukushima coastal area

    Radionuclides were released into the atmosphere and ocean from the accident at the Fukushima Daiichi Nuclear Power Plant (NPP) in March 2011. Numerical simulations were performed to evaluate the distribution of 137Cs in the ocean with considering directly release and deposition from the atmosphere. Calculated concentrations of 137Cs in the sea water and seabed are compared with the measured data, and atmospheric transport model has been also used to calculate the rates of atmospheric deposition on the sea surface. Lots of atmospheric deposition was occurred on sea surface in northeast direction from Fukushima NPP due to westerly wind and precipitation on March 15-31, 2011. Most modelling for the marine dispersion of 137Cs after Fukushima accident was considered as a perfectly conservative radionuclide, thus scavenging processes and adsorption on seabed sediments are neglected. The processes of the non-conservative from the numerical simulations have been well described and the calculated results are also in agreement with the measured data. The concentrations of 137Cs in seabed sediments were mainly contributed by the atmospheric deposition and long residence time showed in northeast coastal area off Fukushima NPP

    The dispersion of 137Cs released from Fukushima nuclear power plant to the sea after the March 11th 2011 tsunami has been studied using numerical models. The 3D dispersion model consists of an advec tion/diffusion equation with terms describing uptake/release reactions between water and seabed sedi ments. The dispersion model has been fed with daily currents provided by HYCOM and JCOPE2 ocean models. Seabed sediment 137Cs patterns obtained using both current data set have been compared. The impact of tides and of atmospheric deposition has been evaluated as well.     Should we measure plutonium concentrations in marine sediments near Fukushima?

    Pu contamination originating from Fukushima accident has not been detected in marine sediment samples collected outside a 30 km circle around the plant. It is shown, by means of numerical modelling, that if any impact from the accident has occurred, this would remain in a very close area to Fukushima because of the low Pu mobility in the marine environment. Since the situation inside the 30 km zone remains unknown, further studies on the determination of Pu isotopes in sediments within this area are required

    Echocardiographic Features in Canine Myxomatous Mitral Valve Disease: An Animal Model for Human Mitral Valve Prolapse

    Myxomatous mitral valve disease (MMVD) is the most common heart disease in dogs and has many similarities to human mitral valve prolapse (MVP). Transthoracic echocardiography is a non-invasive method for making a diagnosis and predicting the progression of heart failure (HF) in dogs and humans with mitral regurgitation (MR). It enables clinicians to detect the mitral valve (MV) lesions, to evaluate MR severity, and to assess its impact on cardiac remodeling, myocardial function, left ventricular (LV) filling pressures, as well as pulmonary arterial pressure. Furthermore, advanced ultrasound technologies such as tissue Doppler imaging (TDI), strain and strain rate imaging, and two-dimensional (2D) speckle tracking echocardiography (STE) provide a better assessment of global and regional myocardial function. Although the severity of MR and HF in dogs with MMVD is being evaluated as similar to human cardiology, the veterinary cardiologists are more focused on the severity of cardiac remodeling and cardiac dysfunction caused by MR, because surgical restoration of defected mitral apparatus is rarely done in dogs. The chapter will review conventional echocardiographic features of MMVD in dogs to provide a better understanding of the similarities and discrepancies between canine MMVD and human MVP to veterinary and human cardiologists and researchers

    Poly(L-histidine)-tagged 5-aminolevulinic acid prodrugs: new photosensitizing precursors of protoporphyrin IX for photodynamic colon cancer therapy

The main drawback associated with ALA-based photodynamic therapy (ALA-PDT) and ALA fluorescence diagnosis results from the hydrophilic nature of ALA and lack of selectivity for tumor versus nontumor cells. The application of certain triggers, such as pH, into conventional sensitizers for controllable 1O2 release is a promising strategy for tumor-targeted treatment.Methods: A series of pH-sensitive ALA-poly(L-histidine) [p(L-His)n] prodrugs were synthesized via ring opening polymerization of 1-benzyl-N-carboxy-L-histidine anhydride initiated by the amine hydrochloride group of ALA itself. As an alternative to ALA for PDT, the synthesized prodrugs were used to treat a cultured human colon cancer HCT116 cell line under different pH conditions. The effect of ALA-p(L-His)n derivatives was evaluated by monitoring the fluorescence intensity of protoporphyrin IX, and measuring the cell survival rate after suitable light irradiation.Results: The cytotoxicity and dark toxicity of ALA and synthesized ALA-p(L-His) derivatives in HEK293T and HCT116 cells in the absence of light at pH 7.4 and 6.8 shows that the cell viability was relatively higher than 100%. ALA-p(L-His)n showed high phototoxicity and selectivity in different pH conditions compared with ALA alone. Because the length of the histidine chain increases in the ALA-p(L-His)n prodrugs, the PDT effect was found to be more powerful. In particular, high phototoxicity was observed when the cells were treated with ALA-p(L-His)15, compared with treatment using ALA alone.Conclusion: The newly synthesized ALA-p(L-His)n derivatives are an effective alternative to ALA for enhancing protoporphyrin IX production and the selectivity of the phototoxic effect in tumor cells.Keywords: 5-aminolevulinic acid, photodynamic therapy, poly(L-histidine), bioconjugate, cancer cell

    Simultaneous deletion of floxed genes mediated by CaMKIIa-Cre in the brain and in male germ cells: application to conditional and conventional disruption of Go-alfa

    The Cre/LoxP system is a well-established approach to spatially and temporally control genetic inactivation. The calcium/calmodulin-dependent protein kinase II alpha subunit (CaMKIIα) promoter limits expression to specific regions of the forebrain and thus has been utilized for the brain-specific inactivation of the genes. Here, we show that CaMKIIα-Cre can be utilized for simultaneous inactivation of genes in the adult brain and in male germ cells. Double transgenic Rosa26+/stop-lacZ::CaMKIIα-Cre+/Cre mice generated by crossing CaMKIIα-Cre+/Cre mice with floxed ROSA26 lacZ reporter (Rosa26+/stop-lacZ) mice exhibited lacZ expression in the brain and testis. When these mice were mated to wild-type females, about 27% of the offspring were whole body blue by X-gal staining without inheriting the Cre transgene. These results indicate that recombination can occur in the germ cells of male Rosa26+/stop-lacZ::CaMKIIα-Cre+/Cre mice. Similarly, when double transgenic Gnao+/f::CaMKIIα-Cre+/Cre mice carrying a floxed Go-alpha gene (Gnaof/f) were backcrossed to wild-type females, approximately 22% of the offspring carried the disrupted allele (GnaoΔ) without inheriting the Cre transgene. The GnaoΔ/Δ mice closely resembled conventional Go-alpha knockout mice (Gnao−/−) with respect to impairment of their behavior. Thus, we conclude that CaMKIIα-Cre mice afford recombination for both tissue- and time-controlled inactivation of floxed target genes in the brain and for their permanent disruption.
