    Finding a third archetypal technical system in architectural phenomenology

    Within the scope of phenomenology and in order to understand architecture, the role of the technical system is as important as those of the purpose of the building or its form. Mass construction and skeletal construction relate to the architectural theory concepts stereotomy and tectonics respectively, which are suitable for describing the fundamental structural and constructive form of architecture. These two systems became established as man built his first shelters and, so far, represented opposite sides of the building industry’s possibilities. The development of new construction techniques and the relationship between research and technology have a great impact on architecture, although new processing methods and materials may not necessarily cause genuine tectonic changes. The technical dimension of architecture is analysed in this work describing how technical elements are built from materials, and then organised in systems. First, the paper examines the division of technical systems in two categories (massive systems and skeletal systems); then it studies timber’s modern production technologies and subsequently the paper critically analyses how these influence the architectural form. The paper concludes that a third archetypal technical system can be perceived with the assembly of surface elements, joining both the multifunctional aspect of the massive systems and the flexibility of the skeletal systems, this third category being fundamental in phenomenological terms

    A new species of Dermopristis Kearn, Whittington & Evans-Gowing, 2010 (Monogenea: Microbothriidae), with observations on associations between the gut diverticula and reproductive system and on the presence of denticles in the nasal fossae of the host Glaucostegus typus (Bennett) (Elasmobranchii: Rhinobatidae)

    Dermopristis cairae n. sp. (Microbothriidae) is described from the skin and possibly from the nasal fossae of the giant shovelnosed ray Glaucostegus typus (Bennett). The new species is distinguished from D. paradoxus Kearn, Whittington & Evans-Gowing, 2010 by its larger size, body shape, lack of transverse ridges on the ventral surface and absence of a seminal receptacle. Extensive short gut branches lie dorsal to the testes and adjacent to the coiled region of the vas deferens and the oo¨type, possibly reflecting high metabolic demand in these areas. Denticles are present in the lining of the nasal fossae of G. typus, providing a firm substrate for the cement-based attachment of a microbothriid. However, confirmation that D. cairae inhabits the nasal fossae of G. typus is required

    Twelve years of clinical practice guideline development, dissemination and evaluation in Canada (1994 to 2005)

    Background - Despite the growing availability of clinical practice guidelines since the early 1990's, little is known about how guideline development and dissemination may have changed over time in Canada. This study compares Canadian guideline development, dissemination, and evaluation in two six year periods from 1994–1999 and 2000–2005. // Methods - Survey of guideline developers who submitted their clinical practice guidelines to the Canadian Medical Association Infobase (a Canadian guideline repository) between 1994 and 2005. Survey items included information about the developers, aspects of guideline development, and dissemination and evaluation activities. // Results - Surveys were sent to the developers of 2341 guidelines in the CMA Infobase over the 12 year period, 1664 surveys were returned (response rate 71%). Of these, 730 unique guidelines were released from 1994–1999, and 630 were released from 2000–2005. Compared to the earlier period, more recent guidelines were being produced in English only. There has been little change in the type of organizations developing guidelines with most developed by provincial and national organizations. In the recent period, developers were more likely to report using computerized search strategies (94% versus 88%), publishing the search strategy (42% versus 34%), reaching consensus using open discussion (95% versus 78%), and evaluating effectiveness of the dissemination strategies (12% versus 6%) and the impact of the CPGs on health outcomes (24% versus 5%). Recent guidelines were less likely to be based on literature reviews (94% versus 99.6%) and were disseminated using fewer strategies (mean 4.78 versus 4.12). // Conclusion - Given that guideline development processes have improved in some areas over the past 12 years yet not in others, ongoing monitoring of guideline quality is required. Guidelines produced more recently in Canada are less likely to be based on a review of the evidence and only about half discuss levels of evidence underlying recommendations. Guideline dissemination and implementation activities have actually decreased. Unfortunately, the potential positive impact on patient health outcomes will not be realized until the recommendations are adopted and acted upon

    Common colorectal cancer risk alleles contribute to the multiple colorectal adenoma phenotype, but do not influence colonic polyposis in FAP

    The presence of multiple (5-100) colorectal adenomas suggests an inherited predisposition, but the genetic aetiology of this phenotype is undetermined if patients test negative for Mendelian polyposis syndromes such as familial adenomatous polyposis (FAP) and MUTYH-associated polyposis (MAP). We investigated whether 18 common colorectal cancer (CRC) predisposition single-nucleotide polymorphisms (SNPs) could help to explain some cases with multiple adenomas who phenocopied FAP or MAP, but had no pathogenic APC or MUTYH variant. No multiple adenoma case had an outlying number of CRC SNP risk alleles, but multiple adenoma patients did have a significantly higher number of risk alleles than population controls (P=5.7 × 10(-7)). The association was stronger in those with ≥10 adenomas. The CRC SNPs accounted for 4.3% of the variation in multiple adenoma risk, with three SNPs (rs6983267, rs10795668, rs3802842) explaining 3.0% of the variation. In FAP patients, the CRC risk score did not differ significantly from the controls, as we expected given the overwhelming effect of pathogenic germline APC variants on the phenotype of these cases. More unexpectedly, we found no evidence that the CRC SNPs act as modifier genes for the number of colorectal adenomas in FAP patients. In conclusion, common colorectal tumour risk alleles contribute to the development of multiple adenomas in patients without pathogenic germline APC or MUTYH variants. This phenotype may have 'polygenic' or monogenic origins. The risk of CRC in relatives of multiple adenoma cases is probably much lower for cases with polygenic disease, and this should be taken into account when counselling such patients

    High Spectral Resolution Observations of the Massive Stars in the Galactic Center

    We present high-resolution near-infrared spectra, obtained with the NIRSPEC spectrograph on the W. M. Keck II Telescope, of a collection of hot, massive stars within the central 25 arcseconds of the Galactic center. We have identified a total of twenty-one emission-line stars, seven of which are new radial velocity detections with five of those being classified as He I emission-line stars for the first time. These stars fall into two categories based on their spectral properties: 1) those with narrow 2.112, 2.113 micron He I doublet absorption lines, and 2) those with broad 2.058 micron He I emission lines. These data have the highest spectral resolution ever obtained for these sources and, as a result, both components of the absorption doublet are separately resolved for the first time. We use these spectral features to measure radial velocities. The majority of the measured radial velocities have relative errors of 20 kms, smaller than those previously obtained with proper-motion or radial velocity measurements for similar stellar samples in the Galactic center. The radial velocities estimated from the He I absorption doublet are more robust than those previously estimated from the 2.058 micron emission line, since they do not suffer from confusion due to emission from the surrounding ISM. Using this velocity information, we agree that the stars are orbiting in a somewhat coherent manner but are not as defined into a disk or disks as previously thought. Finally, multi-epoch radial velocity measurements for IRS 16NE show a change in its velocity presumably due to an unseen stellar companion.Comment: ApJ accepted, 42 pages, 16 figure

    Elevated intracellular cAMP concentration mediates growth suppression in glioma cells.

    Supressed levels of intracellular cAMP have been associated with malignancy. Thus, elevating cAMP through activation of adenylyl cyclase (AC) or by inhibition of phosphodiesterase (PDE) may be therapeutically beneficial. Here, we demonstrate that elevated cAMP levels suppress growth in C6 cells (a model of glioma) through treatment with forskolin, an AC activator, or a range of small molecule PDE inhibitors with differing selectivity profiles. Forskolin suppressed cell growth in a PKA-dependent manner by inducing a G2/M phase cell cycle arrest. In contrast, trequinsin (a non-selective PDE2/3/7 inhibitor), not only inhibited cell growth via PKA, but also stimulated (independent of PKA) caspase-3/-7 and induced an aneuploidy phenotype. Interestingly, a cocktail of individual PDE 2,3,7 inhibitors suppressed cell growth in a manner analogous to forskolin but not trequinsin. Finally, we demonstrate that concomitant targeting of both AC and PDEs synergistically elevated intracellular cAMP levels thereby potentiating their antiproliferative actions
