    On the reliability of inflaton potential reconstruction

    If primordial scalar and tensor perturbation spectra can be inferred from observations of the cosmic background radiation and large-scale structure, then one might hope to reconstruct a unique single-field inflaton potential capable of generating the observed spectra. In this paper we examine conditions under which such a potential can be reliably reconstructed. For it to be possible at all, the spectra must be well fit by a Taylor series expansion. A complete reconstruction requires a statistically-significant tensor mode to be measured in the microwave background. We find that the observational uncertainties dominate the theoretical error from use of the slow-roll approximation, and conclude that the reconstruction procedure will never insidiously lead to an irrelevant potential.Comment: 16 page LaTeX file with eight postscript figures embedded with epsf; no special macros neede

    A Hamilton-Jacobi approach to non-slow-roll inflation

    I describe a general approach to characterizing cosmological inflation outside the standard slow-roll approximation, based on the Hamilton-Jacobi formulation of scalar field dynamics. The basic idea is to view the equation of state of the scalar field matter as the fundamental dynamical variable, as opposed to the field value or the expansion rate. I discuss how to formulate the equations of motion for scalar and tensor fluctuations in situations where the assumption of slow roll is not valid. I apply the general results to the simple case of inflation from an ``inverted'' polynomial potential, and to the more complicated case of hybrid inflation.Comment: 21 pages, RevTeX (minor revisions to match published version

    Probing Planckian physics: resonant production of particles during inflation and features in the primordial power spectrum

    The phenomenon of resonant production of particles {\it after} inflation has received much attention in the past few years. In a new application of resonant production of particles, we consider the effect of a resonance {\em during} inflation. We show that if the inflaton is coupled to a massive particle, resonant production of the particle during inflation modifies the evolution of the inflaton, and may leave an imprint in the form of sharp features in the primordial power spectrum. Precision measurements of microwave background anisotropies and large-scale structure surveys could be sensitive to the features, and probe the spectrum of particles as massive as the Planck scale.Comment: 19 pages, 11 eps figure

    Retrieving sequences of enzymes experimentally characterized but erroneously annotated : the case of the putrescine carbamoyltransferase

    BACKGROUND: Annotating genomes remains an hazardous task. Mistakes or gaps in such a complex process may occur when relevant knowledge is ignored, whether lost, forgotten or overlooked. This paper exemplifies an approach which could help to ressucitate such meaningful data. RESULTS: We show that a set of closely related sequences which have been annotated as ornithine carbamoyltransferases are actually putrescine carbamoyltransferases. This demonstration is based on the following points : (i) use of enzymatic data which had been overlooked, (ii) rediscovery of a short NH(2)-terminal sequence allowing to reannotate a wrongly annotated ornithine carbamoyltransferase as a putrescine carbamoyltransferase, (iii) identification of conserved motifs allowing to distinguish unambiguously between the two kinds of carbamoyltransferases, and (iv) comparative study of the gene context of these different sequences. CONCLUSIONS: We explain why this specific case of misannotation had not yet been described and draw attention to the fact that analogous instances must be rather frequent. We urge to be especially cautious when high sequence similarity is coupled with an apparent lack of biochemical information. Moreover, from the point of view of genome annotation, proteins which have been studied experimentally but are not correlated with sequence data in current databases qualify as "orphans", just as unassigned genomic open reading frames do. The strategy we used in this paper to bridge such gaps in knowledge could work whenever it is possible to collect a body of facts about experimental data, homology, unnoticed sequence data, and accurate informations about gene context

    The cost-effectiveness of providing antenatal lifestyle advice for women who are overweight or obese: the LIMIT randomised trial

    Background: Overweight and obesity during pregnancy is common, although robust evidence about the economic implications of providing an antenatal dietary and lifestyle intervention for women who are overweight or obese is lacking. We conducted a health economic evaluation in parallel with the LIMIT randomised trial. Women with a singleton pregnancy, between 10+0-20+0weeks, and BMI ≥ 25 kg/m2were randomised to Lifestyle Advice (a comprehensive antenatal dietary and lifestyle intervention) or Standard Care. The economic evaluation took the perspective of the health care system and its patients, and compared costs encountered from the additional use of resources from time of randomisation until six weeks postpartum. Increments in health outcomes for both the woman and infant were considered in the cost-effectiveness analysis. Mean costs and effects in the treatment groups allocated at randomisation were compared, and incremental cost effectiveness ratios (ICERs) and confidence intervals (95%) calculated. Bootstrapping was used to confirm the estimated confidence intervals, and to generate acceptability curves representing the probability of the intervention being cost-effective at alternative monetary equivalent values for the outcomes avoiding high infant birth weight, and respiratory distress syndrome. Analyses utilised intention to treat principles. Results: Overall, the increase in mean costs associated with providing the intervention was offset by savings associated with improved immediate neonatal outcomes, rendering the intervention cost neutral (Lifestyle Advice Group 11261.19±14573.97 versus Standard Care Group 11306.70±14562.02; p=0.094). Using a monetary value of 20,000asathresholdvalueforavoidinganadditionalinfantwithbirthweightabove4kg,theprobabilitythattheantenatalinterventioniscost−effectiveis0.85,whichincreasesto0.95whenthethresholdmonetaryvalueincreasesto20,000 as a threshold value for avoiding an additional infant with birth weight above 4 kg, the probability that the antenatal intervention is cost-effective is 0.85, which increases to 0.95 when the threshold monetary value increases to 45,000. Conclusions: Providing an antenatal dietary and lifestyle intervention for pregnant women who are overweight or obese is not associated with increased costs or cost savings, but is associated with a high probability of cost effectiveness. Ongoing participant follow-up into childhood is required to determine the medium to long-term impact of the observed, short-term endpoints, to more accurately estimate the value of the intervention on risk of obesity, and associated costs and health outcomes

    Density and Graviton Perturbations in the Cosmic Microwave Background

    This paper evaluates and compares the gravitational wave and density perturbation contributions to the cosmic microwave background radiation, on the basis of the same power law inflationary model. The inflation to radiation transition is treated as instantaneous, but a model is constructed to allow for a smooth transition from the radiation to the matter dominated eras. The equations are numerically investigated and integrated, without any basic approximations being made. Use is made of the synchronous gauge, with appropriate gauge invariant variables, thus eliminating any confusion arising from unphysical gauge modes. We find a non- negligible gravitational wave contribution, which becomes dominant for a power law expansion with exponent q<13q < 13. We also explore the dependence of our results with the main characteristic of the transition region, its length.Comment: 19 page

    Height and timing of growth spurt during puberty in young people living with vertically acquired HIV in Europe and Thailand.

    OBJECTIVE: The aim of this study was to describe growth during puberty in young people with vertically acquired HIV. DESIGN: Pooled data from 12 paediatric HIV cohorts in Europe and Thailand. METHODS: One thousand and ninety-four children initiating a nonnucleoside reverse transcriptase inhibitor or boosted protease inhibitor based regimen aged 1-10 years were included. Super Imposition by Translation And Rotation (SITAR) models described growth from age 8 years using three parameters (average height, timing and shape of the growth spurt), dependent on age and height-for-age z-score (HAZ) (WHO references) at antiretroviral therapy (ART) initiation. Multivariate regression explored characteristics associated with these three parameters. RESULTS: At ART initiation, median age and HAZ was 6.4 [interquartile range (IQR): 2.8, 9.0] years and -1.2 (IQR: -2.3 to -0.2), respectively. Median follow-up was 9.1 (IQR: 6.9, 11.4) years. In girls, older age and lower HAZ at ART initiation were independently associated with a growth spurt which occurred 0.41 (95% confidence interval 0.20-0.62) years later in children starting ART age 6 to 10 years compared with 1 to 2 years and 1.50 (1.21-1.78) years later in those starting with HAZ less than -3 compared with HAZ at least -1. Later growth spurts in girls resulted in continued height growth into later adolescence. In boys starting ART with HAZ less than -1, growth spurts were later in children starting ART in the oldest age group, but for HAZ at least -1, there was no association with age. Girls and boys who initiated ART with HAZ at least -1 maintained a similar height to the WHO reference mean. CONCLUSION: Stunting at ART initiation was associated with later growth spurts in girls. Children with HAZ at least -1 at ART initiation grew in height at the level expected in HIV negative children of a comparable age
