BCC vs. HCP - The Effect of Crystal Symmetry on the High Temperature Mobility of Solid He
We report results of torsional oscillator (TO) experiments on solid He at
temperatures above 1K. We have previously found that single crystals, once
disordered, show some mobility (decoupled mass) even at these rather high
temperatures. The decoupled mass fraction with single crystals is typically 20-
30%. In the present work we performed similar measurements on polycrystalline
solid samples. The decoupled mass with polycrystals is much smaller, 1%,
similar to what is observed by other groups. In particular, we compared the
properties of samples grown with the TO's rotation axis at different
orientations with respect to gravity. We found that the decoupled mass fraction
of bcc samples is independent of the angle between the rotation axis and
gravity. In contrast, hcp samples showed a significant difference in the
fraction of decoupled mass as the angle between the rotation axis and gravity
was varied between zero and 85 degrees. Dislocation dynamics in the solid

The mitochondrial Na+/Ca2+ exchanger upregulates glucose dependent Ca2+ signalling linked to insulin secretion.
Mitochondria mediate dual metabolic and Ca(2+) shuttling activities. While the former is required for Ca(2+) signalling linked to insulin secretion, the role of the latter in Ξ² cell function has not been well understood, primarily because the molecular identity of the mitochondrial Ca(2+) transporters were elusive and the selectivity of their inhibitors was questionable. This study focuses on NCLX, the recently discovered mitochondrial Na(+)/Ca(2+) exchanger that is linked to Ca(2+) signalling in MIN6 and primary Ξ² cells. Suppression either of NCLX expression, using a siRNA construct (siNCLX) or of its activity, by a dominant negative construct (dnNCLX), enhanced mitochondrial Ca(2+) influx and blocked efflux induced by glucose or by cell depolarization. In addition, NCLX regulated basal, but not glucose-dependent changes, in metabolic rate, mitochondrial membrane potential and mitochondrial resting Ca(2+). Importantly, NCLX controlled the rate and amplitude of cytosolic Ca(2+) changes induced by depolarization or high glucose, indicating that NCLX is a critical and rate limiting component in the cross talk between mitochondrial and plasma membrane Ca(2+) signalling. Finally, knockdown of NCLX expression was followed by a delay in glucose-dependent insulin secretion. These findings suggest that the mitochondrial Na(+)/Ca(2+) exchanger, NCLX, shapes glucose-dependent mitochondrial and cytosolic Ca(2+) signals thereby regulating the temporal pattern of insulin secretion in Ξ² cells
Fairness and Efficiency in DAG-based Cryptocurrencies
Bitcoin is a decentralised digital currency that serves as an alternative to
existing transaction systems based on an external central authority for
security. Although Bitcoin has many desirable properties, one of its
fundamental shortcomings is its inability to process transactions at high
rates. To address this challenge, many subsequent protocols either modify the
rules of block acceptance (longest chain rule) and reward, or alter the
graphical structure of the public ledger from a tree to a directed acyclic
graph (DAG). Motivated by these approaches, we introduce a new general
framework that captures ledger growth for a large class of DAG-based
implementations. With this in hand, and by assuming honest miner behaviour, we
(experimentally) explore how different DAG-based protocols perform in terms of
fairness, i.e., if the block reward of a miner is proportional to their hash
power, as well as efficiency, i.e. what proportion of user transactions a
ledger deems valid after a certain length of time. Our results demonstrate
fundamental structural limits on how well DAG-based ledger protocols cope with
a high transaction load. More specifically, we show that even in a scenario
where every miner on the system is honest in terms of when they publish blocks,
what they point to, and what transactions each block contains, fairness and
efficiency of the ledger can break down at specific hash rates if miners have
differing levels of connectivity to the P2P network sustaining the protocol
Inferring short-term volatility indicators from Bitcoin blockchain
In this paper, we study the possibility of inferring early warning indicators
(EWIs) for periods of extreme bitcoin price volatility using features obtained
from Bitcoin daily transaction graphs. We infer the low-dimensional
representations of transaction graphs in the time period from 2012 to 2017
using Bitcoin blockchain, and demonstrate how these representations can be used
to predict extreme price volatility events. Our EWI, which is obtained with a
non-negative decomposition, contains more predictive information than those
obtained with singular value decomposition or scalar value of the total Bitcoin
transaction volume
Harnessing Soluble NK Cell Killer Receptors for the Generation of Novel Cancer Immune Therapy
The natural cytotoxic receptors (NCRs) are a unique set of activating proteins expressed mainly on the surface of natural killer (NK) cells. The NCRs, which include three members; NKp46, NKp44 and NKp30, are critically involved in NK cytotoxicity against different targets, including a wide range of tumor cells derived from various origins. Even though the tumor ligands of the NCRs have not been identified yet, the selective manner by which these receptors target tumor cells may provide an excellent basis for the development of novel anti-tumor therapies. To test the potential use of the NCRs as anti-tumor agents, we generated soluble NCR-Ig fusion proteins in which the constant region of human IgG1 was fused to the extracellular portion of the receptor. We demonstrate, using two different human prostate cancer cell lines, that treatment with NKp30-Ig, dramatically inhibits tumor growth in vivo. Activated macrophages were shown to mediate an ADCC response against the NKp30-Ig coated prostate cell lines. Finally, the Ig fusion proteins were also demonstrated to discriminate between benign prostate hyperplasia and prostate cancer. This may provide a novel diagnostic modality in the difficult task of differentiating between these highly common pathological conditions
On The Mobile Behavior of Solid He at High Temperatures
We report studies of solid helium contained inside a torsional oscillator, at
temperatures between 1.07K and 1.87K. We grew single crystals inside the
oscillator using commercially pure He and He-He mixtures containing
100 ppm He. Crystals were grown at constant temperature and pressure on the
melting curve. At the end of the growth, the crystals were disordered,
following which they partially decoupled from the oscillator. The fraction of
the decoupled He mass was temperature and velocity dependent. Around 1K, the
decoupled mass fraction for crystals grown from the mixture reached a limiting
value of around 35%. In the case of crystals grown using commercially pure
He at temperatures below 1.3K, this fraction was much smaller. This
difference could possibly be associated with the roughening transition at the
solid-liquid interface.Comment: 15 pages, 6 figure
Game On? Smoking Cessation Through the Gamification of mHealth: A Longitudinal Qualitative Study
BACKGROUND: Finding ways to increase and sustain engagement with mHealth interventions has become a challenge during application development. While gamification shows promise and has proven effective in many fields, critical questions remain concerning how to use gamification to modify health behavior. OBJECTIVE: The objective of this study is to investigate how the gamification of mHealth interventions leads to a change in health behavior, specifically with respect to smoking cessation. METHODS: We conducted a qualitative longitudinal study using a sample of 16 smokers divided into 2 cohorts (one used a gamified intervention and the other used a nongamified intervention). Each participant underwent 4 semistructured interviews over a period of 5 weeks. Semistructured interviews were also conducted with 4 experts in gamification, mHealth, and smoking cessation. Interviews were transcribed verbatim and thematic analysis undertaken. RESULTS: Results indicated perceived behavioral control and intrinsic motivation acted as positive drivers to game engagement and consequently positive health behavior. Importantly, external social influences exerted a negative effect. We identified 3 critical factors, whose presence was necessary for game engagement: purpose (explicit purpose known by the user), user alignment (congruency of game and user objectives), and functional utility (a well-designed game). We summarize these findings in a framework to guide the future development of gamified mHealth interventions. CONCLUSIONS: Gamification holds the potential for a low-cost, highly effective mHealth solution that may replace or supplement the behavioral support component found in current smoking cessation programs. The framework reported here has been built on evidence specific to smoking cessation, however it can be adapted to health interventions in other disease categories. Future research is required to evaluate the generalizability and effectiveness of the framework, directly against current behavioral support therapy interventions in smoking cessation and beyond
