    The Krushinsky – Molodkina genetic rat strain as a unique experimental model of seizure states

    The study of genetic mechanisms, which underlie normal and abnormal behavioral traits, are important not only for fundamental knowledge of CNS function, but also for human well-being, as well as in the aspect of treatment of brain diseases. Accumulation of knowledge concerning the functions of genes, which are expressed in the CNS and are involved in the specific brain functions determined now by the success of molecular genetics, but it could not overshadow the importance of phenotype expression investigation of genetically determined traits, especially pathological ones. Epilepsy, as one of CNS diseases, occupies an important place in the row of these studies. In spite of numerous anticonvulsant drugs a significant proportion of epilepsy cases are still resistant to drug treatments. It means that the study of various aspects of epileptogenesis using animal models should be welcomed as it will help to elucidate those aspects of increased seizure proneness that are now out of scope of research attention. The distinct trait of this domain, which is not easy to analyze genetically, is audiogenic epilepsy (the seizure attack in response to loud sound). The important feature of audiogenic epilepsy is that the intense tonic seizures could be induced and analyzed repeatedly which makes it possible to induce the seizures repeatedly in the course of chronical experiments with the same animal, which is not possible in the cases of pharmacologically or electrically induced seizures. The Krushinsky – Molodkina (KM) inbred rat strain, which was the first among strains selected for audiogenic epilepsy, is briefly characterized. The specific seizure pattern, the rodent proneness for audiogenic epilepsy, as well as abnormal postictal states (catalepsy, in particular) illustrate the importance of this phenomenon not only as a model of certain brain pathology, but also as a matter of general biological importance. The importance of genetic background factor (demonstrated in the case of study of comorbidity (audiogenic epilepsy, anxiety and depression)) could be crucial in the studies of other CNS anomalies.     TELECOMMUNICATION TECHNOLOGIES FOR MONITORING OF FUNCTIONAL STATUS OF DRIVERS DURING THEIR WORK

    The article is devoted to development of modern telemetric methods for an on-line evaluation of functional status of vehicle drivers during their work. The study of occupational load effect on cardiovascular system was performed with the use of mobile devices included portable sensors and storage device to collect information (smartphone). The developed computer programs were used to calculate spectral indices of heart rhythm variability (HRV). A high self-descriptiveness of wavelet transform for identification offunctional status of cardiovascular system was shown. The reduction of HRV, increase of sympathetic component in regulation and increase of vegetative balance index were observed in drivers by fatigue. The intensity of HRV changes met with occupational load level and characterized degree of individual response as well as allowed to detect exposure induced the stress. A wireless cardiorhythmography is a perspective method for creating personal monitoring system for increasing quality of the drivers' life andfor their health preservation

    The study of the endothelial dysfunction and the vegetative homeostasis have been made in patients with hyperplastic processes in genitals. The examination included 60 women with hyperplastic processes in genitals and 30 gynaecologicaly healthy volunteers. The endothelial dysfunction, prevalence influence sympathetic nervous system have been detected in women with hyperplastic processes in genitals. The optimum results of the therapy have been obtain by inclusion clinical use buserelin.У пациенток с гиперпластическими заболеваниями гениталий проведено изучение эндотелиальной регуляции тонуса сосудов в сопоставлении с показателями вегетативного гомеостаза. В исследование были включены 60 женщин с гиперпластическими процессами гениталий и 30 гинекологически здоровых женщин, составляющих контрольную группу. У женщин с гинекологической патологией выявлены дисфункция эндотелия, снижение адаптационных резервов организма и преобладание влияния симпатического отдела ВНС на деятельность сердца. Оптимальные результаты по достижению нормализации гормонального гомеостаза, параметров ВНС и показателей функции эндотелия у женщин с гиперпластическими заболеваниями гениталий достигнуты при включении в схему лечения А-ГнРГ (бусерелина)

    Carcinogenic pollution of air as occupational risk factor among driver and traffic policemen

    The study ot air pollution in the breathing zone ot urban bus driver and traffic-controller policemen showed the presence of carcinogenic substances. The authors determined carcinogenic risk in these occupational groups, carcinogenic risk for urban v bus driver was at permissible levels, carcinogenic risk for traffic-controller policemen was 1.3X10‾⁴ that provided evidence about potential carcinogenic risk. As benzene and formaldehyde are main carcinogenic substances caused carcinogenic risk, it should be perform monitoring of these substances during evaluation of working condition in the occupational groups.Исследование загрязненности воздуха в зоне дыхания водителей городских автобусов и автоинспекторов-регулировщиков дорожного движения выявило присутствие в нем канцерогенных веществ. Определен канцерогенный риск для этих профессиональных групп, который у водителей находится в допустимых пределах, а у регулировщиков составляет 1,3x10‾⁴, что свидетельствует о потенциальной канцерогенной опасности. Поскольку основными веществами. определяющими канцерогенный риск, являются бензол и формальдегид, необходим мониторинг этих веществ при оценке условий труда данных профессиональных групп

    Analysis of recombinant VEGF gene expression by genetically modified umbilical cord blood mononuclear cells in experiment in vivo

    To obtain a significant therapeutic effect transplanted genetically modified cells should have an enhanced ability to survive and active expression of the therapeutic gene. In this paper, by using immunofluorescent staining we investigated the functional activity of the gene-cell formulation designed to deliver a therapeutic gene into the area of regeneration. As a model we used transgenic SOD1-G93A mice with amyotrophic lateral sclerosis phenotype which received xenotransplantation of human umbilical cord blood mononuclear cells, genetically modified with adenoviral expression vector encoding vascular endothelial growth factor (VEGF) and the reporter green fluorescent protein (EGFP). Results of the study allowed to establish not only the duration of survival of transplanted cells, but also the efficiency of expression of recombinant genes in genetically modified cells in vivo. Double immunofluorescent staining with antibodies against human nuclear antigen HNA and VEGF detected HNA+/VEGF+ cells in the terminal stage of disease 15 weeks after transplantation. These data suggest that genetically modified umbilical cord blood mononuclear cells, transplanted into SOD1-G93A transgenic mice, are able to penetrate the blood-brain barrier and migrate into the area of degeneration of nerve tissue and survive from the time of transplantation until the death of animals at the terminal stage of disease. At that time adenoviral expression vector encoding therapeutic gene is functionally active in transplanted cells, and secretory products of recombinant gene act on target cells by a paracrine mechanism

    The years-long neglect of the interests of the real economy, particularly the non-oil sector interests, resulted in the formation of a speculative model of the Russian economy where the cash flows are circulating primarily in the financial sector. The current monetary policy that leaves aside the interests of the economic growth, the welfare growth and the employment fits quite well within this model. However, the low inflation cannot be regarded a goal in itself achieved through stifling the economic growth. The current crisis of the Russian economy has exposed the problem of the priority development of the domestic financial sector to the detriment of the real sector and the Russian economy as a whole. The speculative model of the economy developed in the past quarter-century has led to the disintegration of the financial and real sectors of the economy, which neither ensures the financial stability nor promotes the economic growth. Based on the research findings, the paper formulates proposals on how to prevent the “slippage” of the economy into an uncontrollable state and ensure its sustainable development, including the adjustment and coordination of the monetary, foreign exchange (forex) and fiscal policies of Russia.Многолетнее игнорирование интересов реального сектора экономики, особенно его несырьевого сектора, привело к формированию спекулятивной модели российской экономики, в которой денежные потоки циркулируют преимущественно в финансовом секторе. Такой модели соответствует и проводимая денежно-кредитная политика, за рамками которой остаются интересы экономического роста, роста благосостояния населения и его занятости. Вместе с тем достижение низкого уровня инфляции не может являться самоцелью, достигаемой за счет сдерживания экономического роста. Текущий кризис российской экономики обнажил проблемы приоритетного развития отечественного финансового сектора в ущерб интересам реального сектора и российской экономики в целом. Сформированная в прошедшую четверть века спекулятивная модель экономики страны привела к дезинтеграции финансового и реального секторов экономики, что не способствует ни достижению финансовой стабильности, ни обеспечению экономического роста. В статье на основе проведенного исследования сформулированы предложения по предотвращению «сползания» экономики в неуправляемое состояние и обеспечению ее устойчивого развития, включая корректировку и координацию денежно-кредитной, валютной и бюджетно-налоговой политик России

    Embracing Monogenic Parkinson's Disease: The MJFF Global Genetic PD Cohort

    Background: As gene-targeted therapies are increasingly being developed for Parkinson's disease (PD), identifying and characterizing carriers of specific genetic pathogenic variants is imperative. Only a small fraction of the estimated number of subjects with monogenic PD worldwide are currently represented in the literature and availability of clinical data and clinical trial-ready cohorts is limited. Objective: The objectives are to (1) establish an international cohort of affected and unaffected individuals with PD-linked variants; (2) provide harmonized and quality-controlled clinical characterization data for each included individual; and (3) further promote collaboration of researchers in the field of monogenic PD. Methods: We conducted a worldwide, systematic online survey to collect individual-level data on individuals with PD-linked variants in SNCA, LRRK2, VPS35, PRKN, PINK1, DJ-1, as well as selected pathogenic and risk variants in GBA and corresponding demographic, clinical, and genetic data. All registered cases underwent thorough quality checks, and pathogenicity scoring of the variants and genotype–phenotype relationships were analyzed. Results: We collected 3888 variant carriers for our analyses, reported by 92 centers (42 countries) worldwide. Of the included individuals, 3185 had a diagnosis of PD (ie, 1306 LRRK2, 115 SNCA, 23 VPS35, 429 PRKN, 75 PINK1, 13 DJ-1, and 1224 GBA) and 703 were unaffected (ie, 328 LRRK2, 32 SNCA, 3 VPS35, 1 PRKN, 1 PINK1, and 338 GBA). In total, we identified 269 different pathogenic variants; 1322 individuals in our cohort (34%) were indicated as not previously published. Conclusions: Within the MJFF Global Genetic PD Study Group, we (1) established the largest international cohort of affected and unaffected individuals carrying PD-linked variants; (2) provide harmonized and quality-controlled clinical and genetic data for each included individual; (3) promote collaboration in the field of genetic PD with a view toward clinical and genetic stratification of patients for gene-targeted clinical trials. © 2023 The Authors.
