    Widespread occurrence of Mycobacterium tuberculosis DNA from 18th-19th century Hungarians

    A large number (265) of burials from 1731-1838 were discovered in sealed crypts of the Dominican Church, Vac, Hungary in 1994. Many bodies were naturally mummified, so that both soft tissues and bones were available. Contemporary archives enabled the determination of age at death, and the identification of family groups. In some cases, symptoms before death were described and, occasionally, occupation. Initial radiological examination of a small number of individuals had indicated calcified lung lesions and demonstrable acid-fast bacteria suggestive of tuberculosis infection. Tuberculosis was endemic in 18th-19th century Europe, so human remains should contain detectable Mycobacterium tuberculosis complex (MTB) DNA, enabling comparisons with modern isolates. Therefore, a comprehensive examination of 168 individuals for the presence of MTB DNA was undertaken. Specific DNA amplification methods for MTB showed that 55% of individuals were positive and that the incidence varied according to age at death and sampling site in the body. Radiographs were obtained from 27 individuals and revealed an association between gross pathology and the presence of MTB DNA. There was an inverse relationship between PCR positivity and MTB target sequence size. In some cases, the preservation of MTB DNA was excellent, and several target gene sequences could be detected from the same sample. This information, combined with MTB DNA sequencing data and molecular typing techniques, will enable us to study the past epidemiology of TB infection, and extends the timeframe for studying changes in molecular fingerprints.     Detection and characterization of Mycobacterium tuberculosis DNA in 18th Century Hungarians with pulmonary and extra-pulmonary tuberculosis

    Skeletal and naturally mummified tissues from a previously archived group of 18th century Hungarian remains were examined for the presence of Mycobacterium tuberculosis complex (MTB) DNA, using specific nested PCR for the IS6110 locus. Paleopathological changes in bones and from radiographs were noted in a minority of cases. Overall, specimens from 157/232 (67.7 %) of individuals proved positive, ranging from 20/43 (46.5 %) in children, 26/29 in middle-age (89.7 %) and 32/46 individuals aged 65-95 years (69.6 %). Single samples gave a positive result in 67/120 of cases (55.8 %). Most were ribs where the surface adjacent to lungs and pleura was sampled. When multiple sites were examined, 73/93 (78.5 %) individuals were positive; most of these had MTB only in the pulmonary region but 26 had disseminated disease (35.6 %) and 12 (16.4 %) had extra-pulmonary disease only. To distinguish M. tuberculosis from Mycobacterium bovis, well-preserved positive samples were examined for several additional genetic loci including the TbD1 deletion – characteristic of modern European strains of M. tuberculosis, and spoligotyped. No evidence other than of human M. tuberculosis was found, but different strains were detected. Tuberculosis was widespread in this community and whilst some individuals succumbed early in life, the majority co-existed with the infection. Therefore, this study may lead to the identification of host alleles and MTB strains associated with active and latent disease

    New skeletal tuberculosis cases in past populations from Western Hungary (Transdanubia)

    The distribution, antiquity and epidemiology of tuberculosis (TB) have previously been studied in osteoarchaeological material in the eastern part of Hungary, mainly on the Great Plain. The purpose of this study is to map the occurrence of skeletal TB in different centuries in the western part of Hungary, Transdanubia, and to present new cases we have found. Palaeopathological analysis was carried out using macroscopic observation supported by radiographic and molecular methods. A large human osteoarchaeological sample (n = 5684) from Transdanubian archaeological sites ranging from the 2nd to the 18th centuries served as a source of material. Spinal TB was observed in seven individuals (in three specimens with Pott's disease two of which also had cold abscess) and hip TB was assumed in one case. The results of DNA for Mycobacterium tuberculosis were positive in seven of the eight cases identified by paleopathology, and negative in the assumed case of hip TB. However, the molecular results are consistent with highly fragmented DNA, which limited further analysis. Based on the present study and previously published cases, osteotuberculosis was found in Transdanubia mainly during the 9th–13th centuries. However, there are no signs of TB in many other 9th–13th century sites, even in those that lie geographically close to those where osteotuberculous cases were found. This may be due to a true absence of TB caused by the different living conditions, way of life, or origin of these populations. An alternative explanation is that TB was present in some individuals with no typical paleopathology, but that death occurred before skeletal morphological features could develop

    The aim of this research was to know the relationship between the implementation of family health care function to nutritional status of children aged under five years in Padang Bulan village, Pekanbaru. This research was quantitative research design with descriptive cross sectional correlational approach. The total of sample were 82 mothers who have children and conducted by accidental sampling. The instrument used weight scale (Kg) ang height scale (Cm) which interpreted by standard of WHO-NCHS (2005) as the variable of nutritional status. The instrument used the questionnaire to measure the function of family health care which had been tested the validity and reliability. This research used univariate analyse to determine the distribution of frequencies and bivariate analyse Kolmogorof Smirnov. The result of this study showed that most of children had good nutritional status and most of mothers had implemented the family health care functions excellently. It showed, there is a relationship of the implementation of family health care functions to nutritional status of children aged under five years (p value= 0,014). Based on this result, it is recommended to each mother to improve the nutritional status of children aged by doing the function of health family care well

    A tyrosine-rich amelogenin peptide promotes neovasculogenesis in vitro and ex vivo

    The formation of new blood vessels has been shown to be fundamental in the repair of many damaged tissues, and we have recently shown that the adult human periodontal ligament contains multipotent stem/progenitor cells that are capable of undergoing vasculogenic and angiogenic differentiation in vitro and ex vivo. Enamel matrix protein (EMP) is a heterogeneous mixture of mainly amelogenin-derived proteins produced during tooth development and has been reported to be sometimes effective in stimulating these processes, including in clinical regeneration of the periodontal ligament. However, the identity of the specific bioactive component of EMP remains unclear. In the present study we show that, while the high-molecular-weight Fraction A of enamel matrix derivative (a heat-treated form of EMP) is unable to stimulate the vasculogenic differentiation of human periodontal ligament cells (HPC) in vitro, the low-molecular-weight Fraction C significantly up-regulates the expression of the endothelial markers VEGFR2, Tie-1, Tie-2, VE-cadherin and vWF and markedly increases the internalization of low-density lipoprotein. Furthermore, we also demonstrate, for the first time, that the synthetic homolog of the 45-amino acid tyrosine-rich amelogenin peptide (TRAP) present in Fraction C is likely to be responsible for its vasculogenesis-inducing activity. Moreover, the chemically synthesized TRAP peptide is also shown here to be capable of up-regulating the angiogenic differentiation of the HPC, based on its marked stimulation of in vitro cell migration and tubule formation and of blood vessel formation assay in a chick embryo chorioallantoic membrane model ex vivo. This novel peptide, and modified derivatives, might thereby represent a new class of regenerative drug that has the ability to elicit new blood vessel formation and promote wound healing in vivo. © 2013 Acta Materialia Inc.     Notes on Ipomoea L. (Convolvulaceae) in Cuba and neighbouring islands with a checklist of species found in Cuba

    An updated checklist of species of Ipomoea L. found in Cuba is presented with analysis of the different elements represented. I. alterniflora Griseb. is defined broadly to include I. obtusata Griseb. and I. excisa Urb. and its differences from the little-known I. cubensis (House) Urb. are discussed. I. calophylla C. Wright ex Griseb. is reinstated as the correct name for the species generally known as I. lacteola House. I. praecox C. Wright is recognised as a distinct species from I. argentifolia A. Rich. and images are provided to help distinguish the two species. I. flavopurpurea Urb. and I. dajabonensis Alain are shown to be conspecific with I. longeramosa Choisy, whose disjunct distribution is mapped and discussed. The little-known I. montecristina Hadač is described and illustrated and the cited collections show it to be locally common in the Guantánamo region. I. microdonta J. R. I. Wood and Scotland is described as new from Camagüey in central Cuba. Eight species endemic to Cuba collected by Ekman and described by Urban in 1924 – 25 are evaluated but only two, I. balioclada Urb. and I. erosa Urb., are deemed to warrant recognition as distinct endemic species. The origin and typification of I. horsfalliae Hook. are discussed and an epitype designated. Cultivated plants named I. horsfalliae occur in many tropical countries including Cuba but their extreme variation suggests hybrid origin. Four species from Jamaica, I. rubella House, I. lineolata Urb., I. carmesina Proctor and the Jamaican plant called I. horsfalliae are treated as synonyms of a variable I. lineolata, which is endemic to the island. I. saxicola Proctor is treated as var. saxicola J. R. I. Wood and Scotland of I. ternata Jacq. I. cyanantha Griseb. is treated as a synonym of I. lindenii M. Martens and Galeotti. Lectotypes are designated for I. cyanantha, I. lindenii, I. praecox, I. punctata C. Wright, I. geranioides Meisn. and I. grisebachii Urb
