Realizing quantum gates with optically-addressable Yb ion qudits
The use of multilevel information carriers, also known as qudits, is a
promising path for exploring scalability of quantum computing devices. In this
work, we present a proof-of-principle realization of a quantum processor that
uses optically-addressed Yb ion qudits in a linear trap. The rich
level structure of Yb ions makes it possible to use the Zeeman
sublevels of the quadrupole clock transition at 435.5 nm for efficient and
robust qudit encoding. We demonstrate the realization of the universal set of
gates consisting of single-qudit rotations and two-qudit entangling operation
with a two-ququart system, which is formally equivalent to a universal
gate-based four-qubit processor. Our results paves a way towards further
studies of more efficient implementations of quantum algorithms with
trapped-ion-based processors.Comment: 9 pages, 3 figure
EMERGENCY PROPHYLAXIS OF EXPERIMENTAL MELIOIDOSIS USING SYNTHETIC IMMUNOMODULATORS AND HETEROLOGOUS VACCINES
Melioidosis is a particularly dangerous infection caused by Burkholderia pseudomallei , against which a vaccine has not yet been created. In this regard, the development of effective treatment regimens and emergency prevention of melioidosis is very relevant. To improve the effectiveness of emergency prophylaxis of melioidosis, synthetic peptides (bestim, imunofan) and thiopoietin preparation glutoxim were used when combined with the antibiotic doxycycline. In addition, in experiments on white mice, the ability of heterologous vaccines (plague and tularemia), used in the emergency prevention mode, to increase the resistance of animals to melioidosis infection was assessed. It was shown that the most effective was imunofan, which, when combined with doxycycline, increased by 20% the survival rate of 5LD50 Burkholderia pseudomallei infection and significantly increased the average life span of mice infected with 5-12 LD50 (p < 0.05). The efficiency of use for stimulation of non-specific resistance to melioidosis of a heterologous plague vaccine EV, once administered 1 day prior to infection, protected 90% of mice from 6 LD50 Burkholderia pseudomallei and 60% - with an increase in the infectious dose of the pathogen of melioidosis up to 15 LD50. The same level of protection from melioidosis was provided by a 3 day course of antibiotic therapy with doxycycline. It was concluded that the tularemia vaccine is not suitable for immunostimulation in melioidosis due to its high residual virulence and reactogenicity

Random transgene integration is a powerful tool for developing new genome-wide screening approaches. These techniques have already been used for functional gene annotation by transposon-insertion sequencing, for identification of transcription factor binding sites and regulatory sequences, and for dissecting chromatin position effects. Precise localization of transgenes and accurate artifact filtration are essential for this type of method. To date, many mapping assays have been developed, including Inverse-PCR, TLA, LAM-PCR, and splinkerette PCR. However, none of them is able to ensure localization of both transgeneβs flanking regions simultaneously, which would be necessary for some applications. Here we proposed a cheap and simple NGS-based approach that overcomes this limitation. The developed assay requires using intentionally designed vectors that lack recognition sites of one or a set of restriction enzymes used for DNA fragmentation. By looping and sequencing these DNA fragments, we obtain special data that allows us to link the two flanking regions of the transposon. This can be useful for precise insertion mapping and for screening approaches in the field of chromosome engineering, where chromosomal recombination events between transgenes occur in a cell population. To demonstrate the methodβs feasibility, we applied it for mapping SB transposon integration in the human HAP1 cell line. Our technique allowed us to efficiently localize genomic transposon integrations, which was confirmed via PCR analysis. For practical application of this approach, we proposed a set of recommendations and a normalization strategy. The research represents working out of non-invasive colorectal cancer screening based on DNA integrity testing of patient's fecal. Polymerase chain reaction method and the following polyacrylamide gel electrophoresis in fecal of patient with colorectal cancer detected long DNA fragments which size was 800 n.p. But the same stool DNA testing of control group detected just short DNA fragments which size was less than 200 n.p. The risks of the emergent ecological and technical situation as a result of using liquid ammonia as a refrigerating medium in construction in Far Northern regions have been considered. Affective zone modeling/simulation during ammonia ejection into the atmosphere has been built. A model of reaction between ammonia and plastic frozen ground has been suggested.
