    Efficiency of Xist-mediated silencing on autosomes is linked to chromosomal domain organisation

    BACKGROUND: X chromosome inactivation, the mechanism used by mammals to equalise dosage of X-linked genes in XX females relative to XY males, is triggered by chromosome-wide localisation of a cis-acting non-coding RNA, Xist. The mechanism of Xist RNA spreading and Xist-dependent silencing is poorly understood. A large body of evidence indicates that silencing is more efficient on the X chromosome than on autosomes, leading to the idea that the X chromosome has acquired sequences that facilitate propagation of silencing. LINE-1 (L1) repeats are relatively enriched on the X chromosome and have been proposed as candidates for these sequences. To determine the requirements for efficient silencing we have analysed the relationship of chromosome features, including L1 repeats, and the extent of silencing in cell lines carrying inducible Xist transgenes located on one of three different autosomes. RESULTS: Our results show that the organisation of the chromosome into large gene-rich and L1-rich domains is a key determinant of silencing efficiency. Specifically genes located in large gene-rich domains with low L1 density are relatively resistant to Xist-mediated silencing whereas genes located in gene-poor domains with high L1 density are silenced more efficiently. These effects are observed shortly after induction of Xist RNA expression, suggesting that chromosomal domain organisation influences establishment rather than long-term maintenance of silencing. The X chromosome and some autosomes have only small gene-rich L1-depleted domains and we suggest that this could confer the capacity for relatively efficient chromosome-wide silencing. CONCLUSIONS: This study provides insight into the requirements for efficient Xist mediated silencing and specifically identifies organisation of the chromosome into gene-rich L1-depleted and gene-poor L1-dense domains as a major influence on the ability of Xist-mediated silencing to be propagated in a continuous manner in cis

    Long-Wave Forcing by the Breaking of Random Gravity Waves on a Beach

    This paper presents new laboratory data on long-wave (surf-beat) forcing by the random breaking of shorter gravity water waves on a plane beach. The data include incident and outgoing wave amplitudes, together with shoreline oscillation amplitudes at long-wave frequencies, from which the correlation between forced long waves and short-wave groups is examined. A detailed analysis of the cross-shore structure of the long-wave motion is presented, and the observations are critically compared with existing theories for two-dimensional surf-beat generation. The surf beat shows a strong dependency on normalized surf-zone width, consistent with long-wave forcing by a time-varying breakpoint, with little evidence of the release and reflection of incident bound long waves for the random-wave simulations considered. The seaward-propagating long waves show a positive correlation with incident short-wave groups and are linearly dependent on short-wave amplitude. The phase relationship between the incident bound long waves and radiated free long waves is also consistent with breakpoint forcing. In combination with previous work, the present data suggest that the breakpoint variability may be the dominant forcing mechanism during conditions with steep incident short waves

    'Sinking and swimming in disability coaching': an autoethnographic account of coaching in a new context

    In terms of achieving wider health and social outcomes, sport coaching promises much for young people with disabilities. Despite this promise, the experiences and practices of those coaches who enter the disability sport arena are underexplored. This is particularly so for coaches who operate in community participation rather than competitive elite environments. Accordingly, this paper uses an autoethnographic approach to explore the experiences of a basketball coach (Colum), who enters a youth club for disabled participants for the first time. Utilising observational data, reflective field notes, and interviews, five relativist vignettes are collaboratively constructed to represent Colum’s (a pseudonym) experiences across 12 basketball sessions. The vignettes reveal that the disability and community context disrupted Colum’s normative coaching behaviours. An emotional laborious journey is recounted that includes significant lessons, which may impact coaching practitioners, researchers and sport development officers. In addition, the post-sport context (Atkinson, 2010a) is introduced to differentiate the youth club context from Colum’s normative sport context. Furthermore, the concepts of liminality and ludic, which are novel to extant coaching literature, are introduced to explain how and why Colum struggled to find structure within the context of a youth club for disabled participants

    Temperature scaling in a dense vibro-fluidised granular material

    The leading order "temperature" of a dense two dimensional granular material fluidised by external vibrations is determined. An asymptotic solution is obtained where the particles are considered to be elastic in the leading approximation. The velocity distribution is a Maxwell-Boltzmann distribution in the leading approximation. The density profile is determined by solving the momentum balance equation in the vertical direction, where the relation between the pressure and density is provided by the virial equation of state. The predictions of the present analysis show good agreement with simulation results at higher densities where theories for a dilute vibrated granular material, with the pressure-density relation provided by the ideal gas law, are in error.     Bosons in Disordered Optical Potentials

    In this work we systematically investigate the condensate properties, superfluid properties and quantum phase transitions in interacting Bose gases trapped in disordered optical potentials. We numerically solve the Bose-Hubbard Hamiltonian exactly for different: (a) types of disorder, (b) disorder strengths, and (c) interatomic interactions. The three types of disorder studied are: quasiperiodic disorder, uniform random disorder and random speckle-type disorder. We find that the Bose glass, as identified by Fisher et al [Phys. Rev.     Revisional Surgery for Hallux Valgus with Serial Osteotomies at Two Levels

    The aetiology and form of hallux valgus (HV) is varied with many corrective procedures described. We report a 39-year-old woman, previously treated with a Chevron osteotomy, who presented with recurrent right HV, metatarsus primus varus, and associated bunion. Osteotomies were performed at two levels as a revisional procedure. This report highlights (1) limitations of the Chevron osteotomy and (2) the revisional procedure of the two level osteotomies: (i) proximal opening-wedge basal osteotomy and (ii) distal short Scarf with medial closing wedges. If a Chevron osteotomy is used inappropriately, for example, in an attempt to correct too large a deformity, it may angulate laterally causing a malunion with an increased distal metatarsal articular angle. Secondly, it is feasible to correct this combined deformity using a combination of proximal opening-wedge and distal short Scarf osteotomies

    Transforming, storing and consuming athletic experiences; a coach’s narrative of using a video app.

    The analysis of sport performance in performance contexts has become synonymous with the use of a range of software applications and hardware e.g. heart rate monitors and gps systems. With the prevalence of technology in mind, a small but growing corpus of literature has begun to consider this phenomenon and its influence upon the coaching process. This study adds to this literature by detailing the autoethnographic experiences of a case study coach; Derek. A contextualised and richly described narrative account of Derek’s experience of using coaching applications is provided. Analysis of Derek’s narrative suggests that technology can be a useful means by which individuals make sense of their experience. Specifically, 1) technology can be a ‘ready-to-hand’ instrument that enhances the coaching process. Unfortunately, 2) technology may become the only and ‘calculative’ means by which individuals come to understand their performance. In such instances, it is important to note that 3) the videos we use to understand our performance are transformed and incomplete representations of lived athletic experiences. Thus, Derek’s story illustrates how technology can be both an enabler and barrier to athletes who wish to holistically understand their own lived experiences and engage in coach-athlete relationships. The accompanying analysis draws upon concepts from Heideggerian philosophy to add insight into the use of technology within the coaching process. In so doing, the study prompts coaches to critically view their coach-athlete relationships as situated in a wider world which contains, and can be mediated by, technology. In addition, Derek’s story (re)directs researchers and coaching practitioners interested in technology to a useful literature (philosophy of technology) which may further inform their understanding of coach-athlete relationships
