    Benthic algae and seagrasses of the Walpole and Nornalup Inlets Marine Park, Western Australia

    A survey of the marine plants of the Walpole and Nornalup Inlets Marine Park has recorded 49 species of marine benthic algae and seagrasses, including 15 green algae, 11 brown algae, 18 red algae, 4 seagrasses, and one cyanobacterium, representing a substantial increase on the 14 previously recorded species. Most species are relatively common elements of the south-western Australian marine flora, but several are of taxonomic or biogeographic and ecological interest. Included in this group are: a new species of the green algal genus Codium, the first records of previously unknown reproductive phases in the red algae Mazoyerella australis and Spermothamnion cymosum, and a new distribution record for Ossiella pacifica, a species hitherto known only from warmer waters of the Pacific Ocean and not recorded for mainland Australia. The species diversity in the inlets decreases markedly with increasing distance from the ocean, reflecting a reducing marine and increasing estuarine influence

    Enhancement of feline immunodeficiency virus infection after immunization with envelope glycoprotein subunit vaccines.

    Cats were immunized three times with different recombinant feline immunodeficiency virus (FIV) candidate vaccines. Recombinant vaccinia virus (rVV)-expressed envelope glycoprotein with (vGR657) or without (vGR657 x 15) the cleavage site and an FIV envelope bacterial fusion protein (beta-Galactosidase-Env) were incorporated into immune-stimulating complexes or adjuvanted with Quil A. Although all immunized cats developed antibodies against the envelope protein, only the cats vaccinated with the rVV-expressed envelope glycoproteins developed antibodies which neutralized FIV infection of Crandell feline kidney cells. These antibodies failed to neutralize infection of thymocytes with a molecularly cloned homologous FIV. After the third immunization the cats were challenged with homologous FIV. Two weeks after challenge the cell-associated viral load proved to be significantly higher in the cats immunized with vGR657 and vGR657 x 15 than in the other cats. The cats immunized with vGR657 and vGR657 x 15 also developed antibodies against the Gag proteins more rapidly than the cats immunized with beta-Galactosidase-Env or the control cats. This suggested that immunization with rVV-expressed glycoprotein of FIV results in enhanced infectivity of FIV. It was shown that the observed enhancement could be transferred to naive cats with plasma collected at the day of challenge

    Reflectance confocal microscopy as a non-invasive imaging tool in vulvar high-grade squamous intraepithelial lesions and lichen sclerosus:A descriptive morphological study in patients and healthy volunteers

    Incorrect and delayed diagnosis of vulvar high-grade squamous intraepithelial neoplasia (vHSIL) and lichen sclerosus (LS) increases malignant progression risks and negatively impacts prognosis and quality of life. There is a need to improve diagnosis and monitoring. Reflectance confocal microscopy is a non-invasive imaging tool that visualizes skin structures at cellular resolution. The objectives were to explore feasibility and patient acceptability of vulvar RCM imaging and to identify RCM characteristics that are discriminative for vulvar HSIL and LS. This was a prospective, cross-sectional, observational clinical trial in patients with vHSIL and LS compared to healthy volunteers. RCM images and vulvar tissue samples were obtained. Five (5) patients with vHSIL, 10 patients with LS and 10 healthy volunteers were enrolled. In total, 100 image series of vulvar skin were obtained, including lesional and nonlesional sites. The RCM technique was considered acceptable for application by patients and healthy controls. Healthy vulvar skin was characterized by a homogenous, normal honeycomb patterned epidermis and a clear epidermal-dermal junctions. Vulvar HSIL and LS displayed an atypical honeycomb pattern of the epidermis and lymphocytic influx with presence of melanophages. Distinct features specifically observed in LS included the presence of hyalinised vessels and sclerotic areas in the dermis. RCM is a non-invasive imaging technique that is feasible and clinically acceptable to apply on vulvar skin, both in patients with premalignant lesions and healthy controls. Recognition and validation of disease-specific characteristics could make reflectance confocal microscopy a clinical tool to non-invasively aid identification of vulvar premalignancies.</p

    Making the invisible stratigraphy visible: a grid-based, multi-proxy geoarchaeological study of Umhlatuzana rockshelter, South Africa

    Umhlatuzana rockshelter is an archaeological site with an occupational record covering the Middle Stone Age, Later Stone Age, and Iron Age. The presence of both Middle and Later Stone Age assemblages makes Umhlatuzana the ideal location for the study of the MSA-LSA transition (20-40 ka) in southern Africa. This transitional period is characterized by important modifications in stone tool technology, from prepared core technology to a toolkit based on microlith production. These changes are argued to have occurred in response to changes in climate and environment leading up to the Last Glacial Maximum. The deposits bearing the transitional assemblages at Umhlatuzana rockshelter appear homogeneous with no visible stratigraphic boundaries. This study integrates geoarchaeological techniques in order to explore fine-resolution geochemical differentiations of the sediments that are macroscopically invisible, and that will provide insight into (post-)depositional processes over time. Samples were systematically retrieved from the western profile of the site following a grid-based sampling strategy and analyzed for pH, elemental composition (XRF), and Magnetic Susceptibility. Additionally, the results were integrated with preliminary micromorphological observations. Our study reveals a steady, gradual change in the geochemistry of the deposits throughout the Pleistocene, related to a combination of environmental change and occupation intensity. We suggest that the part of the sequence reported to bear Middle to Later Stone Age transitional industries is characterized by wetter environmental conditions compared to the underlying deposits. Additionally, we support results from previous studies that excluded large scale post-depositional movement of the sedimentary sequence. Our study offers a successful multi-proxy approach to systematically sample and study archaeological deposits at the macro and micro scale, integrating a variety of geoarchaeological techniques. The approach provides insight into the depositional and post-depositional history of the site, and allows for questions of stratigraphic integrity, anthropogenic input, preservation, and environmental change to be addressed.Thrombosis and Hemostasi

    Umhlatuzana rockshelter is an archaeological site with an occupational record spanning the last ~70,000 years, covering the Middle Stone Age (MSA), Later Stone Age (LSA), and Iron Age. The deposits bearing Pleistocene archaeological assemblages at Umhlatuzana rockshelter appear homogeneous with no macroscopically visible stratigraphic boundaries. This means the integrity of the archaeological assemblages is difficult to ascertain. Moreover, the sedimentation rate, taphonomic history, and the environmental context across the sequence are unclear. This study aims to resolve these issues by integrating different geoarchaeological techniques in order to explore fine-resolution geochemical differentiations of the sediments that are macroscopically invisible. Samples were systematically retrieved from the western profile of the site following a grid-based sampling strategy and analysed for pH, elemental composition (XRF), and Magnetic Susceptibility. These methods were chosen because they provide insight into ‘invisible’ geoarchaeological dynamics, related to sediment input (geogenic and anthropogenic), taphonomy, and environmental conditions. Additionally, the results were integrated with preliminary micromorphological observations. Our study reveals a gradual change in the geochemistry of the deposits throughout the Pleistocene, related to a combination of environmental change and occupation intensity. Furthermore, the gradual change within the geochemical data indicates that no large-scale sediment mixing took place (contrary to previous suggestions), while small-scale mixing related to bioturbation is visible in the micromorphological thin sections. Our study offers a successful multi-proxy approach to systematically sample and analyse archaeological deposits at the macro and micro scale, integrating a variety of geoarchaeological techniques.     High rate of autonomic neuropathy in Cornelia de Lange Syndrome

    Background: Cornelia de Lange Syndrome (CdLS) is a rare congenital disorder characterized by typical facial features, growth failure, limb abnormalities, and gastroesophageal dysfunction that may be caused by mutations in several genes that disrupt gene regulation early in development. Symptoms in individuals with CdLS suggest that the peripheral nervous system (PNS) is involved, yet there is little direct evidence. Method: Somatic nervous system was evaluated by conventional motor and sensory nerve conduction studies and autonomic nervous system by heart rate variability, sympathetic skin response and sudomotor testing. CdLS Clinical Score and genetic studies were also obtained. Results: Sympathetic skin response and sudomotor test were pathological in 35% and 34% of the individuals with CdLS, respectively. Nevertheless, normal values in large fiber nerve function studies. Conclusions: Autonomic nervous system (ANS) dysfunction is found in many individuals with Cornelia de Lange Syndrome, and could be related to premature aging

    Surface layering of liquids: The role of surface tension

    Recent measurements show that the free surfaces of liquid metals and alloys are always layered, regardless of composition and surface tension; a result supported by three decades of simulations and theory. Recent theoretical work claims, however, that at low enough temperatures the free surfaces of all liquids should become layered, unless preempted by bulk freezing. Using x-ray reflectivity and diffuse scattering measurements we show that there is no observable surface-induced layering in water at T=298 K, thus highlighting a fundamental difference between dielectric and metallic liquids.     Dermatoscopy and Optical Coherence Tomography in Vulvar High-Grade Squamous Intraepithelial Lesions and Lichen Sclerosus:A Prospective Observational Trial

    Objective This study aimed to examine potential discriminatory characteristics of dermatoscopy and dynamic optical coherence tomography (D-OCT) on vulvar high-grade squamous intraepithelial lesions (vHSIL) and lichen sclerosus (LS) compared with healthy vulvar skin. Methods A prospective observational clinical trial was performed in 10 healthy volunteers, 5 vHSIL and 10 LS patients. Noninvasive imaging measurements using dermatoscopy and D-OCT were obtained at several time points, including lesional and nonlesional vulvar skin. Morphologic features of vHSIL and LS were compared with healthy controls. Epidermal thickness and blood flow were determined using D-OCT. Patients reported tolerability of each study procedure, including reference vulvar biopsies. The main outcome measures were feasibility and tolerability of imaging modalities, dermatoscopy and OCT characteristics, OCT epidermal thickness and D-OCT dermal blood flow. Results The application of dermatoscopy and D-OCT is feasible and tolerable. In vHSIL, dermatoscopic warty structures were present. In LS, sclerotic areas and arborizing vessels were observed. Structural OCT in the vulvar area aligned with histology for hyperkeratosis and dermal-epidermal junction visualization. Currently, the OCT algorithm is unable to calculate the epidermal thickness of the uneven vulvar area. Dynamic optical coherence tomography showed statistically significant increased blood flow in LS patients (mean ± SD, 0.053 ± 0.029) to healthy controls (0.040 ± 0.012; p =.0024). Conclusions The application of dermatoscopy and D-OCT is feasible and tolerable in vHSIL and LS patients. Using dermatoscopy and D-OCT, the authors describe potential characteristics to aid differentiation of diseased from healthy vulvar skin, which could complement clinical assessments.</p

    Developing an interactive Tool for evaluating sand nourishment strategies along the Holland coast in perspective of benthos, fish nursery and dune quality

    Sand nourishments can affect the coastal ecosystem in various ways. Direct effects are the burial of benthic species under a layer of sand. In the direct vicinity, suffocation of benthos can occur due to the settling of a plume of suspended sediment particles. A plume of fine particles may also increase turbidity and thereby affect primary production and the foraging success of filter-feeding benthos and fish. Indirect effects are habitat change, such as altered morphology and sedimentology. It was tested by investigating whether it is possible to optimise nourishment configuration, location and timing, to: - minimise the impact on benthos, - increase nursery area and/or quality, and - enhance dune quality
