    Electrical stimulation alleviates depressive-like behaviors of rats: investigation of brain targets and potential mechanisms

    Deep brain stimulation (DBS) is a promising therapy for patients with refractory depression. However, key questions remain with regard to which brain target(s) should be used for stimulation, and which mechanisms underlie the therapeutic effects. Here, we investigated the effect of DBS, with low- and high-frequency stimulation (LFS, HFS), in different brain regions (ventromedial prefrontal cortex, vmPFC; cingulate cortex, Cg; nucleus accumbens (NAc) core or shell; lateral habenula, LHb; and ventral tegmental area) on a variety of depressive-like behaviors using rat models. In the naive animal study, we found that HFS of the Cg, vmPFC, NAc core and LHb reduced anxiety levels and increased motivation for food. In the chronic unpredictable stress model, there was a robust depressive-like behavioral phenotype. Moreover, vmPFC HFS, in a comparison of all stimulated targets, produced the most profound antidepressant effects with enhanced hedonia, reduced anxiety and decreased forced-swim immobility. In the following set of electrophysiological and histochemical experiments designed to unravel some of the underlying mechanisms, we found that vmPFC HFS evoked a specific modulation of the serotonergic neurons in the dorsal raphe nucleus (DRN), which have long been linked to mood. Finally, using a neuronal mapping approach by means of c-Fos expression, we found that vmPFC HFS modulated a brain circuit linked to the DRN and known to be involved in affect. In conclusion, HFS of the vmPFC produced the most potent antidepressant effects in naive rats and rats subjected to stress by mechanisms also including the DRN.postprin

    Ion source developments for stable and radioactive ion beams at GANIL

    Since now many years, the Ganil ion source team has in charge to develop ion sources with three main purposes. The first one concerns the radioactive ion production that implies high efficiency ion sources as the amount of created exotic atoms is very low (between 10 to 108 particle per second). The second one deals with high intensities of stable metallic ion beams for the injectors of the accelerator while the last one tries to increase the intensities of very high charge state ion beams for atomic physic. Concerning radioactive ion production, the recent results obtained, in collaboration with the ISN Grenoble group, with the 1+/n+ method drove us to develop a new concept of ecr ion source for monocharged ion production. The results of the first tests of this source will be given. This new idea for the construction of ecr ion source can be applied to multicharged ion production. Concerning the high charge state ion beam production, a new source called SUPERSHYPIE has been built that allow to increase by a factor 2 the length of the plasma of an ECR4M source. This new concept has just been started and has produced arround 50 nAe of Ar17+ . The first results of this new source will be presented.     Transcriptomic alterations in the sweet orange vasculature correlate with growth repression induced by a variant of citrus tristeza virus

    Citrus tristeza virus (CTV, family Closteroviridae) is an economically important pathogen of citrus. CTV resides in the phloem of the infected plants and induces a range of disease phenotypes, including stem pitting and quick decline as well as a number of other deleterious syndromes. To uncover the biological processes underlying the poorly understood damaging symptoms of CTV, we profiled the transcriptome of sweet orange (Citrus sinensis) phloem-rich bark tissues of non-infected, mock-inoculated trees and trees singly infected with two distinct variants of CTV, T36 or T68-1. The T36 and T68-1 variants accumulated in the infected plants at similar titers. With that, young trees infected with T68-1 were markedly repressed in growth, while the growth rate of the trees infected with T36 was comparable to the mock-inoculated trees. Only a small number of differentially expressed genes (DEGs) were identified in the nearly asymptomatic T36-infected trees, whereas almost fourfold the number of DEGs were identified with the growth-restricting T68-1 infection. DEGs were validated using quantitative reverse transcription-PCR. While T36 did not induce many noteworthy changes, T68-1 altered the expression of numerous host mRNAs encoding proteins within significant biological pathways, including immunity and stress response proteins, papain-like cysteine proteases (PLCPs), cell-wall modifying enzymes, vascular development proteins and others. The transcriptomic alterations in the T68-1-infected trees, in particular, the strong and persistent increase in the expression levels of PLCPs, appear to contribute to the observed stem growth repression. On the other hand, analysis of the viral small interfering RNAs revealed that the host RNA silencing-based response to the infection by T36 and that by T68-1 was comparable, and thus, the induction of this antiviral mechanism may not contribute to the difference in the observed symptoms. The DEGs identified in this study promote our understanding of the underlying mechanisms of the yet unexplained growth repression induced by severe CTV isolates in sweet orange trees

    Coherent state quantization of a particle in de Sitter space

    We present a coherent state quantization of the dynamics of a relativistic test particle on a one-sheet hyperboloid embedded in a three-dimensional Minkowski space. The group SO_0(1,2) is considered to be the symmetry group of the system. Our procedure relies on the choice of coherent states of the motion on a circle.     Negative emotional stimuli reduce contextual cueing but not response times in inefficient search

    In visual search, previous work has shown that negative stimuli narrow the focus of attention and speed reaction times (RTs). This paper investigates these two effects by first asking whether negative emotional stimuli narrow the focus of attention to reduce the learning of a display context in a contextual cueing task and, second, whether exposure to negative stimuli also reduces RTs in inefficient search tasks. In Experiment 1, participants viewed either negative or neutral images (faces or scenes) prior to a contextual cueing task. In a typical contextual cueing experiment, RTs are reduced if displays are repeated across the experiment compared with novel displays that are not repeated. The results showed that a smaller contextual cueing effect was obtained after participants viewed negative stimuli than when they viewed neutral stimuli. However, in contrast to previous work, overall search RTs were not faster after viewing negative stimuli (Experiments 2 to 4). The findings are discussed in terms of the impact of emotional content on visual processing and the ability to use scene context to help facilitate search

    Ion sources at GANIL

    International audienceThe GANIL produces since many years heavy ion beams with Electron Cyclotron Resonance ion sources. Different facilities have been constructed during the last years in order to allow experiments in a large range of energy (from some tens of kV to 100 MeV/nucleon). The list of available ions has been greatly extended with the construction of the SPIRAL1 facility that produces and accelerates radioactives ions . An overview of the different developments made at GANIL for stable and radioactive ion beam production including the sources for the SPIRAL2 project is given in this paper
