    Adaptation kinetics in bacterial chemotaxis

    Cells of Escherichia coli, tethered to glass by a single flagellum, were subjected to constant flow of a medium containing the attractant alpha-methyl-DL-aspartate. The concentration of this chemical was varied with a programmable mixing apparatus over a range spanning the dissociation constant of the chemoreceptor at rates comparable to those experienced by cells swimming in spatial gradients. When an exponentially increasing ramp was turned on (a ramp that increases the chemoreceptor occupancy linearly), the rotational bias of the cells (the fraction of time spent spinning counterclockwise) changed rapidly to a higher stable level, which persisted for the duration of the ramp. The change in bias increased with ramp rate, i.e., with the time rate of change of chemoreceptor occupancy. This behavior can be accounted for by a model for adaptation involving proportional control, in which the flagellar motors respond to an error signal proportional to the difference between the current occupancy and the occupancy averaged over the recent past. Distributions of clockwise and counterclockwise rotation intervals were found to be exponential. This result cannot be explained by a response regular model in which transitions between rotational states are generated by threshold crossings of a regular subject to statistical fluctuation; this mechanism generates distributions with far too many long events. However, the data can be fit by a model in which transitions between rotational states are governed by first-order rate constants. The error signal acts as a bias regulator, controlling the values of these constants

    The main objective of wastewater treatment is to purify the water by degradation of organic matter in the water to anenvironmentally friendly status. To achieve this objective, some effluent (waste water) quality parameters such asChemical oxygen demand (COD) and Biochemical oxygen demand (BOD5) should be measured continuously in orderto meet up with the said objective and regulatory demands. However, through the prediction on water qualityparameters, effective guidance can be provided to comply with such demand without necessarily engaging in rigorouslaboratory analysis. Box-Jenkin’s Auto Regressive Integrated Moving Average (ARIMA) technique is one of the mostrefined extrapolation techniques for prediction while Artificial Neural Network (ANN) is a modern non-linear methodalso used for prediction. The Mean Absolute Error (MAE), Mean Absolute Percentage Error (MAPE), Root MeanSquare Error (RMSE) and Correlation coefficient (r) are used to evaluate the accuracy of the above-mentionedmodels. This paper examined the efficiency of ARIMA and ANN models in prediction of two major water qualityparameters (COD and BOD5) in a wastewater treatment plant. With the aid of R software, it was concluded that in allthe error estimates, ANNs models performed better than the ARIMA model, hence it can be used in the operation ofthe treatment system

    Coordination of flagella on filamentous cells of Escherichia coli

    Video techniques were used to study the coordination of different flagella on single filamentous cells of Escherichia coli. Filamentous, nonseptate cells were produced by introducing a cell division mutation into a strain that was polyhook but otherwise wild type for chemotaxis. Markers for its flagellar motors (ordinary polyhook cells that had been fixed with glutaraldehyde) were attached with antihook antibodies. The markers were driven alternately clockwise and counterclockwise, at angular velocities comparable to those observed when wild-type cells are tethered to glass. The directions of rotation of different markers on the same cell were not correlated; reversals of the flagellar motors occurred asynchronously. The bias of the motors (the fraction of time spent spinning counterclockwise) changed with time. Variations in bias were correlated, provided that the motors were within a few micrometers of one another. Thus, although the directions of rotation of flagellar motors are not controlled by a common intracellular signal, their biases are. This signal appears to have a limited range

    Composite Flexible Blanket Insulation

    Composite flexible multilayer insulation systems (MLI) were evaluated for thermal performance and compared with the currently used fibrous silica (baseline) insulation system. The systems described are multilayer insulations consisting of alternating layers of metal foil and scrim ceramic cloth or vacuum metallized polymeric films quilted together using ceramic thread. A silicon carbide thread for use in the quilting and the method of making it are also described. These systems are useful in providing lightweight insulation for a variety of uses, particularly on the surface of aerospace vehicles subject to very high temperatures during flight

    Compositional modulation in AlxGa1−xAs epilayers grown by molecular beam epitaxy on the (111) facets of grooves in a nonplanar substrate

    We report the first observation of a lateral junction formed in an alloy due to an abrupt transition from segregated to random AlGaAs alloy compositions. Al0.25Ga0.75As epilayers were grown by molecular beam epitaxy on [011-bar] oriented grooves in a nonplanar (100) GaAs substrate. A quasi-periodic modulation of the aluminum concentration occurs spontaneously in material grown on the (111) facets of the groove, with a period of 50–70 Å along the [111] direction. The compositional modulation is associated with a reduction of the band gap by 130 meV, with respect to the random alloy. While segregation of the AlGaAs alloy has been seen previously, this is the first observation of segregation of AlGaAs grown on a (111) surface. The compositional modulation terminates abruptly at the boundaries of the (111) facet, forming abrupt lateral junctions in the AlGaAs layers grown on a groove

    It was determined that an onboard vortex wake detection system using existing, proven instrumentation is technically feasible. This system might be incorporated into existing onboard systems such as a wind shear detection system, and might provide the pilot with the location of a vortex wake, as well as an evasive maneuver so that the landing separations may be reduced. It is suggested that this system might be introduced into our nation's commuter aircraft fleet and major air carrier fleet and permit a reduction of current landing separation standards, thereby reducing takeoff and departure delays
