Therapies directed at augmenting regulatory T cell (Treg) activities in vivo as a systemic treatment for autoimmune disorders and transplantation may be associated with significant off-target effects, including a generalized immunosuppression that may compromise beneficial immune responses to infections and cancer cells. Adoptive cellular therapies using purified expanded Tregs represents an attractive alternative to systemic treatments, with results from animal studies noting increased therapeutic potency of antigen-specific Tregs over polyclonal populations. However, current methodologies are limited in terms of the capacity to isolate and expand a sufficient quantity of endogenous antigen-specific Tregs for therapeutic intervention. Moreover, FOXP3+ Tregs fall largely within the CD4+ T cell subset and are thus routinely MHC class II-specific, whereas class I-specific Tregs may function optimally in vivo by facilitating direct tissue recognition.To overcome these limitations, we have developed a novel means for generating large numbers of antigen-specific Tregs involving lentiviral T cell receptor (TCR) gene transfer into in vitro expanded polyclonal natural Treg populations. Tregs redirected with a high-avidity class I-specific TCR were capable of recognizing the melanoma antigen tyrosinase in the context of HLA-A*0201 and could be further enriched during the expansion process by antigen-specific reactivation with peptide loaded artificial antigen presenting cells. These in vitro expanded Tregs continued to express FOXP3 and functional TCRs, and maintained the capacity to suppress conventional T cell responses directed against tyrosinase, as well as bystander T cell responses. Using this methodology in a model tumor system, murine Tregs designed to express the tyrosinase TCR effectively blocked antigen-specific effector T cell (Teff) activity as determined by tumor cell growth and luciferase reporter-based imaging.These results support the feasibility of class I-restricted TCR transfer as a promising strategy to redirect the functional properties of Tregs and provide for a more efficacious adoptive cell therapy

The production of tt‾ , W+bb‾ and W+cc‾ is studied in the forward region of proton–proton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98±0.02 fb−1 . The W bosons are reconstructed in the decays W→ℓν , where ℓ denotes muon or electron, while the b and c quarks are reconstructed as jets. All measured cross-sections are in agreement with next-to-leading-order Standard Model predictions.The production of tt, W+bb and W+cc is studied in the forward region of proton-proton collisions collected at a centre-of-mass energy of 8 TeV by the LHCb experiment, corresponding to an integrated luminosity of 1.98 ± 0.02 \mbox{fb}^{-1}. The W bosons are reconstructed in the decays W→ℓν, where ℓ denotes muon or electron, while the b and c quarks are reconstructed as jets. All measured cross-sections are in agreement with next-to-leading-order Standard Model predictions

A first study of CP violation in the decay modes B±→[KS0K±π∓]Dh± and B±→[KS0K∓π±]Dh±, where h labels a K or π meson and D labels a D0 or D0 meson, is performed. The analysis uses the LHCb data set collected in pp collisions, corresponding to an integrated luminosity of 3 fb−1. The analysis is sensitive to the CP-violating CKM phase γ through seven observables: one charge asymmetry in each of the four modes and three ratios of the charge-integrated yields. The results are consistent with measurements of γ using other decay modes

A search for the rare decays Bs0→π+π−μ+μ− and B0→π+π−μ+μ− is performed in a data set corresponding to an integrated luminosity of 3.0 fb−1 collected by the LHCb detector in proton-proton collisions at centre-of-mass energies of 7 and 8 TeV. Decay candidates with pion pairs that have invariant mass in the range 0.5-1.3 GeV/c2 and with muon pairs that do not originate from a resonance are considered. Proton–proton collision data recorded in 2011 and 2012 by the LHCb experiment, corresponding to an integrated luminosity of 3.0 fb−1 , are analysed to search for the charmless B0→ρ0ρ0 decay. More than 600 B0→(π+π−)(π+π−) signal decays are selected and used to perform an amplitude analysis, under the assumption of no CP violation in the decay, from which the B0→ρ0ρ0 decay is observed for the first time with 7.1 standard deviations significance. The fraction of B0→ρ0ρ0 decays yielding a longitudinally polarised final state is measured to be fL=0.745−0.058+0.048(stat)±0.034(syst) . The B0→ρ0ρ0 branching fraction, using the B0→ϕK⁎(892)0 decay as reference, is also reported as B(B0→ρ0ρ0)=(0.94±0.17(stat)±0.09(syst)±0.06(BF))×10−6

The production of the ηc(1S) state in proton-proton collisions is probed via its decay to the ppˉ final state with the LHCb detector, in the rapidity range 2.06.5 GeV/c. The cross-section for prompt production of ηc(1S) mesons relative to the prompt J/ψ cross-section is measured, for the first time, to be σηc(1S)/σJ/ψ=1.74±0.29±0.28±0.18B at a centre-of-mass energy s=7 TeV using data corresponding to an integrated luminosity of 0.7 fb−1, and σηc(1S)/σJ/ψ=1.60±0.29±0.25±0.17B at s=8 TeV using 2.0 fb−1. The uncertainties quoted are, in order, statistical, systematic, and that on the ratio of branching fractions of the ηc(1S) and J/ψ decays to the ppˉ final state. In addition, the inclusive branching fraction of b-hadron decays into ηc(1S) mesons is measured, for the first time, to be B(b→ηcX)=(4.88±0.64±0.25±0.67B)×10−3, where the third uncertainty includes also the uncertainty on the J/ψ inclusive branching fraction from b-hadron decays. The cross-section for prompt production of ηc(1S) mesons relative to the prompt J/ψ cross-section is measured, for the first time, to be σηc(1S)/σJ/ψ=1.74±0.29±0.28±0.18B at a centre-of-mass energy s=7 TeV using data corresponding to an integrated luminosity of 0.7 fb−1 , and σηc(1S)/σJ/ψ=1.60±0.29±0.25±0.17B at s=8 TeV using 2.0 fb−1 . The uncertainties quoted are, in order, statistical, systematic, and that on the ratio of branching fractions of the ηc(1S) and J/ψ decays to the pp final state. In addition, the inclusive branching fraction of b -hadron decays into ηc(1S) mesons is measured, for the first time, to be B(b→ηcX)=(4.88±0.64±0.29±0.67B)×10−3 , where the third uncertainty includes also the uncertainty on the J/ψ inclusive branching fraction from b -hadron decays. The difference between the J/ψ and ηc(1S) meson masses is determined to be 114.7±1.5±0.1MeV/c2 .The production of the ηc(1S) state in proton-proton collisions is probed via its decay to the ppˉ final state with the LHCb detector, in the rapidity range 2.06.5 GeV/c. A search for the lepton flavour violating decay τ−→μ−μ+μ− is performed with the LHCb experiment. The data sample corresponds to an integrated luminosity of 1.0 fb−1 of proton-proton collisions at a centre-of-mass energy of 7 TeV and 2.0 fb−1 at 8 TeV. The associated production of a Z boson or an off-shell photon γ∗ with a bottom quark in the forward region is studied using proton-proton collisions at a centre-of-mass energy of 7TeV. The Z bosons are reconstructed in the Z/γ∗→μ+μ− final state from muons with a transverse momentum larger than 20GeV, while two transverse momentum thresholds are considered for jets (10GeV and 20GeV). Both muons and jets are reconstructed in the pseudorapidity range 2.010GeV, and \sigma(\text{\text{Z}/\gamma^*(\mu^{+}\mu^{-})+b-jet}) = 167 \pm 47 (\text{stat}) \pm 29 (\text{syst}) \pm 6 (\text{lumi}) {\,{fb}} for {p_{\rm T}}(jet)>20GeV
