    The Inclusive City of Johannesburg and the Challenge of Affordable Housing

    Johannesburg, as South Africa’s largest city and the most important economic center, still suffers under the patterns of inequality of the past. Although the spatial change has been rapid since 1994 after the post-apartheid era, the spatial divisions along class lines still remain. Strategic spatial planning strategies of the last two and a half decades were targeting the reduction of spatial patterns of inequality and exclusion by fostering compact and integrated spatial development on national, regional and urban level and currently the local provision of inclusionary housing. This study, based on literature review and census data evaluation, investigates the current challenges of Johannesburg, such as growing population, existing inequalities and the affordable housing backlog. Moreover, the current affordable housing programmes and new policy requirements in Johannesburg are described and analysed. Despite the release of numerous city-wide policies, such as the Spatial Development Framework 2040 (2016) or the Inclusionary Housing Policy (2019) the research shows that the situation has not significantly improved yet, which is indicated by a lack of restrictions in the requirements, implementation deficiencies and limited policy uptake in planning practice

    Inclusivity insights:two urban development projects in Johannesburg

    Johannesburg, which is South Africa’s largest city and economic center, is still influenced by patterns of inequality of the past. Although spatial transformation has been rapid since 1994, spatial divisions along class lines are still prevalent long after the apartheid era. This is despite the fact that societal values such as inclusivity, spatial justice and equal access to resources have become the core goals of Johannesburg’s spatial and urban development. This is particularly true when addressing housing, public open spaces, transport and social infrastructure embedded in a suitable land-use mix. However, despite the adoption of numerous policies by government, this research indicates that even recent urban development projects such as Fleurhof and Waterfall are falling short in delivering those objectives. Based on a case study analysis, we show that significant disfunctionalities become evident when evaluating these two projects, despite their meeting relevant indicators of spatial inclusion on paper. Tensions are identified between theoretical approaches and the implementation of societally relevant policy goals. These include inclusivity and spatial justice by mostly privatized provision of housing and services, and deficiencies in public maintenance of infrastructure. The research reveals, that ‘ticking-the-boxes’ behavior on policy and project level does not produce equitable, inclusive neighborhoods and urban spatial patterns, but rather reproduces spatial inequalities of the past. If these policies are to result in real spatial change and improvement in the lives of Johannesburg residents, a more proactive approach by the public sector at different levels will be necessary

    Predictors of increasing BMI during the course of diabetes in children and adolescents with type 1 diabetes: data from the German/Austrian DPV multicentre survey

    Objective: Increased weight gain has been reported prior to disease onset (accelerator hypothesis) and as a side effect of intensified insulin therapy in type 1 diabetes (T1D). Paediatric studies are complicated by the age-dependency and gender-dependency of BMI, and also by a trend towards obesity in the general population. The aim of this study was to evaluate factors related to the increase in BMI during the course of diabetes in children and adolescents with T1D in a large multicentre survey. Design: Within the DPV database (Diabetespatienten Verlaufsdokumentation) a standardised, prospective, computer-based documentation programme, data of 53 108 patients with T1D, aged <20 years, were recorded in 248 centres. 12 774 patients (53% male, mean age 13.4+/-3.9, mean diabetes duration 4.7+/-3.0 years and mean age at diabetes onset 8.7+/-4.0 years) were included in this analysis. Population-based German reference data were used to calculate BMI-SDS and define overweight and obesity. Results: 12.5% of T1D patients were overweight and 2.8% were obese. Multiple longitudinal regression analysis revealed that female gender, low BMI at diabetes onset, intensified insulin therapy and higher insulin dose, as well as pubertal diabetes onset, long diabetes duration and onset in earlier calendar years among girls, were related to higher BMI-SDS increase during the course of diabetes (p<0.01; all). Conclusions: Intensified insulin regimen is associated with weight gain during T1D treatment, in addition to demographic variables. Optimisation of diabetes management, especially in females, might limit weight gain in order to reduce overweight and obesity together with comorbidities among paediatric T1D patients

    Genome-wide meta-analysis associates HLA-DQA1/DRB1 and LPA and lifestyle factors with human longevity

    Genomic analysis of longevity offers the potential to illuminate the biology of human aging. Here, using genome-wide association meta-analysis of 606,059 parents’ survival, we discover two regions associated with longevity (HLA- DQA1/DRB1 and LPA). We also validate previous suggestions that APOE, CHRNA3/5, CDKN2A/B, SH2B3 and FOXO3A influence longevity. Next we show that giving up smoking, educational attainment, openness to new experience and high-density lipoprotein (HDL) cholesterol levels are most positively genetically correlated with lifespan while susceptibility to coronary artery disease (CAD), cigarettes smoked per day, lung cancer, insulin resistance and body fat are most negatively correlated. We suggest that the effect of education on lifespan is principally mediated through smoking while the effect of obesity appears to act via CAD. Using instrumental variables, we suggest that an increase of one body mass index unit reduces lifespan by 7 months while 1 year of education adds 11 months to expected lifespan

    Novel genetic loci associated with hippocampal volume

    The hippocampal formation is a brain structure integrally involved in episodic memory, spatial navigation, cognition and stress responsiveness. Structural abnormalities in hippocampal volume and shape are found in several common neuropsychiatric disorders. To identify the genetic underpinnings of hippocampal structure here we perform a genome-wide association study (GWAS) of 33,536 individuals and discover six independent loci significantly associated with hippocampal volume, four of them novel. Of the novel loci, three lie within genes (ASTN2, DPP4 and MAST4) and one is found 200 kb upstream of SHH. A hippocampal subfield analysis shows that a locus within the MSRB3 gene shows evidence of a localized effect along the dentate gyrus, subiculum, CA1 and fissure. Further, we show that genetic variants associated with decreased hippocampal volume are also associated with increased risk for Alzheimer’s disease (rg ÂŒ 0.155). Our findings suggest novel biological pathways through which human genetic variation influences hippocampal volume and risk for neuropsychiatric illness

    Identification of microRNA-mRNA functional interactions in UVB-induced senescence of human diploid fibroblasts

    BACKGROUND: Cellular senescence can be induced by a variety of extrinsic stimuli, and sustained exposure to sunlight is a key factor in photoaging of the skin. Accordingly, irradiation of skin fibroblasts by UVB light triggers cellular senescence, which is thought to contribute to extrinsic skin aging, although molecular mechanisms are incompletely understood. Here, we addressed molecular mechanisms underlying UVB induced senescence of human diploid fibroblasts. RESULTS: We observed a parallel activation of the p53/p21(WAF1) and p16(INK4a)/pRb pathways. Using genome-wide transcriptome analysis, we identified a transcriptional signature of UVB-induced senescence that was conserved in three independent strains of human diploid fibroblasts (HDF) from skin. In parallel, a comprehensive screen for microRNAs regulated during UVB-induced senescence was performed which identified five microRNAs that are significantly regulated during the process. Bioinformatic analysis of miRNA-mRNA networks was performed to identify new functional mRNA targets with high confidence for miR-15a, miR-20a, miR-20b, miR-93, and miR-101. Already known targets of these miRNAs were identified in each case, validating the approach. Several new targets were identified for all of these miRNAs, with the potential to provide new insight in the process of UVB-induced senescence at a genome-wide level. Subsequent analysis was focused on miR-101 and its putative target gene Ezh2. We confirmed that Ezh2 is regulated by miR-101 in human fibroblasts, and found that both overexpression of miR-101 and downregulation of Ezh2 independently induce senescence in the absence of UVB irradiation. However, the downregulation of miR-101 was not sufficient to block the phenotype of UVB-induced senescence, suggesting that other UVB-induced processes induce the senescence response in a pathway redundant with upregulation of miR-101. CONCLUSION: We performed a comprehensive screen for UVB-regulated microRNAs in human diploid fibroblasts, and identified a network of miRNA-mRNA interactions mediating UVB-induced senescence. In addition, miR-101 and Ezh2 were identified as key players in UVB-induced senescence of HDF
