    The Effect of Star Formation History on the Inferred Initial Stellar Mass Function

    Peaks and lulls in the star formation rate (SFR) over the history of the Galaxy produce plateaux and declines in the present day mass function (PDMF) where the main-sequence lifetime overlaps the age and duration of the SFR variation. These PDMF features can be misinterpreted as the form of the intrinsic stellar initial mass function (IMF) if the star formation rate is assumed to be constant or slowly varying with time. This effect applies to all regions that have formed stars for longer than the age of the most massive stars, including OB associations, star complexes, and especially galactic field stars. Related problems may apply to embedded clusters. Evidence is summarized for temporal SFR variations from parsec scales to entire galaxies, all of which should contribute to inferred IMF distortions. We give examples of various star formation histories to demonstrate the types of false IMF structures that might be seen. These include short-duration bursts, stochastic histories with log-normal amplitude distributions, and oscillating histories with various periods and phases. The inferred IMF should appear steeper than the intrinsic IMF over mass ranges where the stellar lifetimes correspond to times of decreasing SFRs; shallow portions of the inferred IMF correspond to times of increasing SFRs. If field regions are populated by dispersed clusters and defined by their low current SFRs, then they should have steeper inferred IMFs than the clusters. The SFRs required to give the steep field IMFs in the LMC and SMC are determined.     Observations and Implications of the Star Formation History of the LMC

    We present derivations of star formation histories based on color-magnitude diagrams of three fields in the LMC from HST/WFPC2 observations. A significant component of stars older than 4 Gyr is required to match the observed color-magnitude diagrams. Models with a dispersion-free age-metallicity relation are unable to reproduce the width of the observed main sequence; models with a range of metallicity at a given age provide a much better fit. Such models allow us to construct complete ``population boxes'' for the LMC based entirely on color-magnitude diagrams; remarkably, these qualitatively reproduce the age-metallicity relation observed in LMC clusters. We discuss some of the uncertainties in deriving star formation histories. We find, independently of the models, that the LMC bar field has a larger relative component of older stars than the outer fields. The main implications suggested by this study are: 1) the star formation history of field stars appears to differ from the age distribution of clusters, 2) there is no obvious evidence for bursty star formation, but our ability to measure bursts shorter in duration than \sim 25% of any given age is limited by the statistics of the observed number of stars, 3) there may be some correlation of the star formation rate with the last close passage of the LMC/SMC/Milky Way, but there is no dramatic effect, and 4) the derived star formation history is probably consistent with observed abundances, based on recent chemical evolution models.Comment: Accepted by AJ, 36 pages including 12 figure

    Integration of Pretreatment With Simultaneous Counter-Current Extraction of Energy Sorghum for High-Titer Mixed Sugar Production

    Sorghum (Sorghum bicolor L. Moench) offers substantial potential as a feedstock for the production of sugar-derived biofuels and biochemical products from cell wall polysaccharides (i. e., cellulose and hemicelluloses) and water-extractable sugars (i.e., glucose, fructose, sucrose, and starch). A number of preprocessing schemes can be envisioned that involve processes such as sugar extraction, pretreatment, and densification that could be employed in decentralized, regional-scale biomass processing depots. In this work, an energy sorghum exhibiting a combination of high biomass productivity and high sugar accumulation was evaluated for its potential for integration into several potential biomass preprocessing schemes. This included counter-current extraction of water-soluble sugars followed by mild NaOH or liquid hot water pretreatment of the extracted bagasse. A novel processing scheme was investigated that could integrate with current diffuser-type extraction systems for sugar extraction. In this approach, mild NaOH pretreatment (i.e., <90°C) was performed as a counter-current extraction to yield both an extracted, pretreated bagasse and a high-concentration mixed sugar stream. Following hydrolysis of the bagasse, the combined hydrolysates derived from cellulosic sugars and extractable sugars were demonstrated to be fermentable to high ethanol titers (>8%) at high metabolic yields without detoxification using a Saccharomyces cerevisiae strain metabolically engineered and evolved to ferment xylose

    The Effectiveness of Teleglaucoma versus In-Patient Examination for Glaucoma Screening: A Systematic Review and Meta-Analysis

    BACKGROUND: Glaucoma is the leading cause of irreversible visual impairment in the world affecting 60.5 million people worldwide in 2010, which is expected to increase to approximately 79.6 million by 2020. Therefore, glaucoma screening is important to detect, diagnose, and treat patients at the earlier stages to prevent disease progression and vision loss. Teleglaucoma uses stereoscopic digital imaging to take ocular images, which are transmitted electronically to an ocular specialist. The purpose is to synthesize literature to evaluate teleglaucoma, its diagnostic accuracy, healthcare system benefits, and cost-effectiveness. METHODS: A systematic search was conducted to help locate published and unpublished studies. Studies which evaluate teleglaucoma as a screening device for glaucoma were included. A meta-analysis was conducted to provide estimates of diagnostic accuracy, diagnostic odds ratio, and the relative percentage of glaucoma cases detected. The improvements to healthcare service quality and cost data were assessed. RESULTS: Of 11237 studies reviewed, 45 were included. Our results indicated that, teleglaucoma is more specific and less sensitive than in-person examination. The pooled estimates of sensitivity was 0.832 [95% CI 0.770, 0.881] and specificity was 0.790 [95% CI 0.668, 0.876]. The relative odds of a positive screen test in glaucoma cases are 18.7 times more likely than a negative screen test in a non-glaucoma cases. Additionally, the mean cost for every case of glaucoma detected was 1098.67USandofteleglaucomaperpatientscreenedwas1098.67 US and of teleglaucoma per patient screened was 922.77 US. CONCLUSION: Teleglaucoma can accurately discriminate between screen test results with greater odds for positive cases. It detects more cases of glaucoma than in-person examination. Both patients and the healthcare systems benefit from early detection, reduction in wait and travel times, increased specialist referral rates, and cost savings. Teleglaucoma is an effective screening tool for glaucoma specifically for remote and under-services communities

    Apolipoprotein E O-glycosylation is associated with amyloid plaques and APOE genotype

    Although the APOE ε4 allele is the strongest genetic risk factor for sporadic Alzheimer\u27s disease (AD), the relationship between apolipoprotein (apoE) and AD pathophysiology is not yet fully understood. Relatively little is known about the apoE protein species, including post-translational modifications, that exist in the human periphery and CNS. To better understand these apoE species, we developed a LC-MS/MS assay that simultaneously quantifies both unmodified and O-glycosylated apoE peptides. The study cohort included 47 older individuals (age 75.6 ± 5.7 years [mean ± standard deviation]), including 23 individuals (49%) with cognitive impairment. Paired plasma and cerebrospinal fluid samples underwent analysis. We quantified O-glycosylation of two apoE protein residues - one in the hinge region and one in the C-terminal region - and found that glycosylation occupancy of the hinge region in the plasma was significantly correlated with plasma total apoE levels, APOE genotype and amyloid status as determined by CSF Aβ42/Aβ40. A model with plasma glycosylation occupancy, plasma total apoE concentration, and APOE genotype distinguished amyloid status with an AUROC of 0.89. These results suggest that plasma apoE glycosylation levels could be a marker of brain amyloidosis, and that apoE glycosylation may play a role in the pathophysiology of AD

    Poxvirus-based vaccine therapy for patients with advanced pancreatic cancer

    <p>Abstract</p> <p>Purpose</p> <p>An open-label Phase 1 study of recombinant prime-boost poxviruses targeting CEA and MUC-1 in patients with advanced pancreatic cancer was conducted to determine safety, tolerability and obtain preliminary data on immune response and survival.</p> <p>Patients and methods</p> <p>Ten patients with advanced pancreatic cancer were treated on a Phase I clinical trial. The vaccination regimen consisted of vaccinia virus expressing tumor antigens carcinoembryonic antigen (CEA) and mucin-1 (MUC-1) with three costimulatory molecules B7.1, ICAM-1 and LFA-3 (TRICOM) (PANVAC-V) and fowlpox virus expressing the same antigens and costimulatory molecules (PANVAC-F). Patients were primed with PANVAC-V followed by three booster vaccinations using PANVAC-F. Granulocyte-macrophage colony-stimulating factor (GM-CSF) was used as a local adjuvant after each vaccination and for 3 consecutive days thereafter. Monthly booster vaccinations for up to 12 months were provided for patients without progressive disease. Peripheral blood was collected before, during and after vaccinations for immune analysis.</p> <p>Results</p> <p>The most common treatment-related adverse events were mild injection-site reactions. Antibody responses against vaccinia virus was observed in all 10 patients and antigen-specific T cell responses were observed in 5 out of 8 evaluable patients (62.5%). Median overall survival was 6.3 months and a significant increase in overall survival was noted in patients who generated anti CEA- and/or MUC-1-specific immune responses compared with those who did not (15.1 vs 3.9 months, respectively; <it>P </it>= .002).</p> <p>Conclusion</p> <p>Poxvirus vaccination is safe, well tolerated, and capable of generating antigen-specific immune responses in patients with advanced pancreatic cancer.</p

    NIRCAM image simulations for NGST wavefront sensing

    The Next Generation Space Telescope (NGST) will be a segmented, deployable, infrared-optimized 6.5m space telescope. Its active primary segments will be aligned, co-phased, and then fine-tuned in order to deliver image quality sufficient for the telescope's intended scientific goals. Wavefront sensing used to drive this tuning will come from the analysis of focussed and defocussed images taken with its near-IR science camera, NIRCAM. There is a pressing need to verify that this will be possible with the near-IR detectors that are still under development for NGST. We create simulated NIRCAM images to test the maintenance phase of this plan. Our simulations incorporate Poisson and electronics read noise, and are designed to be able to include various detector and electronics non-linearities. We present our first such simulation, using known or predicted properties of HAWAII HgCdTe focal plane array detectors. Detector effects characterized by the Independent Detector Testing Laboratory will be included as they become available. Simulating InSb detectors can also be done within this framework in future. We generate Point-Spread Functions (PSF's) for a segmented aperture geometry with various wavefront aberrations, and convolve this with typical galaxy backgrounds and stellar foregrounds. We then simulate up-the-ramp (MULTIACCUM in HST parlance) exposures with cosmic ray hits. We pass these images through the HST NICMOS `CALNICA' calibration task to filter out cosmic ray hits. The final images are to be fed to wavefront sensing software, in order to find the ranges of exposure times, filter bandpass, defocus, and calibration star magnitude required to keep the NGST image within its specifications

    Ethanol in alcoholic beverages is a causative agent for colorectal cancer. Colorectal cancer is a biologically heterogeneous disease, and molecular subtypes defined by the presence of somatic mutations in BRAF and KRAS are known to exist. We examined associations between lifetime alcohol intake and molecular and anatomic subtypes of colorectal cancer. We calculated usual alcohol intake for 10-year periods from age 20 using recalled frequency and quantity of beverage-specific consumption for 38,149 participants aged 40-69 years from the Melbourne Collaborative Cohort Study. Cox regression was performed to derive hazard ratios (HRs) and 95% confidence intervals (CIs) for the association between lifetime alcohol intake and colorectal cancer risk. Heterogeneity in the HRs across subtypes of colorectal cancer was assessed. A positive dose-dependent association between lifetime alcohol intake and overall colorectal cancer risk (mean follow-up = 14.6 years; n = 596 colon and n = 326 rectal cancer) was observed (HR = 1.08, 95% CI: 1.04-1.12 per 10 g/day increment). The risk was greater for rectal than colon cancer (phomogeneity  = 0.02). Alcohol intake was associated with increased risks of KRAS+ (HR = 1.07, 95% CI: 1.00-1.15) and BRAF-/KRAS- (HR = 1.05, 95% CI: 1.00-1.11) but not BRAF+ tumors (HR = 0.89, 95% CI: 0.78-1.01; phomogeneity  = 0.01). Alcohol intake is associated with an increased risk of KRAS+ and BRAF-/KRAS- tumors originating via specific molecular pathways including the traditional adenoma-carcinoma pathway but not with BRAF+ tumors originating via the serrated pathway. Therefore, limiting alcohol intake from a young age might reduce colorectal cancer originating via the traditional adenoma-carcinoma pathway

    The Recent Evolution of the Dwarf Starburst Galaxy NGC 625 from Hubble Space Telescope Imaging

    New HST/WFPC2 imaging of the dwarf starburst galaxy NGC 625 is presented. These data, 80% complete to V and I magnitudes of 26.0 and 25.3, respectively, allow us to study the recent star formation history of NGC 625. We derive a tip of the red giant branch (TRGB) distance modulus of 27.95+/-0.07, corresponding to a distance of 3.89+/-0.22 Mpc, and a location on the far side of the Sculptor Group. NGC 625 has a well-defined radial stellar population gradient, evidenced by a central concentration of young MS stars and an RGB/AGB ratio that increases with galactocentric distance. The prominent AGB is very red, and RGB stars are detected far from the central star forming regions. Using H Alpha and H Beta narrow band imaging and previous optical spectroscopy we identify substantial and varying internal extinction (A_V = 0.0 to 0.6 mag) associated with the central active star formation regions. To better understand the effects of internal extinction on the analysis of young stellar populations, synthetic models are presented which, for the first time, examine and account for this effect. Using the luminous blue helium burning stars, we construct a simple model of the recent (< 100 Myr) star formation in which an elevated but declining star formation rate has been present over this entire period. This is at odds with the presence of spectroscopic W-R features in the major star formation region which imply a short duration (<= 5 Myr) for the recent starburst. This suggests that starbursts displaying W-R features are not necessarily all of a short duration. Finally, we speculate on the possible causes of the present burst of star formation in this apparently isolated galaxy, and compare it to other nearby, well-studied dwarf starburst systems.Comment: 56 pages, including 15 figures (2 in color).     Urban-rural disparities in smoking behaviour in Germany

    BACKGROUND: It is currently not clear whether individuals living in metropolitan areas differ from individuals living in rural and urban areas with respect to smoking behaviours. Therefore, we sought to explore the relation between residential area and smoking behaviours in Germany. METHODS: We used a nationwide German census representative for the general population of Germany. A number of 181,324 subjects aged 10 years or older were included. Information on the average daily usage of cigarettes that have or had been smoked formerly or currently was available in subjects who have ever smoked. A daily consumption of more than 20 cigarettes was considered heavy smoking. Logistic regression analyses were performed sex-stratified and adjusted for relevant confounders. RESULTS: Analyses revealed inhabitants of metropolitan areas to be more likely current smokers than inhabitants of rural areas (odds ratio 1.56, 95%-confidence interval 1.51; 1.62). Among current and former smokers those who lived in urban communities had also increased odds for being heavy smokers than those who lived in rural communities. CONCLUSION: We conclude that living in an urban and particularly living in a metropolitan area is a determinant of both smoking and severity of current smoking. Tobacco control programs should recognize the difference in living conditions between rural and urban areas
