    The Evaluation of Ester Functionalised TCF‐based Fluorescent Probes for the Detection of Bacterial Species

    The ester functionality is commonly seen in the areas of chemical biology and medicinal chemistry for the design of cell‐permeable active molecules. Ester‐based pro‐drug/pro‐sensor strategies are employed to mask polar functional groups (i. e. carboxylic acids) and improve the overall cell permeability of these functional molecules. However, their use as reactive units for sensing applications, including bacterial detection, has not been fully explored. Herein, we synthesised two TCF‐based fluorescent probes, TCF‐OAc and TCF‐OBu. As expected, both TCF‐OAc and TCF‐OBu demonstrated a significant fluorescence (22‐ and 43‐fold, respectively) and colorimetric response (yellow to purple) towards porcine liver esterase (PLE) with a limit of detection of 1.18 mU/mL and 0.45 mU/mL, respectively. With these results in hand, the ability of these probes to detect planktonic suspensions of gram‐positive Staphylococcus aureus (S. aureus) and gram‐negative Pseudomonas aeruginosa (P. aeruginosa), and Escherichia coli (E. coli) were evaluated. Different fluorescence responses for gram‐positive and gram‐negative bacteria were observed between TCF‐OAc and TCF‐OBu. After 1 h incubation, TCF‐OAc proved more sensitive towards S. aureus, demonstrating a significant fluorescence “turn on” response (16‐fold); whereas, TCF‐OBu was more selective towards P. aeruginosa, with a 22‐fold increase in the fluorescence response observed.     Understanding how perceptions of tobacco constituents and the FDA relate to effective and credible tobacco risk messaging: A national phone survey of U.S. adults, 2014–2015

    As reported in the original paper [1], the Center for Regulatory Research on Tobacco Communication conducted a telephone survey in 2014–2015 with a national sample of adults ages 18 and older living in the United States (N = 5014). Poverty level was determined using the household size and income reported by the respondents and applying the federal poverty numbers available from the U.S. Department of Health and Human Services in 2014. A coding error was made during the data recoding process such that 2.7% of respondents (n = 129) were incorrectly classified as living above the poverty line. Below are updated Tables 1, 2 and 4 presenting both the original and corrected estimates. No substantive conclusions reported in the paper were affected by this correction

    Examination of the efficacy of acute L-alanyl-L-glutamine ingestion during hydration stress in endurance exercise

    <p>Abstract</p> <p>Background</p> <p>The effect of acute L-alanyl-L-glutamine (AG; Sustamineℱ) ingestion on performance changes and markers of fluid regulation, immune, inflammatory, oxidative stress, and recovery was examined in response to exhaustive endurance exercise, during and in the absence of dehydration.</p> <p>Methods</p> <p>Ten physically active males (20.8 ± 0.6 y; 176.8 ± 7.2 cm; 77.4 ± 10.5 kg; 12.3 ± 4.6% body fat) volunteered to participate in this study. During the first visit (T1) subjects reported to the laboratory in a euhydrated state to provide a baseline (BL) blood draw and perform a maximal exercise test. In the four subsequent randomly ordered trials, subjects dehydrated to -2.5% of their baseline body mass. For T2, subjects achieved their goal weight and were not rehydrated. During T3 - T5, subjects reached their goal weight and then rehydrated to 1.5% of their baseline body mass by drinking either water (T3) or two different doses (T4 and T5) of the AG supplement (0.05 g·kg<sup>-1 </sup>and 0.2 g·kg<sup>-1</sup>, respectively). Subjects then exercised at a workload that elicited 75% of their VO<sub>2 </sub>max on a cycle ergometer. During T2 - T5 blood draws occurred once goal body mass was achieved (DHY), immediately prior to the exercise stress (RHY), and immediately following the exercise protocol (IP). Resting 24 hour (24P) blood samples were also obtained. Blood samples were analyzed for glutamine, potassium, sodium, aldosterone, arginine vasopressin (AVP), C-reactive protein (CRP), interleukin-6 (IL-6), malondialdehyde (MDA), testosterone, cortisol, ACTH, growth hormone and creatine kinase. Statistical evaluation of performance, hormonal and biochemical changes was accomplished using a repeated measures analysis of variance.</p> <p>Results</p> <p>Glutamine concentrations for T5 were significantly higher at RHY and IP than T2 - T4. When examining performance changes (difference between T2 - T5 and T1), significantly greater times to exhaustion occurred during T4 (130.2 ± 340.2 sec) and T5 (157.4 ± 263.1 sec) compared to T2 (455.6 ± 245.0 sec). Plasma sodium concentrations were greater (p < 0.05) at RHY and IP for T2 than all other trials. Aldosterone concentrations at RHY and IP were significantly lower than that at BL and DHY. AVP was significantly elevated at DHY, RHY and IP compared to BL measures. No significant differences were observed between trials in CRP, IL-6, MDA, or in any of the other hormonal or biochemical measures.</p> <p>Conclusion</p> <p>Results demonstrate that AG supplementation provided a significant ergogenic benefit by increasing time to exhaustion during a mild hydration stress. This ergogenic effect was likely mediated by an enhanced fluid and electrolyte uptake.</p

    A comparison of different investigative interviewing techniques in generating differential recall enhancement and detecting deception

    This study compared the Stepwise Interview, Cognitive Interview, and Reality Interview in detecting deception with inmates. The dependent measures were the amount of unique details provided during the free narrative and mnemonics and the number of words provided during the free narrative and mnemonics of each interview. The Stepwise Interview generated 58.3% accuracy, the Cognitive Interview generated 70.0% accuracy, and the Reality Interview generated 93.3% accuracy. The different tasks of these interviews increased the differences between honest and deceptive statements and therefore, increased the accuracy in detection of deception.     Mesoscale variability of the summer bloom over the northern Ross Sea shelf: A tale of two banks

    Multi-year satellite records indicate an asymmetric spatial pattern in the summer bloom in the Northern Ross Sea, with the largest blooms over the shallows of Pennell Bank compared to Mawson Bank. In 2010–2011, high-resolution spatiotemporal in situ sampling focused on these two banks to better understand factors contributing to this pattern. Dissolved and particulate Fe profiles suggested similar surface water depletion of dissolved Fe on both banks. The surface sediments and velocity observations indicate a more energetic water column over Mawson Bank. Consequently, the surface mixed layer over Pennell Bank was more homogeneous and shallower. Over Mawson Bank we observed a thicker more homogeneous bottom boundary layer resulting from stronger tidal and sub-tidal currents. These stronger currents scour the seafloor resulting in sediments less likely to release additional sedimentary iron. Estimates of the quantum yield of photosynthesis and the initial slope of the photosynthesis-irradiance response were lower over Mawson Bank, indicating higher iron stress over Mawson Bank. Overall, the apparent additional sedimentary source of iron to, and longer surface residence time over Pennell Bank, as well as the reduced fluxes from the more isolated bottom mixed layer over Mawson Bank, sustain the observed asymmetric pattern across both banks
