    Structural effects of phosphate groups on apatite formation in a copolymer modified with Ca2+ in a simulated body fluid

    Organicā€“inorganic composites are novel bone substitutes that can ameliorate the mismatch of Young\u27s moduli between natural bone and implanted ceramics. Phosphate groups contribute to the formation of apatite in a simulated body fluid (SBF) and the adhesion of osteoblast-like cells. Therefore, modification of a polymer with these functional groups is expected to enhance the ability of the organicā€“inorganic composite to bond with bone. Two phosphate groups have been used, phosphonic acid (ā€“Cā€“PO3H2) and phosphoric acid (ā€“Oā€“PO3H2). However, the effects of structural differences between these phosphate groups have not been clarified. In this study, the apatite formation of copolymers modified with Ca2+ and either ā€“Cā€“PO3H2 or ā€“Oā€“PO3H2 was examined. The mechanism of apatite formation is discussed based on analytical and computational approaches. The copolymers containing ā€“Oā€“PO3H2, but not those containing ā€“Cā€“PO3H2, formed apatite in the SBF, although both released similar amounts of Ca2+ into the SBF. Adsorption of HPO42āˆ’ from ā€“Oā€“PO3H2 in the SBF following Ca2+ adsorption was confirmed by zeta-potential measurement and X-ray photoelectron spectroscopy. The measurement of the complex formation constant revealed that the ā€“Oā€“PO32āˆ’ā‹ÆCa2+ complex was thermodynamically unstable enough to convert into CaHPO4, which was not the case with ā€“Cā€“PO32āˆ’ā‹ÆCa2+. The formation of CaHPO4-based clusters was found to be a key factor for apatite nucleation. In conclusion, this study revealed that modification with ā€“Oā€“PO3H2 was more effective for enhancing apatite formation compared with ā€“Cā€“PO3H2

    The EP4-ERK-dependent pathway stimulates osteo-adipogenic progenitor proliferation resulting in increased adipogenesis in fetal rat calvaria cell cultures

    We previously reported that fetal rat calvaria (RC) cells are osteo-adipogenic bipotential and that PGE2 receptors EP2 and EP4 are involved in bone nodule formation via both common and distinct MAPK pathways in RC cell cultures. Because PGE2 participates in multiple biological processes including adipogenesis, it is of interest to determine the additional role(s) of PGE2 in RC cells. PGE2 increased the number of adipocyte colonies when RC cells were treated during proliferation but not other development stages. Of four EP agonists tested, the EP4 agonist ONO-AE1-437 (EP4A) was the most effective in promoting adipogenesis. Concomitantly, EP4A increased the number of cells with BrdU labeling and gene expression of CCAAT/enhancer binding protein (C/EBP)Ī“ and c-fos but not peroxisome proliferator-activated receptor Ī³2 and C/EBPĪ±. Amongst MAPK inhibitors, U0126, an inhibitor of MEK1/2, abrogated the EP4A-dependent effects. Our results suggest that the PGE2ā€“EP4-ERK pathway increases the number of osteo-adipogenic bipotential progenitor cells, with a resultant increase in adipogenesis in RC cell cultures.This work was supported in part by grants from the Ministry of Education, Science, Sports and Culture of Japan (13771074 to YY)and Ono Pharmaceutical Co.     1Ī±,25-dihydroxyvitamin D3 acts predominately in mature osteoblasts under conditions of high extracellular phosphate to increase fibroblast growth factor 23 production in vitro

    Osteoblasts/osteocytes are the principle sources of fibroblast growth factor 23 (FGF23), a phosphaturic hormone, but the regulation of FGF23 expression during osteoblast development remains uncertain. Because 1Ī±,25-dihydroxyvitamin D3 (1,25(OH)2D3) and inorganic phosphate (Pi) may act as potent activators of FGF23 expression, we estimated how these molecules regulate FGF23 expression during rat osteoblast development in vitro. 1,25(OH)2D3-dependent FGF23 production was restricted largely to mature cells in correlation with increased vitamin D receptor (VDR) mRNA levels, in particular, when Pi was present. Pi alone and more so in combination with 1,25(OH)2D3 increased FGF23 production and VDR mRNA expression. Parathyroid hormone, stanniocalcin 1, prostaglandin E2, FGF2, and foscarnet did not increase FGF23 mRNA expression. Thus, these results suggest that 1,25(OH)2D3 may exert its largest effect on FGF23 expression/production when exposed to high levels of extracellular Pi in osteoblasts/osteocytes

    Lipomatous Pseudohypertrophy of the Pancreas Taking the Form of Huge Massive Lesion of the Pancreatic Head

    A 70-year-old woman presented with hypogastric pain. Computed tomography and magnetic resonance imaging revealed a retroperitoneal tumor 18.0 cm in diameter with fatty tissue density, ventrally compressing the pancreatic head. We suspected a well-differentiated liposarcoma compressing the pancreas. At laparotomy, the tumor mass was the size of an infant's head; its center was located in the area corresponding to the pancreatic uncus. It was continuous with the pancreatic parenchyma through a poorly demarcated border, and we resected as much of the tumor mass as possible while conserving the pancreatic capsule. Histopathological examination indicated lipomatous pseudohypertrophy of the pancreas with proliferation of mature fatty tissue as the main constituent. At the periphery, islands of acinar tissue were retained among the fatty infiltration, which also contained branches of the pancreatic duct and islets of Langerhans. Previous reports have stated that this disorder only causes fatty replacements throughout the pancreas or in the pancreatic body and tail; however, in this patient, imaging and macroscopic examination revealed no fatty replacements in the pancreatic body and tail. We report this case, which we consider extremely rare, along with a brief review of the literature

    Aluminum Silicate Nanotube Coating of Siloxane-Poly(lactic acid)-Vaterite Composite Fibermats for Bone Regeneration

    In our earlier work, a flexible fibermat consisting of a biodegradable composite with soluble silicate species, which has been reported to enhance bone formation, was prepared successfully using poly(L-lactic acid) and siloxane-containing calcium carbonate particles by electrospinning. The fibermat showed enhanced bone formation in an in vivo test. In the present work, to improve the hydrophilicity of skeletal fibers in a fibermat, they were coated with nanotubular aluminum silicate crystals, which have a hydrophilic surface that has excellent affinity to body fluids and a high surface area advantageous for pronounced protein adsorption. The nanotubes were coated easily on the fiber surface using an electrophoretic method. In a conventional contact angle test, a drop of water rapidly penetrated into the nanotube-coated fibermat. The culture test using murine osteoblast-like cells (MC3T3-E1) showed that the cell attachment to the nanotube-coated fibermat at an early stage after seeding was enhanced in comparison with that to the noncoated one. This approach may provide a new method of improving the surface of polymer-based biomaterials

    Preparation and Rheological Characterization of Imogolite Hydrogels

    Imogolite, one of the aluminium silicates, has a nanotube structure and has been known to form gel under alkaline condition. Imogolite nanotubes were synthesized in an acidic solution with various tube lengths by controlling the aging time from 1ā€‰d to 14ā€‰d. The length of the nanotubes grew from 100ā€‰nm to severalā€‰Ī¼m as the aging time. Pure imogolite hydrogels were prepared by applying a salting-out method and centrifugation from its dispersed solutions with various tube lengths and solution pH. Imogolite hydrogel can be classified as the physically cross-linked one; the structure of the gel network is considered to be the entanglements and hydrogen bonding among nanotubes. The theoretical water contents of the prepared hydrogels were calculated as āˆ¼99.7% in average. Gelation percentage significantly increased as the length of imogolite nanotubes. Whereas hydrogel prepared from 4ā€‰d aging sample showed the highest storage modulus of āˆ¼970ā€‰Pa, it was found that the hydrogel could be prepared in the pH range from 6 to 10. The gel strength reached the highest value of 1000ā€‰Pa when the gel was prepared from the imogolite dispersed solution of pH 8. It could be explained by the surface charge variation of the imogolite
