    Studies on the influence of mineral ions on bacterial respiration Report I: The decrease of respiration by washing organisms and the influence of mineral ions on it

    This experiment was performed to study the influence of washing on bacterial respiration and, moreover, to study the effects of various mineral ions on it. Organisms: Bacillus dysenteriae (Komagome B III and Ohno's bacillus) Bacillus coli communis, Bacillus coli communior, Staphylococcus aureus, Staphylococcus albus, Bacillus pneumoniae, Bacillus pyocyaneus, Bacillus proteus (H and Otype) and Bacillus cholerae. Substrates: glucose, α-ketoglutaric acid, pyruvic acid, succinic acid and malic acid. Mineral ions: Fe(++), Fe(+++), Mg(++), Cu(++) and Zn(++) 1) The decrease of respiration by washing varies according to the species of Organisms and the sorts of substrates. 2) The organisms of which respiration is remarkably decreased by washing are Staphylococcus and Bacillus coli. However, the respiration of Bacillus proteus and Bacillus pneumoniae is little decreased by washing. 3) The substrates in which the washing of organisms results in noticeable decrease of bacterial respiration are glucose and pyruvic acid. However, it shows little decrease in malic acid. 4) The addition of mineral ions can restore the respiration decreased by washing, of which the degree of restoration is, however, different according to the species of organisms and the sorts of substrates. 5) The most effective method for adding the mineral ions is to wash organisms with buffer containing the mineral ions and then suspend the washed organisms in the same buffer as that used for washing

    Cardiomyocytes fuse with surrounding noncardiomyocytes and reenter the cell cycle

    The concept of the plasticity or transdifferentiation of adult stem cells has been challenged by the phenomenon of cell fusion. In this work, we examined whether neonatal cardiomyocytes fuse with various somatic cells including endothelial cells, cardiac fibroblasts, bone marrow cells, and endothelial progenitor cells spontaneously in vitro. When cardiomyocytes were cocultured with endothelial cells or cardiac fibroblasts, they fused and showed phenotypes of cardiomyocytes. Furthermore, cardiomyocytes reentered the G2-M phase in the cell cycle after fusing with proliferative noncardiomyocytes. Transplanted endothelial cells or skeletal muscle–derived cells fused with adult cardiomyocytes in vivo. In the cryoinjured heart, there were Ki67-positive cells that expressed both cardiac and endothelial lineage marker proteins. These results suggest that cardiomyocytes fuse with other cells and enter the cell cycle by maintaining their phenotypes

    Paeoniflorin ameliorates acquisition impairment of a simple operant discrimination performance caused by unilateral nucleus basalis magnocellularis lesion in rats

    The effect of paeoniflorin on learning impairment of operant brightness discrimination performance was investigated in rats with unilateral nucleus basalis magnocellularis(NBM)lesion. The animals with unilateral NBM lesion exhibited a significant acquisition impairment of brightness discrimination task during the early phase of a training period(1-5days after starting the training session). When administered daily during a training period,paeoniflorin significantly improved the learning impairment of unilateral NBM-lesioned rats at 0.01 but not 0.1mg/kg/day(p.o.). Tacrine (0.3mg/kg/day,p.o.), a cholinesterase inhibitor,also significantly ameliorated the learning deficit. These results suggest that paeoniflorin improves the impairment of non-spatial learning performance caused by cholinergic dysfunction in rats and that it may have a beneficial effect on senile dementia

    以下の報告は、平成14年8月26日に開催された岡山大学教育学部附属教育実践総合センター研修講座の教育臨床部門の分科会「シンポジウム:教育臨床における評価・見立てについて」での報告内容をもとに、発表者がそれぞれに再構成して書き下ろしたものである。シンポジウムのコーディネーターは塚原千秋が行った。そして、まず心理臨床の視点から山本が教育臨床の見立てとは何かについて模索した見解を報告した。続いて専任スクールカウンセラーの立場から西山が米国のスクールカウンセリングをモデルにした評価の仕方を詳しい報告にした。最後に学校でのブリーフセラピーの可能性を探る教師の視点から赤沢が実践的な方法と認識を報告した。This paper is aimed at reporting the matter of symposium on the subject of clinical evaluation and assessment in school setting. The focus of discussion is especially on our needs to evaluate and assess students, personnel, parents, ourselves and "school guidance and counseling program". Each symposist expresses ones views and ideas, which are implemented through their work in school setting, and have to be elaborated in the future

    Novel Cyclic Dipeptide Dehydrogenase and Assay Method for Activity

    The cell-free extract prepared from cells of an albonoursin-producing actinomycete Streptomyces sp. KO2388 was found to catalyze the dehydrogenation of cyclo (L-Phe-L-Leu) (CFL) to albonoursin. This is the first report for the dehydrogenation at the α,β-positions of amino acid residues. The simple method for determining the dehydrogenation activity was devised by measuring the increase in UV absorption of the reaction mixture at 317nm, λmax(ε25,400) of albonoursin, where CFL had no absorption. Phenazine methosulfate was the most active cofactor for the dehydrogenation among several hydrogen acceptors.アルボノルシン生産菌、Streptomyces sp. KO2388株の無細胞抽出液が、cyclo(L-Phe-L-Leu)からアルボノルシンへの脱水素反応を触媒することを明らかにした。アミノ酸残基のα,β-位での脱水素反応を酵素レベルで明らかにしたのはこの報告が初めてである。本反応の簡便な測定法として、基質cyclo(L-Phe-L-Leu)には認められない。アルボノルシンの特異的な317nmにおける紫外吸収の増加を測定する方法を考案した。本方法を用いて数種の水素受容体を試験したところ、フェナジンメトサルフェートが最も有効であると判った

    A novel LIM protein Cal promotes cardiac differentiation by association with CSX/NKX2-5

    The cardiac homeobox transcription factor CSX/NKX2-5 plays an important role in vertebrate heart development. Using a yeast two-hybrid screening, we identified a novel LIM domain–containing protein, named CSX-associated LIM protein (Cal), that interacts with CSX/NKX2-5. CSX/NKX2-5 and Cal associate with each other both in vivo and in vitro, and the LIM domains of Cal and the homeodomain of CSX/NKX2-5 were necessary for mutual binding. Cal itself possessed the transcription-promoting activity, and cotransfection of Cal enhanced CSX/NKX2-5–induced activation of atrial natriuretic peptide gene promoter. Cal contained a functional nuclear export signal and shuttled from the cytoplasm into the nucleus in response to calcium. Accumulation of Cal in the nucleus of P19CL6 cells promoted myocardial cell differentiation accompanied by increased expression levels of the target genes of CSX/NKX2-5. These results suggest that a novel LIM protein Cal induces cardiomyocyte differentiation through its dynamic intracellular shuttling and association with CSX/NKX2-5
