    Differences in strategies for coping with menopausal symptoms in full-time workers and part-time workers in Japan

    Working hours and working conditions are different for full-time workers and part-time workers. In this study, it was hypothesized that the strategies for coping with menopausal symptoms, such as hot flashes, night sweats and insomnia, and the proportion of women receiving clinical care differ between full-time and part-time workers. Four hundred and eighty-seven Japanese female workers aged 45 to 60 years responded to a health questionnaire about their understanding of menopause and their strategies for coping with menopausal symptoms. Based on the surveyed responses, the proportions of women with menopausal symptoms were 43.0% (80/186) in full-time workers and 48.2% (145/301) in part-time workers. There was no significant difference in the female worker response rate, nor were there significant differences in the proportions of full-time workers and part-time workers who had an understanding of menopause causes and treatments and who were able to cope with menopausal-related symptoms. However, there were significantly more full-time workers than part-time workers who received routine clinical care and who visited a hospital. On the other hand, significantly more part-time workers than full-time workers exercised and took dietary supplements as a means of coping with their symptoms. There are differences between female full-time and part-time workers regarding the strategies employed for coping with menopausal symptoms. The proportion of women receiving clinical care for their symptoms is greater among full-time employees than part-time employees. A physical examination can be a valuable opportunity for working women with menopausal symptoms to receive advice from a doctor or a nurse practitioner

    Anti-Dermatophagoides farinae type I and II IgE antibodies in allergic rhinitis

    ABSTRACTSera from 27 patients with mite-sensitive allergic rhinitis, without atopic dermatitis and bronchial asthma, were examined for anti-Der f I and anti-Der f II IgE antibody contents by enzyme-linked immunosorbent assay (ELISA). Anti-Der f I and anti-Der f II IgE antibody levels were 14.78 Ā± 1.34 and 32.68 Ā± 0.88 ng/mL (mean Ā± SEM), respectively. The anti-Der f II IgE antibody was predominant over the anti-Der f I IgE antibody in these patients.In comparison with the results of a previous study the present study indicates that the ratio between serum anti-Der f I and II IgE antibodies in patients with allergic rhinitis indicated the same pattern as in that of patients with bronchial asthma, while the inverse was the case in patients with atopic dermatitis.These results indicate that immunological features and major allergen molecules could be different in different atopic diseases. At present it is not clear where this difference comes from, but the route of immunological sensitization (via respiratory tract vs via skin) might result in the difference

    Objective: The main objective was to ascertain the typical menopausal symptoms and job-related stress factors in Japanese nurses during the menopausal transition, and the associations of menopausal symptoms with job-related stress. A supplementary objective was to determine whether there were any differences in menopausal symptoms and job-related stress factors among nurses in managerial positions. Methods: One thousand seven hundred female registered nurses aged 45ā€“60 years who were working in hospitals in Japan were asked to complete a self-administered survey that included Greeneā€™s Climacteric Scale and the Brief Job Stress Questionnaire. Results: The proportions of nurses who reported feelings of tiredness, irritability and difficulty in concentration were higher than the proportions with other menopausal symptoms. The proportions of nurses reporting feeling unhappy or depressed and having crying spells were higher among nurses in managerial positions than among other nurses. Stresses related to ā€˜quantitative overloadā€™ on the Brief Job Stress Questionnaire among nurses in managerial positions were significantly greater than among nurses not in managerial positions, while stresses related to ā€˜physical overloadā€™, ā€˜job controlā€™, ā€˜skill discretionā€™, ā€˜workplace environmentā€™ and ā€˜job satisfactionā€™ among nurses not in managerial positions were significantly greater than they were among nurses in managerial positions. Psychological symptoms were significantly correlated with poor job-related interpersonal relationships. Conclusions: Health care practitioners should be aware that menopausal symptoms are associated with job-related stress during the menopausal transition. Information on the differences in these associations between nurses in managerial positions and other nurses is important as it will allow their health care to be managed on a more individual basis

    Contribution of Thyrotropin-Releasing Hormone to Cerebellar Long-Term Depression and Motor Learning

    Thyrotropin-releasing hormone (TRH) regulates various physiological activities through activation of receptors expressed in a broad range of cells in the central nervous system. The cerebellum expresses TRH receptors in granule cells and molecular layer interneurons. However, the function of TRH in the cerebellum remains to be clarified. Here, using TRH knockout (KO) mice we studied the role of TRH in the cerebellum. Immunohistochemistry showed no gross morphological differences between KO mice and wild-type (WT) littermates in the cerebellum. In the rotarod test, the initial performance of KO mice was comparable to that of WT littermates, but the learning speed of KO mice was significantly lower than that of WT littermates, suggesting impaired motor learning. The motor learning deficit in KO mice was rescued by intraperitoneal injection of TRH. Electrophysiology revealed absence of long-term depression (LTD) at parallel fiber-Purkinje cell synapses in KO mice, which was rescued by bath-application of TRH. TRH was shown to increase cyclic guanosine monophosphate (cGMP) content in the cerebellum. Since nitric oxide (NO) stimulates cGMP synthesis in the cerebellum, we examined whether NO-cGMP pathway was involved in TRH-mediated LTD rescue in KO mice. Pharmacological blockade of NO synthase and subsequent cGMP production prevented TRH-induced LTD expression in KO mice, whereas increase in cGMP signal in Purkinje cells by 8-bromoguanosine cyclic 3ā€™,5ā€™-monophosphate, a membrane-permeable cGMP analog, restored LTD without TRH application. These results suggest that TRH is involved in cerebellar LTD presumably by upregulating the basal cGMP level in Purkinje cells, and, consequently, in motor learning

    Protein kinase CĪ³ negatively regulates the intrinsic excitability in zebrin-negative cerebellar Purkinje cells

    Protein kinase C Ī³ (PKCĪ³), a neuronal isoform present exclusively in the central nervous system, is most abundantly expressed in cerebellar Purkinje cells (PCs). Targeted deletion of PKCĪ³ causes a climbing fiber synapse elimination in developing PCs and motor deficit. However, physiological roles of PKCĪ³ in adult mouse PCs are little understood. In this study, we aimed to unravel the roles of PKCĪ³ in mature mouse PCs by deleting PKCĪ³ from adult mouse PCs of PKCĪ³fl/fl mice via cerebellar injection of adeno-associated virus (AAV) vectors expressing Cre recombinase under the control of the PC-specific L7-6 promoter. Whole cell patch-clamp recording of PCs showed higher intrinsic excitability in PCs virally lacking PKCĪ³ [PKCĪ³-conditional knockout (PKCĪ³-cKO) PCs] than in wild-type (WT) mouse PCs in the zebrin-negative module, but not in the zebrin-positive module. AAV-mediated PKCĪ³ re-expression in PKCĪ³-deficient mouse PCs in the zebrin-negative module restored the enhanced intrinsic excitability to a level comparable to that of wild-type mouse PCs. In parallel with higher intrinsic excitability, we found larger hyperpolarization-activated cyclic nucleotide-gated (HCN) channel currents in PKCĪ³-cKO PCs located in the zebrin-negative module, compared with those in WT mouse PCs in the same region. However, pharmacological inhibition of the HCN currents did not restore the enhanced intrinsic excitability in PKCĪ³-cKO PCs in the zebrin-negative module. These results suggested that PKCĪ³ suppresses the intrinsic excitability in zebrin-negative PCs, which is likely independent of the HCN current inhibition

    In Vitro Assessment of Factors Affecting the Apparent Diffusion Coefficient of Ramos Cells Using Bio-phantoms

    The roles of cell density, extracellular space, intracellular factors, and apoptosis induced by the molecularly targeted drug rituximab on the apparent diffusion coefficient (ADC) values were investigated using bio-phantoms. In these bio-phantoms, Ramos cells (a human Burkittļ½¾s lymphoma cell line) were encapsulated in gellan gum. The ADC values decreased linearly with the increase in cell density, and declined steeply when the extracellular space became less than 4 Ī¼m. The analysis of ADC values after destruction of the cellular membrane by sonication indicated that approximately 65% of the ADC values of normal cells originate from the cell structures made of membranes and that the remaining 35% originate from intracellular components. Microparticles, defined as particles smaller than the normal cells, increased in number after rituximab treatments, migrated to the extracellular space and significantly decreased the ADC values of bio-phantoms during apoptosis. An in vitro study using bio-phantoms was conducted to quantitatively clarify the roles of cellular factors and of extracellular space in determining the ADC values yielded by tumor cells and the mechanism by which apoptosis changes those values

    Chronic Treatment with a Water-Soluble Extract from the Culture Medium of Ganoderma lucidum

    Type 2 diabetes mellitus has been known to increase systemic oxidative stress by chronic hyperglycemia and visceral obesity and aggravate cerebral ischemic injury. On the basis of our previous study regarding a water-soluble extract from the culture medium of Ganoderma lucidum mycelia (designed as MAK), which exerts antioxidative and neuroprotective effects, the present study was conducted to evaluate the preventive effects of MAK on apoptosis and necroptosis (a programmed necrosis) induced by hypoxia/ischemia (H/I) in type 2 diabetic KKAy mice. H/I was induced by a combination of unilateral common carotid artery ligation with hypoxia (8% O2 for 20ā€‰min) and subsequent reoxygenation. Pretreatment with MAK (1ā€‰g/kg, p.o.) for a week significantly reduced H/I-induced neurological deficits and brain infarction volume assessed at 24ā€‰h of reoxygenation. Histochemical analysis showed that MAK significantly suppressed superoxide production, neuronal cell death, and vacuolation in the ischemic penumbra, which was accompanied by a decrease in the numbers of TUNEL- or cleaved caspase-3-positive cells. Furthermore, MAK decreased the expression of receptor-interacting protein kinase 3 mRNA and protein, a key molecule for necroptosis. These results suggest that MAK confers resistance to apoptotic and necroptotic cell death and relieves H/I-induced cerebral ischemic injury in type 2 diabetic mice

    Crisis-management, Anti-stigma, and Mental Health Literacy Program for University Students (CAMPUS): A preliminary evaluation of suicide prevention [version 2; peer review: 2 approved]

    Background: University students have specific risk factors for suicide, necessitating targeted prevention programs. This preliminary study evaluated the efficacy of the Crisis-management, Anti-stigma, Mental health literacy Program for University Students (CAMPUS) for reduction of risk factors and promotion of preventative behaviors. Methods: A total of 136 medical students attended the CAMPUS as a required course at the national university in Japan. The CAMPUS consisted of a lecture and two group sessions covering mental health literacy, self-stigma, and gatekeeper efficacy (e.g., identifying and helping at-risk individuals). The students were asked to role-play based on a movie about gatekeepers and scripts about self-stigma and suicide-related issues. Participants completed questionnaires on suicidal thoughts, depression, help-seeking intentions, self-efficacy as gatekeepers, self-concealment, and self-acceptance. A total of 121 students completed the questionnaires pre- and post-program, and 107 students also responded six months later. Results: Students demonstrated significantly reduced overall suicide thoughts six months post-program compared to before the program. In addition, gatekeeper self-efficacy, help-seeking intentions for formal resources, and self-acceptance were improved in the students six month after the program. Conclusions: The CAMPUS suggested effective at reducing suicidal people and promoting preventative psychological tendencies among medial students. This study was a one-group pre post design study without control group. The CAMPUS program was delivered as a mandatory requirement to a group with relatively low suicide risk. Further studies are required to assess its suitability for the general university student population

    In Vitro Assessment of Factors Affecting the Apparent Diffusion Coefficient of Jurkat Cells Using Bio-phantoms

    It is well known that many tumor tissues show lower apparent diffusion coefficient (ADC) values, and that several factors are involved in the reduction of ADC values. The aim of this study was to clarify how much each factor contributes to decreases in ADC values. We investigate the roles of cell density, extracellular space, intracellular factors, apoptosis and necrosis in ADC values using bio-phantoms. The ADC values of bio-phantoms, in which Jurkat cells were encapsulated by gellan gum, were measured by a 1.5-Tesla magnetic resonance imaging device with constant diffusion time of 30sec. Heating at 42ā„ƒ was used to induce apoptosis while heating at 48ā„ƒ was used to induce necrosis. Cell death after heating was evaluated by flow cytometric analysis and electron microscopy. The ADC values of bio-phantoms including non-heated cells decreased linearly with increases in cell density, and showed a steep decline when the distance between cells became less than 3Ī¼m. The analysis of ADC values of cells after destruction of cellular structures by sonication suggested that approximately two-thirds of the ADC values of cells originate from their cellular structures. The ADC values of bio-phantoms including necrotic cells increased while those including apoptotic cells decreased. This study quantitatively clarified the role of the cellular factors and the extracellular space in determining the ADC values produced by tumor cells. The intermediate diffusion time of 30msec might be optimal to distinguish between apoptosis and necrosis
