    STRUCTURAL EVOLUTION OF AN INTRACRATONIC RIFT SYSTEM; MISSISSIPPI VALLEY GRABEN, ROUGH CREEK GRABEN, AND ROME TROUGH OF KENTUCKY, USA

    As indicated by drilling and geophysical data, the Mississippi Valley Graben, the Rough Creek Graben, together with the Rome Trough of eastern Kentucky and West Virginia, are fault-bounded graben structures filled with as much as 27,000 feet of Early to Middle Cambrian sediments. Detailed regional mapping of Cambrian and younger strata within and surrounding these structures indicates that they formed contemporaneously. The proximity of these structures suggests they developed within the same regional stress fields and tectonic environments. These three structures are mechanically and kinematically connected, and formed part of a single continent-scale rift system produced during the breakup of Rodinia and the separation of Laurentia from Amazonia. Data including stratigraphic tops from 1,764 wells, interpretations of 106 seismic profiles, aeromagnetic and gravity survey analysis, and mapped surface geology and structures were used within this project. Seven stratigraphic packages resolvable in both geophysical well logs and reflection seismic profiles were mapped in the subsurface across parts of Kentucky, Ohio, Indiana, Illinois, Missouri, and Tennessee. These stratigraphic units were then analyzed through structure maps, isopachous maps, and across 12 regional well-based cross sections. Detailed analysis of thickness patterns of seven major stratigraphic packages was used to identify the locations and timing of major fault movements within the study area. The regional patterns of fault movements through time were used to investigate how the structures evolved in response to the tectonic episodes in southeastern Laurentia during the Cambrian through Devonian Periods. Active rifting of the Precambrian crystalline bedrock began by the Early Cambrian, and resulted in a thick deposit of Reelfoot Arkose and Eau Claire Formation within the Mississippi Valley and Rough Creek Grabens, and the Rome Formation and Conasauga Group within the Rome Trough. Major tectonic extension ended by the Late Cambrian, prior to the deposition of the Knox Supergroup. Counter-clockwise rotation of the regional sigma-1 stress field between the Middle Ordovician and Early Mississippian (Taconic through Acadian Orogenies) resulted in the reactivation of varying sets of preexisting faults through time.     A Pilot Randomized Clinical Trial of a Teamwork Intervention for Heart Failure Care Dyads

    Background: Dyadic heart failure (HF) management can improve outcomes for patients and caregivers and can be enhanced through eHealth interventions. Objective: To evaluate the feasibility, acceptability, and preliminary efficacy of an eHealth dyadic teamwork intervention, compared to an attention control condition. Methods: We recruited 29 HF patient-caregiver dyads from inpatient units and randomized dyads to an intervention or a control group. We calculated enrollment and retention rates, described acceptability using interview and questionnaire data, and computed intervention effect sizes. Results: 37% of eligible dyads agreed to participate and 93% of randomized participants completed follow-up questionnaires. Participants found both study conditions to be acceptable. Between-group effect sizes suggested that the intervention led to improvements in relationship quality, self-efficacy, and quality of life for patients and caregivers. Conclusions: Dyadic recruitment from acute care settings is challenging. Findings provide initial evidence that our intervention can contribute to better health outcomes for HF dyads

    HIV-1 pol Diversity among Female Bar and Hotel Workers in Northern Tanzania

    A national ART program was launched in Tanzania in October 2004. Due to the existence of multiple HIV-1 subtypes and recombinant viruses co-circulating in Tanzania, it is important to monitor rates of drug resistance. The present study determined the prevalence of HIV-1 drug resistance mutations among ART-naive female bar and hotel workers, a high-risk population for HIV-1 infection in Moshi, Tanzania. A partial HIV-1 pol gene was analyzed by single-genome amplification and sequencing in 45 subjects (622 pol sequences total; median number of sequences per subject, 13; IQR 5–20) in samples collected in 2005. The prevalence of HIV-1 subtypes A1, C, and D, and inter-subtype recombinant viruses, was 36%, 29%, 9% and 27%, respectively. Thirteen different recombination patterns included D/A1/D, C/A1, A1/C/A1, A1/U/A1, C/U/A1, C/A1, U/D/U, D/A1/D, A1/C, A1/C, A2/C/A2, CRF10_CD/C/CRF10_CD and CRF35_AD/A1/CRF35_AD. CRF35_AD was identified in Tanzania for the first time. All recombinant viruses in this study were unique, suggesting ongoing recombination processes among circulating HIV-1 variants. The prevalence of multiple infections in this population was 16% (n = 7). Primary HIV-1 drug resistance mutations to RT inhibitors were identified in three (7%) subjects (K65R plus Y181C; N60D; and V106M). In some subjects, polymorphisms were observed at the RT positions 41, 69, 75, 98, 101, 179, 190, and 215. Secondary mutations associated with NNRTIs were observed at the RT positions 90 (7%) and 138 (6%). In the protease gene, three subjects (7%) had M46I/L mutations. All subjects in this study had HIV-1 subtype-specific natural polymorphisms at positions 36, 69, 89 and 93 that are associated with drug resistance in HIV-1 subtype B. These results suggested that HIV-1 drug resistance mutations and natural polymorphisms existed in this population before the initiation of the national ART program. With increasing use of ARV, these results highlight the importance of drug resistance monitoring in Tanzania

    Fibrinogen production is enhanced in an in-vitro model of non-alcoholic fatty liver disease: An isolated risk factor for cardiovascular events?

    BACKGROUND: Cardiovascular disease (CVD) remains the major cause of excess mortality in patients with non-alcoholic fatty liver disease (NAFLD). The aim of this study was to investigate the individual contribution of NAFLD to CVD risk factors in the absence of pathogenic influences from other comorbidities often found in NAFLD patients, by using an established in-vitro model of hepatic steatosis. METHODS: Histopathological events in non-alcoholic fatty liver disease were recapitulated by focused metabolic nutrient overload of hepatoblastoma C3A cells, using oleate-treated-cells and untreated controls for comparison. Microarray and proteomic data from cell culture experiments were integrated into a custom-built systems biology database and proteogenomics analysis performed. Candidate genes with significant dysregulation and concomitant changes in protein abundance were identified and STRING association and enrichment analysis performed to identify putative pathogenic pathways. RESULTS: The search strategy yielded 3 candidate genes that were specifically and significantly up-regulated in nutrient-overloaded cells compared to untreated controls: fibrinogen alpha chain (2.2 fold), fibrinogen beta chain (2.3 fold) and fibrinogen gamma chain (2.1 fold) (all rank products pfp <0.05). Fibrinogen alpha and gamma chain also demonstrated significant concomitant increases in protein abundance (3.8-fold and 2.0-fold, respectively, p <0.05). CONCLUSIONS: In-vitro modelling of NAFLD and reactive oxygen species formation in nutrient overloaded C3A cells, in the absence of pathogenic influences from other comorbidities, suggests that NAFLD is an isolated determinant of CVD. Nutrient overload-induced up-regulation of all three fibrinogen component subunits of the coagulation cascade provides a possible mechanism to explain the excess CVD mortality observed in NAFLD patients.     Phase I Activities of the Earth Mapping Resources Initiative (Earth MRI) in the Western Kentucky Fluorspar District

    The Western Kentucky Fluorspar District (WKFD), which comprises the Kentucky portion of the Illinois–Kentucky Fluorspar District, is well known for the occurrence of strategically important critical mineral resources, such as fluorite (fluorspar) and rare earth elements (REEs). Due to technological advances, demand has increased for critical minerals used in cell phones, magnets in wind turbines, batteries for electric vehicles, smart weapons, and other devices and systems. The overall objective of the Earth Mapping Resources Initiative (Earth MRI) program, directed by the U.S. Geological Survey (USGS), is to gather information that will help better understand the distribution of critical minerals in the United States. This report details the Phase I Earth MRI activities conducted by the Kentucky Geological Survey (KGS), including a review of more than 1,400 mineral and rock specimens that resulted in the inventory of 486 specimens relevant to future Earth MRI work. Furthermore, 3,590 digital files were reviewed and indexed with keywords and other attributes to create an upgraded online map service, the Kentucky Mineral Resources Information Map Service, with advanced search functions. Thirty-nine probable igneous dikes that may be an economically viable source of REEs were identified using a filtered aeromagnetic dataset provided by the USGS, and these features were incorporated into a 1:50,000-scale geological map that covers the WKFD. The total REE content (including yttrium and scandium) in 66 analyzed dike samples range from 152 ppm to 620 ppm with an average of 341 ppm. The various types of igneous dikes that have been identified—alnöite, aillikite, and rocks in which carbonate alteration predominates—have overlapping but somewhat distinct concentrations of REEs with alnöite samples having the highest, the carbonate alteration predominant samples the lowest, and the aillikite samples intermediate average total REE values. The relatively high REE content in one massive calcite vein (280 ppm) suggests remobilization of REEs and warrants further study of fluid–rock interactions to better understand the mineral system of the WKFD
