LOAD TESTS ON THE FOUR-SPAN STRUCTURE OF THE RAILWAY BRIDGE OVER THE TISZA AT CSONGRÁD-SZENTES
New bridges can be opened to traffic only after a subsequent load test. The minimal
aim of these checks is to show that the structure fulfils the design requirements given in
the corresponding Code and its real behaviour approximates the expected one. However,
these pieces of information do not provide an explanation for all the deviations. Therefore,
if the objective and financial conditions are given, it is advisable to carry out additional
measurements, which can be analyzed later to describe more accurately the behaviour
of the structure, which, in turn, widens the possibilities of assuming a more realistic
structural model and which provides aid and reference point for design practice in the
future
Evolutionarily conserved role and physiological relevance of a STX17/Syx17 (syntaxin 17)-containing SNARE complex in autophagosome fusion with endosomes and lysosomes.
Phagophores engulf cytoplasmic material and give rise to autophagosomes, double-membrane vesicles mediating cargo transport to lysosomes for degradation. The regulation of autophagosome fusion with endosomes and lysosomes during autophagy has remained poorly characterized. Two recent papers conclude that STX17/syntaxin 17 (Syx17 in Drosophila) has an evolutionarily conserved role in autophagosome fusion with endosomes and lysosomes, acting in one SNARE complex with SNAP29 (ubisnap in Drosophila) and the endosomal/lysosomal VAMP8 (CG1599/Vamp7 in Drosophila). Surprisingly, a third report suggests that STX17 might also contribute to proper phagophore assembly. Although several experiments presented in the two human cell culture studies yielded controversial results, the essential role of STX17 in autophagic flux is now firmly established, both in cultured cells and in an animal model. Based on these data, we propose that genetic inhibition of STX17/Syx17 may be a more specific tool in autophagic flux experiments than currently used drug treatments, which impair all lysosomal degradation routes and also inactivate MTOR (mechanistic target of rapamycin), a major negative regulator of autophagy. Finally, the neuronal dysfunction and locomotion defects observed in Syx17 mutant animals point to the possible contribution of defective autophagosome clearance to various human diseases
A note on linear Sperner families
In an earlier work we described Gr\"obner bases of the ideal of polynomials
over a field, which vanish on the set of characteristic vectors of the complete unifom set family over the ground set . In
particular, it turns out that the standard monomials of the above ideal are
{\em ballot monomials}. We give here a partial extension of the latter fact. We
prove that the lexicographic standard monomials for linear Sperner systems are
also ballot monomials. A set family is a linear Sperner system if the
characteristic vectors satisfy a linear equation ,

The uses of Betula pendula Roth among Hungarian Csángós and Székelys in Transylvania, Romania
The aim of this work is to evaluate the ethnobotanical and ethnopharmacological use of Betula pendula Roth in various regions of Transylvania, in East and Central Romania. Silver birch is an important pioneer species in the country, which has traditionally been used in ethnomedicine, households and various customs. Among the ethnic groups, 55 informants from the Csángós and 68 from the Székelys were interviewed in six villages during field studies occurring between 2007 and 2012. Informants were asked questions about the medical and non-medical applications of B. pendula, focusing on the use of the leaf and woody parts in tools and customs, and on the collection and use of birch sap with its temporal change in local ethnomedicinal knowledge. Differences related to the use of birch among the studied areas were observed. The folk remedies prepared from the sap ("virics") and leaves were documented in local customs, with similar symbolic roles assigned to birch used in other countries. Sap collection was performed during interviews and by manual means. Four special section types on the trunk were presented, and data concerning the nutritional and ethnomedicinal use of the sap was recorded. Both the significance and use of birch sap have drastically decreased in Transylvania due to the appearance of new phytotherapeutical sources and to social changes caused by migration of young people. We found that this phenomenon is gradually leading to the disappearance traditional harvesting techniques, frequency of use and ethnomedicinal knowledge concerning B. pendula in the studied regions
Interaction of the HOPS complex with Syntaxin 17 mediates autophagosome clearance in Drosophila
Homotypic fusion and vacuole protein sorting (HOPS) is a tethering complex required for trafficking to the vacuole/lysosome in yeast. Specific interaction of HOPS with certain SNARE (soluble NSF attachment protein receptor) proteins ensures the fusion of appropriate vesicles. HOPS function is less well characterized in metazoans. We show that all six HOPS subunits (Vps11 [vacuolar protein sorting 11]/CG32350, Vps18/Dor, Vps16A, Vps33A/Car, Vps39/CG7146, and Vps41/Lt) are required for fusion of autophagosomes with lysosomes in Drosophila. Loss of these genes results in large-scale accumulation of autophagosomes and blocks autophagic degradation under basal, starvation-induced, and developmental conditions. We find that HOPS colocalizes and interacts with Syntaxin 17 (Syx17), the recently identified autophagosomal SNARE required for fusion in Drosophila and mammals, suggesting their association is critical during tethering and fusion of autophagosomes with lysosomes. HOPS, but not Syx17, is also required for endocytic down-regulation of Notch and Boss in developing eyes and for proper trafficking to lysosomes and eye pigment granules. We also show that the formation of autophagosomes and their fusion with lysosomes is largely unaffected in null mutants of Vps38/UVRAG (UV radiation resistance associated), a suggested binding partner of HOPS in mammals, while endocytic breakdown and lysosome biogenesis is perturbed. Our results establish the role of HOPS and its likely mechanism of action during autophagy in metazoans
Dog feeding test for assessing the nutritional adequacy of practical diets
The nutritive value of dog foods declared by the manufacturer as nutritionally complete and balanced can be best assessed by feeding trials with dogs. A protocol of a feeding trial has been developed and tested with working dogs fed two different commercial complete and balanced diets for 8 weeks. The parameters used for evaluating the effect of diets were general health status, body and hair coat condition, change of body weight, haematological parameters (white blood cell (WBC) count, red blood cell (RBC) count, haemoglobin, packed cell volume), and biochemical parameters in blood serum (alanine aminotransferase, urea, albumin). The trial protocol proved to be appropriate to monitor the dogs' nutritional status and to reveal differences between diets. This method of evaluation is recommended for use in supporting the nutritional claims (labelling) of dog foods
