    Flavonoids and Pectins

    Pectins and flavonoids are two related groups of important secondary metabolites derived from plants. The interaction between pectins and flavonoids can affect their shelf-life stability, functionality, bioavailability, and bioaccessibility. In this chapter, we will concentrate on the current opinions on the flavonoids to understand how to classify this group of secondary metabolites, what biological and pharmacological activities they possess, and how to biosynthesize them in plants. We will then discuss the general strategies for the derivation of these small secondary compounds. The strategies comprise traditional plant extraction, chemical synthesis, and biosynthesis. We will also discuss the advantages and disadvantages of these three production strategies in the derivation of flavonoids and the future research directions in generating health-beneficial flavonoids using the biosynthetic strategy

    Design and Experiment of a Wheel Precision Seed-Metering Device with Cells for Corn

    In this study, a wheel seed-metering device with cells for corn was built to enhance seed-metering performance and shorten the design cycle. The actual seed-metering process was analyzed and the three main factors influencing seed metering were obtained: rotational speed of the seed-metering device, hole diameter, and hole pitch. With the three factors as the test factors, and the QFI, MISS and MULT index as the test indexes, the multiple quadratic rotation orthogonal combination test was carried out. Using Design-Expert 8.0.6 to analyze the test data, the mathematical model between the test factors and the test index was obtained. The parameter optimization results indicate that optimal performance of the seed-metering device can be achieved under a rotational speed of 30.32 rpm, hole diameter of 13 mm, and hole pitch of 45 mm, qualification rate, miss-seeding rate, and reseeding rate were 94.99%, 1.93%, and 3.08%, respectively. Under the optimal parameter combination, the field test proved that the qualification rate, miss-seeding rate, and reseeding rate were 91.81%, 3.08%, and 5.11%, respectively. The test results were close to the optimization results, which could show the agronomic requirements of the precision sowing of maize

    Converting Redox Signaling to Apoptotic Activities by Stress-Responsive Regulators HSF1 and NRF2 in Fenretinide Treated Cancer Cells

    BACKGROUND: Pharmacological intervention of redox balance in cancer cells often results in oxidative stress-mediated apoptosis, attracting much attention for the development of a new generation of targeted therapy in cancer. However, little is known about mechanisms underlying the conversion from oxidative signaling to downstream activities leading cells to death. METHODOLOGY/PRINCIPAL FINDINGS: We here report a systematic detection of transcriptome changes in response to oxidative signals generated in leukemia cells upon fenretinide treatment, implicating the occurrence of numerous stress-responsive events during the fenretinide induced apoptosis, such as redox response, endoplasmic reticulum stress/unfolded protein response, translational repression and proteasome activation. Moreover, the configuration of these relevant events is primarily orchestrated by stress responsive transcription factors, as typically highlighted by NF-E2-related factor-2 (NRF2) and heat shock factor 1 (HSF1). Several lines of evidence suggest that the coordinated regulation of these transcription factors and thus their downstream genes are involved in converting oxidative signaling into downstream stress-responsive events regulating pro-apoptotic and apoptotic activities at the temporal and spatial levels, typifying oxidative stress-mediated programmed death rather than survival in cancer cells. CONCLUSIONS/SIGNIFICANCE: This study provides a roadmap for understanding oxidative stress-mediated apoptosis in cancer cells, which may be further developed into more sophisticated therapeutic protocols, as implicated by synergistic induction of cell apoptosis using proteasome inhibitors with fenretinide

    Predictive model for diabetic retinopathy under limited medical resources: A multicenter diagnostic study

    BackgroundComprehensive eye examinations for diabetic retinopathy is poorly implemented in medically underserved areas. There is a critical need for a widely available and economical tool to aid patient selection for priority retinal screening. We investigated the possibility of a predictive model for retinopathy identification using simple parameters.MethodsClinical data were retrospectively collected from 4, 159 patients with diabetes admitted to five tertiary hospitals. Independent predictors were identified by univariate analysis and least absolute shrinkage and selection operator (LASSO) regression, and a nomogram was developed based on a multivariate logistic regression model. The validity and clinical practicality of this nomogram were assessed using concordance index (C-index), area under the receiver operating characteristic curve (AUROC), calibration curves, decision curve analysis (DCA), and clinical impact curves (CIC).ResultsThe predictive factors in the multivariate model included the duration of diabetes, history of hypertension, and cardiovascular disease. The three-variable model displayed medium prediction ability with an AUROC of 0.722 (95%CI 0.696-0.748) in the training set, 0.715 (95%CI 0.670-0.754) in the internal set, and 0.703 (95%CI 0.552-0.853) in the external dataset. DCA showed that the threshold probability of DR in diabetic patients was 17-55% according to the nomogram, and CIC also showed that the nomogram could be applied clinically if the risk threshold exceeded 30%. An operation interface on a webpage (https://cqmuxss.shinyapps.io/dr_tjj/) was built to improve the clinical utility of the nomogram.ConclusionsThe predictive model developed based on a minimal amount of clinical data available to diabetic patients with restricted medical resources could help primary healthcare practitioners promptly identify potential retinopathy

Dále se věnuji úkolům, které mi byly zadány s jejich implementací a postupem řešení problémů, které se objevily při vývoji. Závěr práce je věnován zhodnocení získaných zkušeností a dosažených výsledků.Purpose of this bachelor thesis is to describe a professional practice in company HS Interactive s.r.o. Practice was focused on the development of mobile application for the operating system Android. The application is a mobile client for social network MatchToMe. In the introduction I describe reasons that led to the selection of professional practice. Then I describe tasks that I have been awarded with their implementations and process of solution issues that have emerged during development.     Salvianolic Acid B Prevents Bone Loss in Prednisone-Treated Rats through Stimulation of Osteogenesis and Bone Marrow Angiogenesis

    Glucocorticoid (GC) induced osteoporosis (GIO) is caused by the long-term use of GC for treatment of autoimmune and inflammatory diseases. The GC related disruption of bone marrow microcirculation and increased adipogenesis contribute to GIO development. However, neither currently available anti-osteoporosis agent is completely addressed to microcirculation and bone marrow adipogenesis. Salvianolic acid B (Sal B) is a polyphenolic compound from a Chinese herbal medicine, Salvia miltiorrhiza Bunge. The aim of this study was to determine the effects of Sal B on osteoblast bone formation, angiogenesis and adipogenesis-associated GIO by performing marrow adipogenesis and microcirculation dilation and bone histomorphometry analyses. (1) In vivo study: Bone loss in GC treated rats was confirmed by significantly decreased BMD, bone strength, cancellous bone mass and architecture, osteoblast distribution, bone formation, marrow microvessel density and diameter along with down-regulation of marrow BMPs expression and increased adipogenesis. Daily treatment with Sal B (40 mg/kg/d) for 12 weeks in GC male rats prevented GC-induced cancellous bone loss and increased adipogenesis while increasing cancellous bone formation rate with improved local microcirculation by capillary dilation. Treatment with Sal B at a higher dose (80 mg/kg/d) not only prevented GC-induced osteopenia, but also increased cancellous bone mass and thickness, associated with increase of marrow BMPs expression, inhibited adipogenesis and further increased microvessel diameters. (2) In vitro study: In concentration from 10−6 mol/L to 10−7 mol/L, Sal B stimulated bone marrow stromal cell (MSC) differentiation to osteoblast and increased osteoblast activities, decreased GC associated adipogenic differentiation by down-regulation of PPARγ mRNA expression, increased Runx2 mRNA expression without osteoblast inducement, and, furthermore, Sal B decreased Dickkopf-1 and increased β-catenin mRNA expression with or without adipocyte inducement in MSC. We conclude that Sal B prevented bone loss in GC-treated rats through stimulation of osteogenesis, bone marrow angiogenesis and inhibition of adipogenesis

    Does Person–Organization Value Fit Affect Job Performance of Post-90s Employees in China? A Moderated Mediation Model Based on Self-Determination Theory

    Nowadays, post-90s employees are becoming the main cohort within organizations in China. They are considered to have strong self-awareness, weak collective consciousness, and low work motivation, making it difficult for managers to improve their job performance. After reviewing the relevant literature, we found that person–organization (PO) value fit is positively related to job performance, but there is a limitation in explaining the psychological characteristics of post-90s employees. This study aims to explore the mechanism that how PO value fit impacts the job performance of post-90s employees in China. From the view of self-determination theory (SDT), we selected happiness as a mediating variable and love of money (LOM) as a moderating variable. Based on valid data collected from 919 employees from all walks of life in China, we utilized both linear regression analysis and a bootstrapping approach to verify our propositions. The results revealed a positive relationship between PO value fit and job performance through happiness. The moderated mediation analysis further indicated that the mediated path bonding happiness with job performance was weaker for post-90s employees with higher levels of LOM. The present study offers a nuanced interpretation of how PO value fit affects the job performance of post-90s employees in China and contributes to providing suggestions for improving the sustainability of organizations
