    Halogen occultation experiment (HALOE) optical witness-plate program

    An optical witness plate program was implemented to monitor buildup of molecular contamination in the clean room during the assembly and testing of the Halogen Occulation Experiment (HALOE) instrument. Travel plates to monitor molecular contamination when the instrument is not in the clean room are also measured. The instrument technique is high-resolution transmission spectroscopy in the 3 micron spectral region using a Fourier transform spectrometer. Witness specimens of low index of refraction, infrared transmitting material are used for contaminant monitoring and for spectral signature analysis. Spectral signatures of possible molecular contamination are presented. No condensible volatile material contamination of HALOE optical witness specimens have yet been found

    Trust in Biotechnology Risk Managers: Insights from the United Kingdom, 1996-2002

    The mid to late 1990s saw a series of negative media events in the United Kingdom (UK) related to biotechnology. According to the trust asymmetry hypothesis, such events ought to cause public trust in risk managers of biotechnology to fall quickly but rise slowly. We present evidence from the Eurobarometer surveys that from 1996 to 1999 public trust in the UK declined, but it increased sharply between 1999 and 2002. We seek to explain this apparent contradiction to the asymmetry hypothesis. We use canonical discriminant analysis of public trust to show that whether people trust or distrust risk managers of biotechnology depends significantly on the amount of knowledge people have about science.     Abundance, distribution, and habitat of leatherback turtles (Dermochelys coriacea) off California, 1990−2003

    Leatherback turtles (Dermochelys coriacea) are regularly seen off the U.S. West Coast, where they forage on jellyfish (Scyphomedusae) during summer and fall. Aerial line-transect surveys were conducted in neritic waters (<92 m depth) off central and northern California during 1990−2003, providing the first foraging population estimates for Pacific leatherback turtles. Males and females of about 1.1 to 2.1 m length were observed. Estimated abundance was linked to the Northern Oscillation Index and ranged from 12 (coefficient of variation [CV] =0.75) in 1995 to 379 (CV= 0.23) in 1990, averaging 178 (CV= 0.15). Greatest densities were found off central California, where oceanographic retention areas or upwelling shadows created favorable habitat for leatherback turtle prey. Results from independent telemetry studies have linked leatherback turtles off the U.S. West Coast to one of the two largest remaining Pacific breeding populations, at Jamursba Medi, Indonesia. Nearshore waters off California thus represent an important foraging region for the critically endangered Pacific leatherback turtle

    Minimally Invasive Optical Biopsy for Oximetry

    The study of localised oxygen saturation in blood vessels can shed light on the etiology and progression of many diseases with which hypoxia is associated. For example, hypoxia in the tendon has been linked to early stages of rheumatoid arthritis, an auto-immune inflammatory disease. Vascular oximetry of deep tissue presents significant challenges as vessels are not optically accessible. In this paper, we present a novel multispectral imaging technique for vascular oximetry, and recent developments made towards its adaptation for minimally invasive imaging. We present proof-of-concept of the system and illumination scheme as well as the analysis technique. We present results of a validation study performed in vivo on mice with acutely inflamed tendons. Adaptation of the technique for minimally invasive microendoscopy is also presented, along with preliminary results of minimally invasive ex vivo vascular oximetry

    Development of polyisocyanurate pour foam formulation for space shuttle external tank thermal protection system

    Four commercially available polyisocyanurate polyurethane spray-foam insulation formulations are used to coat the external tank of the space shuttle. There are several problems associated with these formulations. For example, some do not perform well as pourable closeout/repair systems. Some do not perform well at cryogenic temperatures (poor adhesion to aluminum at liquid nitrogen temperatures). Their thermal stability at elevated temperatures is not adequate. A major defect in all the systems is the lack of detailed chemical information. The formulations are simply supplied to NASA and Martin Marietta, the primary contractor, as components; Part A (isocyanate) and Part B (poly(s) and additives). Because of the lack of chemical information the performance behavior data for the current system, NASA sought the development of a non-proprietary room temperature curable foam insulation. Requirements for the developed system were that it should exhibit equal or better thermal stability both at elevated and cryogenic temperatures with better adhesion to aluminum as compared to the current system.     Relationship between fish size and otolith length for 63 species of fishes from the Eastern North Pacific Ocean

    Otoliths commonly are used to determine the taxon, age, and size of fishes. This information is useful for population management, predator-prey studies, and archaeological research. The relationship between the length of a fish and the length of its otoliths remains unknown for many species of marine fishes in the Pacific Ocean. Therefore, the relationships between fish length and fish weight, and between otolith length and fish length, were developed for 63 species of fishes caught in the eastern North Pacific Ocean. We also summarized similar relationships for 46 eastern North Pacific fish species reported in the literature. The relationship between fish length and otolith length was linear, and most of the variability was explained by a simple least-squares regression (r 2 > 0.700 for 45 of 63 species). The relationship between otolith length and fish length was not significantly different between left and right otoliths for all but one fish species. Images of otoliths from 77 taxa are included to assist in the identification of species. (PDF file contains 38 pages.

    Visualisation and Quantification of Morphogen Gradient Formation in the Zebrafish

    During embryonic development, signalling molecules known as morphogens act in a concentration-dependent manner to provide positional information to responding tissues. In the early zebrafish embryo, graded signalling by members of the nodal family induces the formation of mesoderm and endoderm, thereby patterning the embryo into three germ layers. Nodal signalling has also been implicated in the establishment of the dorso-ventral axis of the embryo. Although one can infer the existence of nodal gradients by comparing gene expression patterns in wild-type embryos and embryos in which nodal signalling is diminished or augmented, real understanding can only come from directly observing the gradients. One approach is to determine local ligand concentrations in the embryo, but this is technically challenging, and the presence of inhibitors might cause the effective concentration of a ligand to differ from its actual concentration. We have therefore taken two approaches to visualise a direct response to nodal signalling. In the first, we have used transgenic embryos to study the nuclear accumulation of a Smad2-Venus fusion protein, and in the second we have used bimolecular fluorescence complementation to visualise the formation of a complex between Smad2 and Smad4. This has allowed us to visualise, in living embryos, the formation of a graded distribution of nodal signalling activity. We have quantified the formation of the gradient in time and space, and our results not only confirm that nodal signalling patterns the embryo into three germ layers, but also shed light on its role in patterning the dorso-ventral axis and highlight unexpected complexities of mesodermal patterning

    Overlapping functionality of the Pht proteins in zinc homeostasis of streptococcus pneumoniae

    Streptococcus pneumoniae is a globally significant pathogen that causes a range of diseases, including pneumonia, sepsis, meningitis, and otitis media. Its ability to cause disease depends upon the acquisition of nutrients from its environment, including transition metal ions such as zinc. The pneumococcus employs a number of surface proteins to achieve this, among which are four highly similar polyhistidine triad (Pht) proteins. It has previously been established that these proteins collectively aid in the delivery of zinc to the ABC transporter substrate-binding protein AdcAII. Here we have investigated the contribution of each individual Pht protein to pneumococcal zinc homeostasis by analyzing mutant strains expressing only one of the four pht genes. Under conditions of low zinc availability, each of these mutants showed superior growth and zinc accumulation profiles relative to a mutant strain lacking all four genes, indicating that any of the four Pht proteins are able to facilitate delivery of zinc to AdcAII. However, optimal growth and zinc accumulation in vitro and pneumococcal survival and proliferation in vivo required production of all four Pht proteins, indicating that, despite their overlapping functionality, the proteins are not dispensable without incurring a fitness cost. We also show that surface-attached forms of the Pht proteins are required for zinc recruitment and that they do not contribute to defense against extracellular zinc stress
