Altering Social Cue Perception Impacts Honey Bee Aggression With Minimal Impacts on Aggression-Related Brain Gene Expression
Gene expression changes resulting from social interactions may give rise to long term behavioral change, or simply reflect the activity of neural circuitry associated with behavioral expression. In honey bees, social cues broadly modulate aggressive behavior and brain gene expression. Previous studies suggest that expression changes are limited to contexts in which social cues give rise to stable, relatively long-term changes in behavior. Here we use a traditional beekeeping approach that inhibits aggression, smoke exposure, to deprive individuals of aggression-inducing olfactory cues and evaluate whether behavioral changes occur in absence of expression variation in a set of four biomarker genes (drat, cyp6g1/2, GB53860, inos) associated with aggression in previous studies. We also evaluate two markers of a brain hypoxic response (hif1α, hsf) to determine whether smoke induces molecular changes at all. We find that bees with blocked sensory perception as a result of smoke exposure show a strong, temporary inhibition of aggression relative to bees allowed to perceive normal social cues. However, blocking sensory perception had minimal impacts on aggression-relevant gene expression, althought it did induce a hypoxic molecular response in the brain. Results suggest that certain genes differentiate social cue-induced changes in aggression from long-term modulation of this phenotype
Mechanobiology of Antimicrobial Resistant Escherichia coli and Listeria innocua
A majority of antibiotic-resistant bacterial infections in the United States are associated with biofilms. Nanoscale biophysical measures are increasingly revealing that adhesive and viscoelastic properties of bacteria play essential roles across multiple stages of biofilm development. Atomic Force Microscopy (AFM) applied to strains with variation in antimicrobial resistance enables new opportunities for investigating the function of adhesive forces (stickiness) in biofilm formation. AFM force spectroscopy analysis of a field strain of Listeria innocua and the strain Escherichia coli K-12 MG1655 revealed differing adhesive forces between antimicrobial resistant and nonresistant strains. Significant increases in stickiness were found at the nanonewton level for strains of Listeria innocua and Escherichia coli in association with benzalkonium chloride and silver nanoparticle resistance respectively. This advancement in the usage of AFM provides for a fast and reliable avenue for analyzing antimicrobial resistant cells and the molecular dynamics of biofilm formation as a protective mechanism
Interferometric Observations of the Nuclear Region of Arp220 at Submillimeter Wavelengths
We report the first submillimeter interferometric observations of an
ultraluminous infrared galaxy. We observed Arp220 in the CO J=3-2 line and
342GHz continuum with the single baseline CSO-JCMT interferometer consisting of
the Caltech Submillimeter Observatory (CSO) and the James Clerk Maxwell
Telescope (JCMT). Models were fit to the measured visibilities to constrain the
structure of the source. The morphologies of the CO J=3-2 line and 342GHz
continuum emission are similar to those seen in published maps at 230 and
110GHz. We clearly detect a binary source separated by about 1 arcsec in the
east-west direction in the 342GHz continuum. The CO J=3-2 visibility
amplitudes, however, indicate a more complicated structure, with evidence for a
compact binary at some velocities and rather more extended structure at others.
Less than 30% of the total CO J=3-2 emission is detected by the interferometer,
which implies the presence of significant quantities of extended gas. We also
obtained single-dish CO J=2-1, CO J=3-2 and HCN J=4-3 spectra. The HCN J=4-3
spectrum, unlike the CO spectra, is dominated by a single redshifted peak. The
HCN J=4-3/CO J=3-2, HCN J=4-3/HCN J=1-0 and CO J=3-2/2-1 line ratios are larger
in the redshifted (eastern) source, which suggests that the two sources may
have different physical conditions. This result might be explained by the
presence of an intense starburst that has begun to deplete or disperse the
densest gas in the western source, while the eastern source harbors undispersed

Barriers to the management of Heart Failure in Ontario Long-Term Care Homes: an Interprofessional Care perspective
Background: With population aging, the prevalence of heart failure (HF) is risingin long-term care (LTC) homes. Given this burden, there is an urgent need to establish effective HF management programs.Methods and Findings: To understand what barriers would need to be addressed to develop such a program, we conducted a series of consultations among various LTC staff, as well as residents and their family caregivers. This article uses data obtained from the consultations to describe the interprofessional (IP) barriers that exist among the various LTC staff roles. Consultation methods included a Delphi survey followed by focus group interviews of LTC staff, and then personal interviews with LTC residents with HF and their family caregivers. Data were interpreted using an IP care framework in which interpersonal relationships among LTC staff provide the most direct influence on collaborative resident-centred practice, within the broader context of conditions within the LTC home, which in turn are housed in the broader context of systemic determinants.Conclusion: Across all data sets, the most consistently mentioned determinant was communication between the resident and the healthcare team, between different healthcare providers, between shifts, between medical specialists, and between the long-term care home and the hospital
A Comparison of Methods for a Priori Bias Correction in Soil Moisture Data Assimilation
Data assimilation is being increasingly used to merge remotely sensed land surface variables such as soil moisture, snow and skin temperature with estimates from land models. Its success, however, depends on unbiased model predictions and unbiased observations. Here, a suite of continental-scale, synthetic soil moisture assimilation experiments is used to compare two approaches that address typical biases in soil moisture prior to data assimilation: (i) parameter estimation to calibrate the land model to the climatology of the soil moisture observations, and (ii) scaling of the observations to the model s soil moisture climatology. To enable this research, an optimization infrastructure was added to the NASA Land Information System (LIS) that includes gradient-based optimization methods and global, heuristic search algorithms. The land model calibration eliminates the bias but does not necessarily result in more realistic model parameters. Nevertheless, the experiments confirm that model calibration yields assimilation estimates of surface and root zone soil moisture that are as skillful as those obtained through scaling of the observations to the model s climatology. Analysis of innovation diagnostics underlines the importance of addressing bias in soil moisture assimilation and confirms that both approaches adequately address the issue
Development of a new learning methodology for discrete event simulation by reutilising previous software experience
New discrete event simulation software available to industry has significantly reduced the modelling efforts of complex manufacturing problems. These tools enable analysts to assess the viability of potential solutions that better conform to previously defined requirements. Thus, analysts must be conversant in new technologies applications to deliver top quality solutions to the enterprises analysed.
Traditional approaches of learning a new technology tend to isolate previous knowledge the analyst possesses in similar application fields and concentrate on features and strengths of the particular application under study. A new approach is therefore needed to capitalise on previous experience an analyst might have, enabling reduction of learning a new technological application by minimising the learning curve effort spent learning the technology, and increasing focus on quantitative and qualitative analysis. Rapid Evolution of Silver Nanoparticle Resistance in Escherichia Coli
The recent exponential increase in the use of engineered nanoparticles (eNPs) means both greater intentional and unintentional exposure of eNPs to microbes. Intentional use includes the use of eNPs as biocides. Unintentional exposure results from the fact that eNPs are included in a variety of commercial products (paints, sunscreens, cosmetics). Many of these eNPs are composed of heavy metals or metal oxides such as silver, gold, zinc, titanium dioxide, and zinc oxide. It is thought that since metallic/metallic oxide NPs impact so many aspects of bacterial physiology that it will difficult for bacteria to evolve resistance to them. This study utilized laboratory experimental evolution to evolve silver nanoparticle (AgNP) resistance in the bacterium Escherichia coli (K-12 MG1655), a bacterium that does not harbor any known silver resistance elements. After 225 generations of exposure to the AgNP environment, the treatment populations demonstrated greater fitness vs. control strains as measured by optical density (OD) and colony forming units (CFU) in the presence of varying concentrations of 10 nm citrate-coated silver nanoparticles (AgNP) or silver nitrate (AgNO3). Genomic analysis shows that changes associated with AgNP resistance were already accumulating within the treatment populations by generation 100, and by generation 200 three mutations had swept to high frequency in the AgNP resistance stocks. This study indicates that despite previous claims to the contrary bacteria can easily evolve resistance to AgNPs, and this occurs by relatively simple genomic changes. These results indicate that care should be taken with regards to the use of eNPs as biocides as well as with regards to unintentional exposure of microbial communities to eNPs in waste products
