Reduction in the risk of human breast cancer by selective cyclooxygenase-2 (COX-2) inhibitors
BACKGROUND: Epidemiologic and laboratory investigations suggest that nonsteroidal anti-inflammatory drugs (NSAIDs) have chemopreventive effects against breast cancer due to their activity against cyclooxygenase-2 (COX-2), the rate-limiting enzyme of the prostaglandin cascade. METHODS: We conducted a case control study of breast cancer designed to compare effects of selective and non-selective COX-2 inhibitors. A total of 323 incident breast cancer patients were ascertained from the James Cancer Hospital, Columbus, Ohio, during 2003–2004 and compared with 649 cancer free controls matched to the cases at a 2:1 ratio on age, race, and county of residence. Data on the past and current use of prescription and over the counter medications and breast cancer risk factors were ascertained using a standardized risk factor questionnaire. Effects of COX-2 inhibiting agents were quantified by calculating odds ratios (OR) and 95% confidence intervals. RESULTS: Results showed significant risk reductions for selective COX-2 inhibitors as a group (OR = 0.29, 95% CI = 0.14–0.59), regular aspirin (OR = 0.49, 95% CI = 0.26–0.94), and ibuprofen or naproxen (0.36, 95% CI = 0.18–0.72). Acetaminophen, a compound with negligible COX-2 activity and low dose aspirin (81 mg) produced no significant change in the risk of breast cancer. CONCLUSION: Selective COX-2 inhibitors (celecoxib and rofecoxib) were only recently approved for use in 1999, and rofecoxib (Vioxx) was withdrawn from the marketplace in 2004. Nevertheless, even in the short window of exposure to these compounds, the selective COX-2 inhibitors produced a significant (71%) reduction in the risk of breast cancer, underscoring their strong potential for breast cancer chemoprevention
A comparison of two-coloured filter systems for treating visual reading difficulties
Copyright @ 2013 Informa UK Ltd.Purpose: Visual disturbances that make it difficult to read text are often termed “visual stress”. Coloured filters in spectacles may help some children overcome reading problems that are often caused by visual stress. It has been suggested that for optimal effect each child requires an individually prescribed colour for each eye, as determined in systems such as the “Harris Foundation” coloured filters. Alternatively, it has been argued that only blue or yellow filters, as used in the “Dyslexia Research Trust” (DRT) filter system, are necessary to affect the underlying physiology. Method: A randomised, double blind trial with 73 delayed readers, was undertaken to compare changes in reading and spelling as well as irregular and non-word reading skills after 3 months of wearing either the Harris or the DRT filters. Results: Reading improved significantly after wearing either type of filter (t = −8.4, p < 0.01), with 40% of the children improving their reading age by 6 months or more during the 3 month trial. However, spelling ability (t = 2.1, p = 0.05) and non-word reading (f = 4.7, p < 0.05) improved significantly more with the DRT than with the Harris filters. Conclusion: Education and rehabilitation professionals should therefore, consider coloured filters as an effective intervention for delayed readers experiencing visual stress
Phosphorus sensitivity in species of Proteaceae (Protea obtusifolia, Leucadendron coniferum and Leucadendron salignum) from different soil habitats : possible candidates for growth on former agricultural soils high in P
The phosphorus (P) uptake capacities and tolerance to high P of three Proteaceae species from acid (Leucadenron coniferum, Leucadendron salignum) and alkaline (Protea obtusifolia, Leucadedndron meridianum) soils in the Cape Floristic Region (CFR) were compared. These species were also investigated as potential candidates for growth on nutrient enriched, post-agricultural soils. In parallel, two crop plant species (Lupinus albus, with cluster roots; and L. angustifolius, without cluster roots) were also compared. The cluster-root forming species (L. albus) was more sensitive to higher P levels and showed toxicity in terms of photosynthesis, efficiency of PSII, transpiration and stomatal conductance. Growth, gaseous exchange and efficiency of photosystem II (as a measure of stress) were determined at 1, 10 and 100 μg P g⁻¹ sand for both lupins and Proteaceae. Kinetics of P uptake (in hydroponics) was determined at a range ( 4 to 2000 μM P) of P concentrations. Phosphorus sensitivity was relatively high in Ld. coniferum as evident from reduced growth, net photosynthetic rate, stomata} conductance and transpiration rates. Protea obtusifolia showed the least sensitivity to increased phosphorus concentrations under acidic and native alkaline conditions. The inability to regulate P uptake rates at elevated phosphorus concentrations and excessive P accumulation were major determinants of P sensitivity. The relatively higher P sensitivity of Lupinus albus compared to L. angustifolius is likely related to the efficient P uptake by cluster roots in the former
The estrogenic activity of phthalate esters in vitro
A large number of phthalate esters were screened for estrogenic activity using a recombinant yeast screen. a selection of these was also tested for mitogenic effect on estrogen-responsive human breast cancer cells. A small number of the commercially available phthalates tested showed extremely weak estrogenic activity. The relative potencies of these descended in the order butyl benzyl phthalate (BBP) > dibutyl phthalate (DBP) > diisobutyl phthalate (DIBP) > diethyl phthalate (DEP) > diisiononyl phthalate (DINP). Potencies ranged from approximately 1 x 10(6) to 5 x 10(7) times less than 17beta-estradiol. The phthalates that were estrogenic in the yeast screen were also mitogenic on the human breast cancer cells. Di(2-ethylhexyl) phthalate (DEHP) showed no estrogenic activity in these in vitro assays. A number of metabolites were tested, including mono-butyl phthalate, mono-benzyl phthalate, mono-ethylhexyl phthalate, mon-n-octyl phthalate; all were wound to be inactive. One of the phthalates, ditridecyl phthalate (DTDP), produced inconsistent results; one sample was weakly estrogenic, whereas another, obtained from a different source, was inactive. analysis by gel chromatography-mass spectometry showed that the preparation exhibiting estrogenic activity contained 0.5% of the ortho-isomer of bisphenol A. It is likely that the presence of this antioxidant in the phthalate standard was responsible for the generation of a dose-response curve--which was not observed with an alternative sample that had not been supplemented with o,p'-bisphenol A--in the yeast screen; hence, DTDP is probably not weakly estrogenic. The activities of simple mixtures of BBP, DBP, and 17beta-estradiol were assessed in the yeast screen. No synergism was observed, although the activities of the mixtures were approximately additive. In summary, a small number of phthalates are weakly estrogenic in vitro. No data has yet been published on whether these are also estrogenic in vitro. No data has yet been published on whether these are also estrogenic in vivo; this will require tests using different classes of vertebrates and different routes of exposure
Promoter methylation regulates cyclooxygenase expression in breast cancer
INTRODUCTION: Overexpression of cyclooxygenase (COX-2) is commonly observed in human cancers. In a murine model of metastatic breast cancer, we observed that COX-2 expression and enzyme activity were associated with enhanced tumorigenic and metastatic potential. In contrast to the high COX-2 expression in metastatic tumors, transplantation of poorly tumorigenic tumor cell lines to syngeneic mice results in less COX-2 expression and less COX-2 activity in vivo. Aberrant CpG island methylation, and subsequent silencing of the COX-2 promoter, has been observed in human cancer cell lines and in some human tumors of the gastrointestinal tract. METHODS: Using bisulfite modification and a methylation-specific PCR, we examined the methylation status of the COX-2 promoter in a series of four closely-related murine mammary tumors differing in COX-2 expression and metastatic potential. RESULTS: We showed that line 410, which does not express COX-2 in vivo, exhibited evidence of promoter methylation. Interestingly, the metastatic counterpart of this cell (line 410.4) displayed only the unmethylated COX-2 promoter, as did two additional cell lines (lines 66.1 and 67). The methylation patterns observed in vitro were maintained when these murine mammary tumor lines were transplanted to syngeneic mice. Treatment with the DNA demethylating agent 5-aza-deoxycytidine increased COX-2 mRNA, increased protein and increased enzyme activity (prostaglandin synthesis). CONCLUSIONS: These results indicate that COX-2 promoter methylation may be one mechanism by which tumor cells regulate COX-2 expression. Upregulation of COX-2 expression in closely related metastatic lesions versus nonmetastatic lesions may represent a shift towards the unmethylated phenotype
Plate-impact loading of cellular structures formed by selective laser melting
Studies of the shock loading of porous material have the potential to improve our understanding of factors such as density, crush strength and pore size on energy absorbing capability. Porous components were manufactured using Selective Laser Melting (SLM) in which layers of metal powder are fused together to create a structure specified by an electronic file. Samples have been manufactured in which a lattice is formed by an array of intersecting rods angled at 45 degrees to the surface of a 6 mm thick x ~100 mm diameter disc. The cell size is 1 mm3 and the density is 44.6% of solid. A 100 mm gas gun has been used to impact the porous samples onto solid stainless steel plates. Het-V laser interferometry was used to measure the velocity vs. time profile of the transmitted shock. The experimental results were compared with three dimensional computer predictions. It was found that the simulations reproduced the main features of the experimental record but tended to underestimate the measured velocities, suggesting that the codes were not calculating the energy absorbed by the lattice correctly. Additional calculations were performed with the aim of building a picture of the processes of energy absorption in cellular materials whose structure is varied systematically. These supporting studies suggest a possible explanation for the observed computational/experimental discrepancies. © 2012 British Crown
Establishing comprehensive oral assessments for children with safeguarding concerns.
The dental profession is well placed to contribute important information in child protection cases but no previous research has been reported that assesses the volume or impact of this information. Comprehensive oral assessment clinics were introduced and established as an integral part of comprehensive medical assessments for children with welfare concerns in Greater Glasgow and Clyde. An assessment protocol and standardised paperwork for comprehensive oral assessments were developed to enhance information sharing and patient access to appropriate care. Building development and roads: implications for the distribution of stone curlews across the Brecks
Background: Substantial new housing and infrastructure development planned within England has the potential to conflict with the nature conservation interests of protected sites. The Breckland area of eastern England (the Brecks) is designated as a Special Protection Area for a number of bird species, including the stone curlew (for which it holds more than 60% of the UK total population). We explore the effect of buildings and roads on the spatial distribution of stone curlew nests across the Brecks in order to inform strategic development plans to avoid adverse effects on such European protected sites. Methodology: Using data across all years (and subsets of years) over the period 1988 – 2006 but restricted to habitat areas of arable land with suitable soils, we assessed nest density in relation to the distances to nearest settlements and to major roads. Measures of the local density of nearby buildings, roads and traffic levels were assessed using normal kernel distance-weighting functions. Quasi-Poisson generalised linear mixed models allowing for spatial auto-correlation were fitted. Results: Significantly lower densities of stone curlew nests were found at distances up to 1500m from settlements, and distances up to 1000m or more from major (trunk) roads. The best fitting models involved optimally distance-weighted variables for the extent of nearby buildings and the trunk road traffic levels. Significance : The results and predictions from this study of past data suggests there is cause for concern that future housing development and associated road infrastructure within the Breckland area could have negative impacts on the nesting stone curlew population. Given the strict legal protection afforded to the SPA the planning and conservation bodies have subsequently agreed precautionary restrictions on building development within the distances identified and used the modelling predictions to agree mitigation measures for proposed trunk road developments

Impact response of additively manufactured metallic hybrid lattice materials
Additive manufacturing (AM) enables the design of new cellular materials for blast and impact mitigation by allowing novel material-geometry combinations to be realised and examined at a laboratory scale. However, design of these materials requires an understanding of the relationship between the AM process and material properties at different length scales: from the microstructure to geometric feature rendition to overall dynamic performance. To date, there remain significant uncertainties about both the potential benefits and pitfalls of using AM to design and optimise cellular materials for dynamic energy absorbing applications. This experimental investigation focuses on the out-of-plane compression of stainless steel cellular materials fabricated using selective laser melting (SLM), and makes two specific contributions. First, we demonstrate how the AM process itself influences the characteristics of these cellular materials across a range of length scales, and, crucially, how this influences the dynamic deformation. Secondly, we demonstrate how an AM route can be used to add geometric complexity to the cell structure, creating a versatile basis for future geometry optimisation. Starting with an AM square honeycomb (the reference case), we add porosity to the walls by replacing them with a lattice truss, while maintaining the same relative density. This geometry hybridisation is an approach uniquely suited to this manufacturing route. It is found that the hybrid lattice-walled honeycomb geometry significantly outperforms previously reported AM lattices in terms of specific strength, specific energy absorption, and energy absorption efficiency. It is also found that the hybrid geometry outperforms the benchmark metallic square honeycomb in terms of energy absorption efficiency in the intermediate impact velocity regime (i.e. between quasi-static loading and loading rates at which wave propagation effects begin to become pronounced), a regime in which the collapse is dominated by dynamic buckling effects.The authors are grateful to AWE for financial support
