    Comparing Wearer DNA Sample Collection Methods for the Recovery of Single Source Profiles

    Wearer DNA is the deposit of epithelial cells on clothing worn by an individual. Detection of the last individual to handle or wear an item is often an important and desirable determination in forensic science. The most commonly used collection methods for wearer DNA include swabbing and scraping. These often result in mixture profiles. Recently, adhesives have been introduced as a possible reliable method for the collection of biological evidence. The goal of the research was to compare the current collection methods of swabbing and scraping with a gel film called Gel-Pak ‘0’ which shares similar properties with adhesives. Gel-Pak ‘0’ has been previously studied in comparison to other adhesives for the collection of epithelial cells, and was shown to recover the top layer of loose particulate. This particulate had a tendency to be deposited by the individual who last came in contact with an item. Therefore, in comparison to the other two collection methods, Gel-Pak ‘0’ was hypothesized to recover single source profiles on clothing items from the most recent wearer. DNA analysis was performed on samples collected by the three methods from various clothing items including baseball hats, t-shirts, sweatpants, socks, and other items commonly submitted to crime labs for DNA analysis. The habitual wearer and the second/last wearer wore each item for a predetermined amount of time. The results of the research showed that Gel-Pak ‘0’ recovered a similar number of CODIS (local and national) eligible profiles as swabbing. However, coupled with the fact that it is time consuming, costly, and cannot be used on all surfaces, Gel-Pak ‘0’ was determined to not make for an effective collection method of the most recent wearer’s DNA. Therefore, Gel-Pak ‘0’ will not be considered for casework. Although Gel-Pak ‘0’ will not be further used, the results did reveal some trends that may shed light on how DNA analysts may approach wearer DNA cases. Swabbing had a tendency to yield smaller amounts of DNA in comparison to scraping, but obtain DNA from the last wearer of the piece of clothing more effectively than the other two methods. Scraping had a tendency to yield greater quantities of DNA, recovering more DNA from the habitual wearer due to its invasive nature. Revealing individuals who last wore an item can be of great importance in forensic science, and therefore, further research with various adhesives and gel films could be vital for solving forensic investigations

    Off-farm Income and Investments in Farm Assets: A Double Hurdle Approach

    The farm household structure is a complex set of inter-relationships between and among a variety of internal and external factors involving consumption, investment, and income-earning activities. In this paper we use ARMS data to explore the contribution of off-farm income to the viability of the farm business. We focus on the link between off-farm income and farm investment and whether off-farm income drives on-farm investment. The results indicate the importance of farm characteristics such as type, size, and location on the probability of investment but lead us to reject the hypothesis that off farm income is driving farm investment. Further research will be needed to further unweave some of the complex relationships involved in the farm household structure.     The Impacts of Wrist Wrap Type and Sex on Bench Press Muscular Strength and Power

    While wrist wraps have become increasingly prevalent in both competitive and recreational demographics, their posited ability to augment bench press performance by enhanced wrist stability still remains unclear. PURPOSE: To determine the effect of varying wrap styles on bench-specific muscular strength and associated power, as well as quantitative and subjective differences between sexes. METHODS: Eighteen resistance trained males and females (9M/9F; 24±4y; 176±33cm; 80±15kg) visited the laboratory on three separate occasions in randomized, crossover, and counterbalanced design to sport either a flexible wrist wrap (FW), stiff wrap (SW), or a no wrap control (NW) condition. All participants underwent a bench press one-repetition maximum (1RM) test and linear position transducer-derived peak power and velocity assessments. Furthermore, subjective stability (SS) and discomfort (SD) were determined promptly following 1RM attempts. Bench press performance and sex-collapsed subjective variables were analyzed using a two-way (condition x sex) mixed model ANOVA with repeated measures and a nonparametric Friedman’s ANOVA, respectively. Both analyses were performed at a p\u3c.05 significance level. RESULTS: Analyses failed to detect any main condition or interaction effects for bench press 1RM, however, a statistically significant main sex effect was observed (p\u3c.001; ηp2=.597) favoring males relative to females (p\u3c.001; 114±22kg vs 68±16kg). Both peak power and velocity failed to reveal any significant main condition or sex effects, nor any interactions. Nonparametric assessments further revealed significant wrist wrap condition effects for both SS (p\u3c.001; Kendall’s W=.628) and SD (p\u3c.001; Kendall’s W=.935), whereby NW was statistically more comfortable (p\u3c.001) than either wrap condition, without any difference between DW and SW (p\u3e.05). CONCLUSION: Although wrist wraps did not significantly alter bench press-specific strength and power, participants nonetheless perceived wrist wraps as subjectively more stable irrespective of increased discomfort.     Influence of Acute Turkesterone Dosing on Resting Metabolic Rate and Substrate Utilization in Recreationally-active Males

    Turkesterone is a relatively novel phytoecdysteroid compound that has become increasingly popular amongst recreationally active adults seeking to improve body composition. Although many of the these hypothetical benefits arose from prior rodent data demonstrating enhanced substrate utilization, no data presently exist amongst humans in this regard. PURPOSE: to determine the effect of multiple turkesterone doses on both resting metabolic rate (RMR) and substrate utilization in a healthy human population. METHODS: Eleven recreationally active males (23.3±2.2y) visited the laboratory on three occasions separated by at least seven days and were randomized in single-blind, placebo-controlled, and counter-balanced crossover fashion to either 2000mg cellulose placebo (PLA), 1000mg turkesterone + 1000mg placebo, (1000T) or 2000mg (2000T) turkesterone. RMR and respiratory exchange ratio were assessed using a metabolic cart for 20 minutes prior to supplement provision (i.e. baseline [PRE)), as well as 60-minutes (POST60M), 120-minutes (POST120M), and 180-minutes (POST180M) post-acute supplementation timepoints at each visit. RMR, as well as both carbohydrate (CHO) and Fat (FAT) oxidation were analyzed using a two-way (condition [PLA, 1000T, 2000T] x time [PRE, POST60M, POST120M, POST180M) ANOVA with repeated measures at a significance level of pRESULTS: Analyses failed to reveal any significant condition, time, nor interaction effects for RMR, nor CHO or FAT oxidation (p\u3e0.05). Nonetheless, both 1000T (2.7%, 5.6%, and 7.8%) and 2000T (0.7%, 4.2%, and 3.6%) increased mean RMR above baseline at POST60M, POST120M, and POST180M timepoints, respectively. Conversely, PLA decreased mean RMR by 0.9% and 0.7% at POST60M and POST120M, respectively. Incidentally, the 1000T condition displayed increased mean FAT oxidation by 1.85, 5.34, and 7.96% at the POST60M, POST120M, and POST180M timepoints, respectively, and when compared to the consistent decreases observed with both PLA and 2000T. CONCLUSION: Although these data fail to display a significant turkesterone-mediated enhancement in the investigated metabolic parameters, there were interesting mean differences that should be further explored to determine any longitudinal and/or exercise-dependent permissive impacts on RMR and substrate utilization

    Tracking aftershock sequences using empirical matched field processing

    Extensive aftershock sequences present a significant problem to seismological data centres attempting to produce near real-time comprehensive seismic event bulletins. An elevated number of events to process and poorer performance of automatic phase association algorithms can lead to large delays in processing and a greatly increased human workload. Global monitoring is often performed using seismic array stations at considerable distances from the events involved. Empirical matched field processing (EMFP) is a narrow-frequency band array signal processing technique that recognizes the inter-sensor phase and amplitude relations associated with wavefronts approaching a sensor array from a given direction. We demonstrate that EMFP, using a template obtained from the first P arrival from the main shock alone, can efficiently detect and identify P arrivals on that array from subsequent events in the aftershock zone with exceptionally few false alarms (signals from other sources). The empirical wavefield template encodes all the narrow-band phase and amplitude relations observed for the main shock signal. These relations are also often robust and repeatable characteristics of signals from nearby events. The EMFP detection statistic compares the phase and amplitude relations at a given time in the incoming data stream with those for the template and is sensitive to very short-duration signals with the required characteristics. Significant deviations from the plane-wavefront model that typically degrade the performance of standard beamforming techniques can enhance signal characterization using EMFP. Waveform correlation techniques typically perform poorly for aftershocks from large earthquakes due to the distances between hypocentres and the wide range of event magnitudes and source mechanisms. EMFP on remote seismic arrays mitigates these difficulties; the narrow-band nature of the procedure makes arrival identification less sensitive to the signals’ temporal form and spectral content. The empirical steering vectors derived for the main shock P arrival can reduce the frequency dependency of the slowness vector estimates. This property helps us to automatically screen out arrivals from outside of the aftershock zone. Standard array processing pipelines could be enhanced by including both plane-wave and empirical matched field steering vectors.     Fate of legume and fertilizer nitrogen-15 in a long-term cropping systems experiment

    Includes bibliographical references (pages 914-915).Relying more on biological N2 fixation has been suggested as a way to meet one of the major challenges of agricultural sustainability. A 15N study was conducted to compare the fate of applied legume and fertilizer N in a long-term cropping systems experiment. Nitrogen-15-1abeled red clover (Trifolium pratense L.) and (NH4)2SO4 ere applied microplots within the low-input and conventional cropping systems of the Farming Systems Trial at the Rodale Institute Research Center in Pennsylvania. The 15SN was applied to soil and traced into corn (Zea mays L.) in 1987 and 1988. Residual 15SN was also traced into second-year spring barley (Hordeum vulgare L.). Legume and fertilizer 15SN remaining in soil was measured and loss of N was calculated by difference. More fertilizer than legume N was recovered by crops (40 vs. 17% of input), more legume than fertilizer N was retained in soil (47 vs. 17% of input), and similar amounts of N from both sources were lost from the cropping systems (39% of input) over the 2-yr period. More fertilizer than legume N was lost during the year of application (38 vs. 18% of input), but more legume than fertilizer N was lost the year after application (17 vs. 4% of input). Residual fertilizer and legume 15SN was distributed similarly among soil fractions. Soil microbial biomass was larger in the legume-based system. A larger, but not necessarily more active, soil microbial biomass was probably responsible for the greater soil N supplying capacity in the legume-based compared with fertilizer-based system
