    Preliminary Sizing Study of Ares-I and Ares-V Liquid Hydrogen Tanks

    A preliminary sizing study of two cryogenic propellant tanks was performed using a FORTRAN optimization program to determine weight efficient orthogrid designs for the tank barrels sections only. Various tensile and compressive failure modes were considered, including general buckling of cylinders with a shell buckling knockdown factor. Eight independent combinations of three design requirements were also considered and their effects on the tanks weight. The approach was to investigate each design case with a variable shell buckling knockdown factor, determining the most weight efficient combination of orthogrid design parameters. Numerous optimization analyses were performed, and the results presented herein compare the effects of the different design requirements and shell buckling knockdown factor. Through a series of comparisons between design requirements or shell buckling knockdown factors, the relative change in overall tank barrel weights is shown. The findings indicate that the design requirements can substantually increase the tank weight while a less conservative shell buckling knockdown factor can modestly reduce the tank weight

    The Effects of Fish Trap Mesh Size on Reef Fish Catch off Southeastern Florida

    Catch and mesh selectivity of wire-meshed fish traps were tested for eleven different mesh sizes ranging from 13 X 13 mm (0.5 x 0.5") to 76 x 152 mm (3 X 6"). A total of 1,810 fish (757 kg) representing 85 species and 28 families were captured during 330 trap hauls off southeastern Florida from December 1986 to July 1988. Mesh size significantly affected catches. The 1.5" hexagonal mesh caught the most fish by number, weight, and value. Catches tended to decline as meshes got smaller or larger. Individual fish size increased with larger meshes. Laboratory mesh retention experiments showed relationships between mesh shape and size and individual retention for snapper (Lutjanidae), grouper (Serranidae), jack (Carangidae), porgy (Sparidae), and surgeonfish (Acanthuridae). These relationships may be used to predict the effect of mesh sizes on catch rates. Because mesh size and shape greatly influenced catchability, regulating mesh size may provide a useful basis for managing the commercial trap fishery

    Optical Flameout Detector

    A device has been developed which monitors the presence of a flame within a combustion chamber. The optical flameout detection system responds to gross changes in combustor light intensity which is monitored in two spectral bands. A photomultiplier tube makes optical measurements in the ultraviolet portion of the spectrum and a silicon photodiode covers the visible region. The detectors, located outside the combustion chamber, receive the light energy radiated from the combustion process through fiber optic probes designed to operate in a high pressure environment. The optical fibers are aimed diagonally through the center of the injector at the combustion chamber wall downstream of the injector. The probe observes events occurring within a narrow conical-shaped field of view so that the system can quickly detect longitudinal movement of the flame front away from the injector. If a change in intensity of the flame is detected, the fuel supply to the combustion chamber is shut off limiting the amount of unburned fuel in the combustion chamber which could reignite

    A hierarchical model of transcriptional dynamics allows robust estimation of transcription rates in populations of single cells with variable gene copy number

    Motivation: cis-regulatory DNA sequence elements, such as enhancers and silencers, function to control the spatial and temporal expression of their target genes. Although the overall levels of gene expression in large cell populations seem to be precisely controlled, transcription of individual genes in single cells is extremely variable in real time. It is, therefore, important to understand how these cis-regulatory elements function to dynamically control transcription at single-cell resolution. Recently, statistical methods have been proposed to back calculate the rates involved in mRNA transcription using parameter estimation of a mathematical model of transcription and translation. However, a major complication in these approaches is that some of the parameters, particularly those corresponding to the gene copy number and transcription rate, cannot be distinguished; therefore, these methods cannot be used when the copy number is unknown. Results: Here, we develop a hierarchical Bayesian model to estimate biokinetic parameters from live cell enhancer–promoter reporter measurements performed on a population of single cells. This allows us to investigate transcriptional dynamics when the copy number is variable across the population. We validate our method using synthetic data and then apply it to quantify the function of two known developmental enhancers in real time and in single cells

    Cdc13 OB2 Dimerization Required for Productive Stn1 Binding and Efficient Telomere Maintenance

    SummaryCdc13 is an essential yeast protein required for telomere length regulation and genome stability. It does so via its telomere-capping properties and by regulating telomerase access to the telomeres. The crystal structure of the Saccharomyces cerevisiae Cdc13 domain located between the recruitment and DNA binding domains reveals an oligonucleotide-oligosaccharide binding fold (OB2) with unusually long loops extending from the core of the protein. These loops are involved in extensive interactions between two Cdc13 OB2 folds leading to stable homodimerization. Interestingly, the functionally impaired cdc13-1 mutation inhibits OB2 dimerization. Biochemical assays indicate OB2 is not involved in telomeric DNA or Stn1 binding. However, disruption of the OB2 dimer in full-length Cdc13 affects Cdc13-Stn1 association, leading to telomere length deregulation, increased temperature sensitivity, and Stn1 binding defects. We therefore propose that dimerization of the OB2 domain of Cdc13 is required for proper Cdc13, Stn1, Ten1 (CST) assembly and productive telomere capping

    NASA Langley Research Center (LaRC) recently developed the LaRC Airborne High Spectral Resolution Lidar (HSRL) to make measurements of aerosol and cloud distribution and optical properties. The Airborne HSRL has undergone as series of test flights and was successfully deployed on the Megacity Initiative: Local and Global Research Observations (MILAGRO) field mission in March 2006 (see Hair et al. in these proceedings). This paper provides an overview of the design of the Airborne HSRL and descriptions of some key subsystems unique to this instrument
