    ‘A productive and happy collaboration’: maximising impact through project design

    This case study describes a highly collaborative project created to develop an online self-assessment tool that offers students the opportunity to plan their preparedness to enter the work place named the Preparation for Placement Assessment Tool. The aim of the project was to develop an artefact that aidsall students to think about and prepare for placements, and more broadly to be work-ready. It also aims to help disabled students to identify and manage their individual needs. Working in collaboration with students is offered as an effective strategy for improving project outcomes and managing the tensions for academics of balancing research, teaching, learning and scholarly activities.This paper discusses the project background, design and delivery, which includes the engagement of a diverse mix of students, and the practice research methodologies used. Discussion focuses on the project’s impact, particularly on the students involved. Four key factors: student power, methodology, the use of funding and slow burn are identified as significant for success

    Reduced glycogen availability is associated with increased AMPKα2 activity, nuclear AMPKα2 protein abundance, and GLUT4 mRNA expression in contracting human skeletal muscle

    Glycogen availability can influence glucose transporter 4 (GLUT4) expression in skeletal muscle through unknown mechanisms. The multisubstrate enzyme AMP-activated protein kinase (AMPK) has also been shown to play an important role in the regulation of GLUT4 expression in skeletal muscle. During contraction, AMPK [alpha]2 translocates to the nucleus and the activity of this AMPK isoform is enhanced when skeletal muscle glycogen is low. In this study, we investigated if decreased pre-exercise muscle glycogen levels and increased AMPK [alpha]2 activity reduced the association of AMPK with glycogen and increased AMPK [alpha]2 translocation to the nucleus and GLUT4 mRNA expression following exercise. Seven males performed 60 min of exercise at ~70% [VO.sub.2] peak on 2 occasions: either with normal (control) or low (LG) carbohydrate pre-exercise muscle glycogen content. Muscle samples were obtained by needle biopsy before and after exercise. Low muscle glycogen was associated with elevated AMPK [alpha]2 activity and acetyl-CoA carboxylase [beta] phosphorylation, increased translocation of AMPK [alpha]2 to the nucleus, and increased GLUT4 mRNA. Transfection of primary human myotubes with a constitutively active AMPK adenovirus also stimulated GLUT4 mRNA, providing direct evidence of a role of AMPK in regulating GLUT4 expression. We suggest that increased activation of AMPK [alpha]2 under conditions of low muscle glycogen enhances AMPK [alpha]2 nuclear translocation and increases GLUT4 mRNA expression in response to exercise in human skeletal muscle. <br /

    Muscle glycogen and metabolic regulation

    Muscle glycogen is an important fuel for contracting skeletal muscle during prolonged strenuous exercise, and glycogen depletion has been implicated in muscle fatigue. It is also apparent that glycogen availability can exert important effects on a range of metabolic and cellular processes. These processes include carbohydrate, fat and protein metabolism during exercise, post-exercise glycogen resynthesis, excitation-contraction coupling, insulin action and gene transcription. For example, low muscle glycogen is associated with reduced muscle glycogenolysis, increased glucose and NEFA uptake and protein degradation, accelerated glycogen resynthesis, impaired excitation-contraction coupling, enhanced insulin action and potentiation of the exercise-induced increases in transcription of metabolic genes.     TraceGen: user activity emulation for digital forensic test image generation

    Digital forensic test images are commonly used across a variety of digital forensic use cases including education and training, tool testing and validation, proficiency testing, malware analysis, and research and development. Using real digital evidence for these purposes is often not viable or permissible, especially when factoring in the ethical and in some cases legal considerations of working with individuals' personal data. Furthermore, when using real data it is not usually known what actions were performed when, i.e., what was the ‘ground truth’. The creation of synthetic digital forensic test images typically involves an arduous, time-consuming process of manually performing a list of actions, or following a ‘story’ to generate artefacts in a subsequently imaged disk. Besides the manual effort and time needed in executing the relevant actions in the scenario, there is often little room to build a realistic volume of non-pertinent wear-and-tear or ‘background noise’ on the suspect device, meaning the resulting disk images are inherently limited and to a certain extent simplistic. This work presents the TraceGen framework, an automated system focused on the emulation of user actions to create realistic and comprehensive artefacts in an auditable and reproducible manner. The framework consists of a series of actions contained within scripts that are executed both externally and internally to a target virtual machine. These actions use existing automation APIs to emulate a real user's behaviour on a Windows system to generate realistic and comprehensive artefacts. These actions can be quickly scripted together to form complex stories or to emulate wear-and-tear on the test image. In addition to the development of the framework, evaluation is also performed in terms of the ability to produce background artefacts at scale, and also the realism of the artefacts compared with their human-generated counterparts

    A Dynamical Fossil in the Ursa Minor Dwarf Spheroidal Galaxy

    The nearby Ursa Minor dwarf spheroidal (UMi dSph) is one of the most dark matter dominated galaxies known, with a central mass to light ratio roughly equal to 70. Somewhat anomalously, it appears to contain morphological substructure in the form of a second peak in the stellar number density. It is often argued that this substructure must be transient because it could not survive for the > 10 Gyr age of the system, given the crossing time implied by UMi's 8.8 km/s internal velocity dispersion. In this paper, however, we present evidence that the substructure has a cold kinematical signature, and argue that UMi's clumpiness could indeed be a primordial artefact. Using numerical simulations, we demonstrate that substructure is incompatible with the cusped dark matter haloes predicted by the prevailing Cold Dark Matter (CDM) paradigm, but is consistent with an unbound stellar cluster sloshing back and forth within the nearly harmonic potential of a cored dark matter halo.     First Clear Signature of an Extended Dark Matter Halo in the Draco Dwarf Spheroidal

    We present the first clear evidence for an extended dark matter halo in the Draco dwarf spheroidal galaxy based on a sample of new radial velocities for 159 giant stars out to large projected radii. Using a two parameter family of halo models spanning a range of density profiles and velocity anisotropies, we are able to rule out (at about the 2.5 sigma confidence level) haloes in which mass follows light. The data strongly favor models in which the dark matter is significantly more extended than the visible dwarf. However, haloes with harmonic cores larger than the light distribution are also excluded. When combined with existing measurements of the proper motion of Draco, our data strongly suggest that Draco has not been tidally truncated within ~1 kpc.     ChatGPT for Digital Forensic Investigation: The Good, The Bad, and The Unknown

    The disruptive application of ChatGPT (GPT-3.5, GPT-4) to a variety of domains has become a topic of much discussion in the scientific community and society at large. Large Language Models (LLMs), e.g., BERT, Bard, Generative Pre-trained Transformers (GPTs), LLaMA, etc., have the ability to take instructions, or prompts, from users and generate answers and solutions based on very large volumes of text-based training data. This paper assesses the impact and potential impact of ChatGPT on the field of digital forensics, specifically looking at its latest pre-trained LLM, GPT-4. A series of experiments are conducted to assess its capability across several digital forensic use cases including artefact understanding, evidence searching, code generation, anomaly detection, incident response, and education. Across these topics, its strengths and risks are outlined and a number of general conclusions are drawn. Overall this paper concludes that while there are some potential low-risk applications of ChatGPT within digital forensics, many are either unsuitable at present, since the evidence would need to be uploaded to the service, or they require sufficient knowledge of the topic being asked of the tool to identify incorrect assumptions, inaccuracies, and mistakes. However, to an appropriately knowledgeable user, it could act as a useful supporting tool in some circumstances

    Participatory politics, environmental journalism and newspaper campaigns

    This is an Author's Accepted Manuscript of an article published in Journalism Studies, 13(2), 210 - 225, 2012, copyright Taylor & Francis, available online at: http://www.tandfonline.com/10.1080/1461670X.2011.646398.This article explores the extent to which approaches to participatory politics might offer a more useful alternative to understanding the role of environmental journalism in a society where the old certainties have collapsed, only to be replaced by acute uncertainty. This uncertainty not only generates acute public anxiety about risks, it has also undermined confidence in the validity of long-standing premises about the ideal role of the media in society and journalistic professionalism. The consequence, this article argues, is that aspirations of objective reportage are outdated and ill-equipped to deal with many of the new risk stories environmental journalism covers. It is not a redrawing of boundaries that is needed but a wholesale relocation of our frameworks into approaches better suited to the socio-political conditions and uncertainties of late modernity. The exploration of participatory approaches is an attempt to suggest one way this might be done
