    Diatoms as indicators of historical water quality: A comparison of samples taken in the Wemmershoek catchment (Western Province, South Africa) in 1960 and 2008

    Historical diatom records provide a means of retrospectively determining water quality and inferring ecological condition in rivers and streams. In this study we re-sampled sites originally sampled 48 years previously. We then determined the scores for the Biological Diatom Index (BDI) and the South African Diatom Index (SADI) for each dataset. The results revealed that the present day conditions in this relatively undisturbed locality were almost identical to those reflected by the samples collected half a century before.     Food-web structure in the hypertrophic Rietvlei Dam based on stable isotope analysis: Specific and general implications for reservoir biomanipulation

    Fish predation on zooplankton is the basic foundation for top-down biomanipulation of lacustrine ecosystems. To test this premise, we determined stable isotope (SI) values (ήC and ήN) of representative samples of major planktonic (phytoplankton, zooplankton), benthic (submerged macrophytes and associated epiphytes, benthic macro-invertebrates) and nektonic (fish) food-web components, collected from 3 to 7 shallow inshore locations (with additional plankton samples at 1 or 2 deep offshore sites) in Rietvlei Dam over a period of 30 months. The resulting ήC values did not indicate significant consumption of zooplankton by fish, while the ήN values for fish confirmed their wide trophic separation from zooplankton. Instead, SI values indicated that fish relied mostly on food resources of benthic origin (through direct consumption or piscivory). The SI signatures of individual fish species were consistent with their known feeding habits. The lack of trophic couplings between zooplankton and fish accords with previous gut content analyses of fish and analyses of zooplankton abundance and size structure in hypertrophic reservoirs. Marginal utilisation of zooplankton by indigenous reservoir fish is attributable to their native origin as riverine species unaccustomed to feeding on zooplankton. These findings indicate that top-down biomanipulation is unlikely to be effective as a management tool in eutrophic South African reservoirs. Primary producer components exhibited surprisingly wide and unsystematic temporal fluctuations in both ήC and ήN values; some potential contributory factors are considered. Changes in phytoplankton ήC values were broadly tracked by zooplankton – their nominal consumers.     The relevance of diatoms for water quality assessment in South Africa: A position paper

    Water quality assessment protocols based on the use of diatoms are now well developed and their value substantiated at an international level. The use of diatoms is not designed or intended to be a “rapid” technology. The detailed level of information generated from the procedure outweighs perceived disadvantages of the additional time required for sample preparation and analysis to species level. The method is applicable across a wide range of aquatic ecosystem types, namely freshwater, brackish, and estuarine, and is inclusive of both lentic and lotic environments, wetlands and their associated damp, marginal and littoral zones. Details provided by diatom assemblages support palaeoecological investigations, historical reconstruction of water quality and the determination of prevailing water quality conditions. Deliberate determination of responses to management strategies or impacts arising from a variety of anthropogenic activities can be achieved via the simple expedient of retrieving living material from introduced artificial substrates. Previous studies in South Africa and elsewhere have shown that on a site-by-site basis the use of diatoms provides a fine level of diagnostic resolution of the causes underlying changes in water quality and environmental condition. The South African Diatom Collection (“the Collection”), a repository of diatom specimens and records that spans the length and breadth of this country, contains an as-yet unutilised wealth of ecological and taxonomic information. More importantly, the historical data analysis records provide an insight into water quality conditions prevailing 40 to 50 years ago – in many cases prior to the “development” of many of our rivers, streams and wetlands. The real value of its existence underpins the great potential for renewed attention to the value of diatom-based approaches to water quality assessments. In addition, the Collection provides a ready-made foundation on which a locally relevant tool for water quality assessment may be established to augment the current use of invertebrate indicators. It is now appropriate that the full potential of the use of diatoms in water quality assessments, and the information contained in the Collection, be developed and utilised for water quality assessment in South Africa.     Diatoms as indicators of water quality in the Jukskei-Crocodile river system in 1956 and 1957, a re-analysis of diatom count data generated by BJ Cholnoky

    South Africa has a long legacy of diatom research. The eminent diatomist Dr BJ Cholnoky spent much of his working life examining and enumerating diatom communities found in Southern Africa. Most if not all of Cholnoky's collected diatom material in the form of mounted material on glass slides accompanied by diatom analysis sheets is stored in the South African Diatom Collection currently housed at the CSIR in Durban. As Cholnoky only employed enumeration methods yielding a margin of error of 2% or less, Cholnoky's results should provide an accurate reflection of the structure of the diatom communities that he examined. It is the aim of the present study to demonstrate the value of these historical diatom analyses for inferring past water quality conditions using the diatom-based index method. Data for the Jukskei-Crocodile River system were obtained from the South African Diatom Collection for the period 1956/1957. The nomenclature of the diatoms listed on Cholnoky's data sheets was modernised and the data then entered into OMNIDIA v3.1. Diatom index scores generated from OMNIDIA v3.1 were in general in agreement with Cholnoky's own assessment of water quality (especially with reference to organic pollution). It is concluded that the diatom analysis records housed in the South African Diatom Collection constitute a valuable resource for the assessment of past conditions of rivers and streams..     Assessing sedimentation equilibrium profiles in analytical ultracentrifugation experiments on macromolecules: from simple average molecular weight analysis to molecular weight distribution and interaction analysis

    Molecular weights (molar masses), molecular weight distributions, dissociation constants and other interaction parameters are fundamental characteristics of proteins, nucleic acids, polysaccharides and glycoconjugates in solution. Sedimentation equilibrium in the analytical ultracentrifugation provides a powerful method with no supplementary immobilization, columns or membranes required. It is particularly powerful when used in conjunction with its sister technique, namely sedimentation velocity analysis. We describe key approaches now available and their application to the characterisation of antibodies polysaccharides and glycoconjugates. We indicate how major complications such as thermodynamic non-ideality can now be routinely dealt with, thanks to a great extent to the extensive contribution of Professor DonWinzor over several decades of research

    Hydrodynamic modelling of protein conformation in solution: ELLIPS and HYDRO

    The last three decades has seen some important advances in our ability to represent the conformation of proteins in solution on the basis of hydrodynamic measurements. Advances in theoretical modeling capabilities have been matched by commensurate advances in the precision of hydrodynamic measurements. We consider the advances in whole-body (simple ellipsoid-based) modeling—still useful for providing an overall idea of molecular shape, particularly for those systems where only a limited amount of data is available—and outline the ELLIPS suite of algorithms which facilitates the use of this approach. We then focus on bead modeling strategies, particularly the surface or shell–bead approaches and the HYDRO suite of algorithms. We demonstrate how these are providing great insights into complex issues such as the conformation of immunoglobulins and other multi-domain complexes

    Variation in styles of rifting in the Gulf of California

The definitive version was published in Nature 448 (2007): 466-469, doi:10.1038/nature06035.The rifting of continental lithosphere is a fundamental solid-earth process that leads to the formation of rifted continental margins and ocean basins. Understanding of this process comes from observations of the geometry of rifted margins and the magmatism resulting from rifting, which inform us about the strength of the lithosphere, the state of the underlying mantle, and the transition from rifting to seafloor spreading. Here we describe results from the PESCADOR seismic experiment in the southern Gulf of California and present the first crustal-scale images across conjugate margins of multiple segments within a single rift that has reached the stage of oceanic spreading. A surprisingly large variation in rifting style and magmatism is observed between these segments, from wide rifting with minor syn-rift magmatism to narrow rifting in magmatically robust segments. These differences encompass much of the variation observed across nearly all other non-end-member continental margins. The characteristics of magmatic endmember margins are typically explained in terms of mantle temperature. Our explanations for the variation in the Gulf of California, in contrast, invoke mantle depletion to account for wide, magma-poor rifting and mantle fertility and possibly the influence of sediments to account for robust rift and post-rift magmatism in the Gulf of California. These factors may vary laterally over small distances in regions that have transitioned from convergence to extension, as is the case for the Gulf of California and many other rifts.This work was funded by a grant from the U.S. NSF-MARGINS program

    Mass Treatment with Azithromycin for Trachoma Control: Participation Clusters in Households

    Trachoma, an infectious disease, continues to cause blindness. A great deal of the trachoma burden is concentrated in developing countries. The World Health Organization recommends mass treatment for entire communities in trachoma-endemic regions. In 32 Tanzanian and 48 Gambian communities with trachoma, mass treatment was directly observed following a census. Community coverage was mostly greater than 80%. Larger-than-expected proportions of households where all children were treated and where none of the children were treated were found in each country. Household clustering of treatment was higher in Tanzania compared to The Gambia. However, children who were not treated were not more likely to be infected compared to children who were treated. We found that treatment and non-treatment within communities does not occur at random but rather clusters within households. These findings impact the design of future coverage surveys and suggest that further research evaluate factors that are associated with familial non-compliance

    A sociological dilemma: race, segregation, and US sociology

    US sociology has been historically segregated in that, at least until the 1960s, there were two distinct institutionally organized traditions of sociological thought – one black and one white. For the most part, however, dominant historiographies have been silent on that segregation and, at best, reproduce it when addressing the US sociological tradition. This is evident in the rarity with which scholars such as WEB Du Bois, E Franklin Frazier, Oliver Cromwell Cox, or other ‘African American Pioneers of Sociology’, as Saint-Arnaud calls them, are presented as core sociological voices within histories of the discipline. This article addresses the absence of African American sociologists from the US sociological canon and, further, discusses the implications of this absence for our understanding of core sociological concepts. With regard to the latter, the article focuses in particular on the debates around equality and emancipation and discusses the ways in which our understanding of these concepts could be extended by taking into account the work of African American sociologists and their different interpretations of core themes
