    Experimental Investigation of Angular Stackgram Filtering for Noise Reduction of SPECT Projection Data: Study with Linear and Nonlinear Filters

    We discuss data filtering prior to image reconstruction. For this kind of filtering, the radial direction of the sinogram is routinely employed. Recently, we have introduced an alternative approach to sinogram data processing, exploiting the angular information in a novel way. This new stackgram representation can be regarded as an intermediate form of the sinogram and image domains. In this experimental study, we compare the radial sinogram and angular stackgram filtering methods using physical SPECT phantoms. Our study is carried out by employing simple linear and nonlinear filters with ten different Gaussian kernels, in order to provide a comparable investigation. According to our results, angular stackgram filtering with the nonlinear filters provides the best resolution-noise tradeoff of the compared methods. Besides, stackgram filtering with these filters seems to preserve the resolution in an exceptional way. Visually, noise in the reconstructed images after stackgram filtering appears more “powdery” in comparison with radial sinogram filtering

    Biochemical and structural characterisation of membrane-containing icosahedral dsDNA bacteriophages infecting thermophilic Thermus thermophilus

    AbstractIcosahedral dsDNA viruses isolated from hot springs and proposed to belong to the Tectiviridae family infect the Gram-negative thermophilic Thermus thermophilus bacterium. Seven such viruses were obtained from the Promega Corporation collection. The structural protein patterns of three of these viruses, growing to a high titer, appeared very similar but not identical. The most stable virus, P23-77, was chosen for more detailed studies. Analysis of highly purified P23-77 by thin layer chromatography for neutral lipids showed lipid association with the virion. Cryo-EM based three-dimensional image reconstruction of P23-77 to 1.4 nm resolution revealed an icosahedrally-ordered protein coat, with spikes on the vertices, and an internal membrane. The capsid architecture of P23-77 is most similar to that of the archaeal virus SH1. These findings further complicate the grouping of icosahedrally-symmetric viruses containing an inner membrane.     Black Mesoporous Silicon as a Contrast Agent for LED-Based 3D Photoacoustic Tomography

    Mesoporous silicon (PSi) nanoparticles have been widely studied in different biomedical imaging modalities due to their several beneficial material properties. However, they have not been found to be suitable for photoacoustic imaging due to their poor photothermal conversion performance. In the present study, biodegradable black mesoporous silicon (BPSi) nanoparticles with strong light absorbance were developed as superior image contrast agents for photoacoustic tomography (PAT), which was realized with a light-emitting diode (LED) instead of the commonly used laser. LED-based PAT offers the advantages of low cost, compactness, good mobility, and easy operation as compared to the traditional laser-based PAT modality. Nevertheless, the poor imaging sensitivity of the LED-PAT systems has been the main barrier to prevent their wide biomedical application because the LED light has low optical energy. The present study demonstrated that the imaging sensitivity of the LED-PAT system was significantly enhanced with the PEGylated BPSi (PEG–BPSi) nanoparticles. The PEG–BPSi nanoparticles were clearly detectable with a low concentration of 0.05 mg/mL in vitro and with an LED radiation energy of 5.2 ÎŒJ. The required concentration of the PEG–BPSi nanoparticles was 10 times lesser than that of the reference gold nanoparticles to reach the corresponding level of the imaging contrast. The ex vivo studies demonstrated that the submillimeter BPSi nanoparticle-based absorbers were distinguishable in chicken breast tissues. The strong contrast provided by the BPSi particles indicated that these particles can be utilized as novel contrast agents in PAT, especially in LED-based systems with low light intensity

    Long-term outcome after treatment of pulmonary atresia with ventricular septal defect : nationwide study of 109 patients born in 1970-2007

    OBJECTIVES: Treatment of pulmonary atresia with ventricular septal defect (PA + VSD) has evolved during recent decades, but it still remains challenging. This study evaluated 41-year experience of outcome, survival and treatment of PA + VSD patients. METHODS: Patient records and angiograms of 109 patients with PA + VSD born in Finland between 1970 and 2007, and treated at the Children's Hospital, Helsinki University Central Hospital, were retrospectively analysed in this nationwide study. RESULTS: Of the 109 patients, 66 (61%) had simple PA + VSD without major aortopulmonary collateral arteries (MAPCAs). Although we observed no difference in overall survival between those with or without MAPCAs, the patients without MAPCAs had better probability to achieve repair (64 vs 28%, P <0.0003). Only 3 patients were treated by compassionate care. Overall survival was affected by the size of true central pulmonary arteries on the first angiogram (P = 0.001) and whether repair was achieved (P <0.0001). After successful repair, the survival rate was 93% at 1 year, 91% from the second year, and functional capacity as assessed by New York Heart Association (NYHA) I-II remained in 85% of patients alive at the end of follow-up. Palliated patients at 1, 5, 10 and 20 years of age had Kaplan-Meier estimated survival rates of 55, 42, 34 and 20%, respectively. Patients who underwent repair attempts but were left palliated with right ventricle (RV)-pulmonary artery connection and septal fenestration had better survival than the rest of the palliated patients (P = 0.001). Further, the McGoon index improved after implementation of a systemic-pulmonary artery shunt in the overall PA + VSD population (P <0.0001). CONCLUSIONS: These findings show that achievement of repair and initial size of true central pulmonary arteries affect survival of patients with PA + VSD. Although the overall survival of patients with MAPCAs showed no difference compared with simple PA + VSD patients, they had a higher risk of remaining palliated. However, palliative surgery may have a role in treatment of PA + VSD because the size of pulmonary arteries increased after placement of systemic-pulmonary artery shunt. In addition, subtotal repair by a RV-pulmonary artery connection and septal fenestration improved survival over extracardiac palliation.Peer reviewe

    Dental age in patients with neurofibromatosis 1

    Neurofibromatosis type 1 (NF1) is a common, hereditary, neurocutaneous skeletal condition with a variety of osseous manifestations. Although NF1 also affects craniofacial structures, the literature has only limited information related to the effect of NF1 on dental development. In this study the dental age of 34 Finnish NF1 patients, 817 yr of age, was estimated using the Demirjian & Goldstein method. The dental age of children with NF1 was similar to that of a Finnish control population. The findings suggest that NF1 does not affect the timing of tooth development.</p

    Bone marrow mesenchymal stem cells do not enhance intra-synovial tendon healing despite engraftment and homing to niches within the synovium

    Intra-synovial tendon injuries display poor healing, which often results in reduced functionality and pain. A lack of effective therapeutic options has led to experimental approaches to augment natural tendon repair with autologous mesenchymal stem cells (MSCs) although the effects of the intra-synovial environment on the distribution, engraftment and functionality of implanted MSCs is not known.
