Accuracy of the Aspartic Acid Racemization Technique in Age Estimation of Mammals and the Influence of Body Temperature
The aspartic acid racemization (AAR) technique has been applied for age estimation of humans and other mammals for more than four decades. In this study, eye lenses from 124 animals representing 25 mammalian species were collected and D/L ratios obtained using the AAR technique. The animals were either of known age or had the age estimated by other methods. The purpose of the study was to: a) estimate the accuracy of the AAR technique, and b) examine the effect of body temperature on racemization rates. Samples from four of the 25 species covered the range of ages that is needed to estimate species-specific racemization rates. The sample size from a single species of known age, the pygmy goat (Capra hircus, n = 35), was also large enough to investigate the accuracy of ages obtained using the AAR technique. The 35 goats were divided into three datasets: all goats (n = 35), goats >0.5 yrs old (n = 26) and goats >2 yrs old (n = 19). Leave-one-out analyses were performed on the three sets of data. Normalized root mean squared errors for the group of goats >0.5 yrs old were found to be the smallest. The higher variation in D/L measurements found for young goats 0.5 yrs old was for three age groups of the goats: 0.934 yrs for young goats 8 yrs (n = 4). Thus, the age of an adult or an old animal can be predicted with approximately 10% accuracy, whereas the age of a young animal is difficult to predict. A goat specific racemization rate, as a 2kAsp value, was estimated to 0.0107 ± 3.8 x 10-4 SE (n = 26). The 2kAsp values from 12 species, four estimated in this study and another eight published, were used to examine the effect of core body temperature on the rate of racemization. A positive relationship between AAR and temperature was found (r2 = 0.321) but results also suggest that other factors besides temperature are involved in the racemization process in living animals. Based on our results we emphasize that non-species-specific racemization rates should be used with care in AAR age estimation studies and that the period of postnatal growth of the eye lens be considered when estimating species-specific D/L0 values and ages of young individuals
Peak Power Output in Loaded Jump Squat Exercise is Affected by Set Structure
International Journal of Exercise Science 11(1): 776-784, 2018. A priority in strength and power exercise might be to train with as high quality as possible for the shortest possible duration. In this context, peak power output could reflect quality. Designing an exercise session as a cluster set structure, as compared to a traditional set structure, may be a way to obtain higher peak power output in the session. But it is unknown whether that is obtainable for non-elite individuals performing loaded jump squat exercise. The aim of the present study was therefore to test the hypothesis that peak power output would be highest in a jump squat exercise session, which was structured with cluster sets, as compared to traditional sets. Ten individuals (2 women, 8 men; 26.5 ± 4.8 years, 1.81 ±0.08 m, 90.9 ± 13.2 kg) performed two loaded jump squat exercise sessions structured with cluster sets and traditional sets, respectively. The sessions were performed on two separate days, in counterbalanced order. The position of the barbell was used to calculate derived values including peak power output. Values calculated as averages across the entire exercise sessions showed peak power output to be 178 ± 181 W, corresponding to 4.1% ± 4.9%, higher in the session with cluster set structure, as compared to the session with traditional set structure (p = 0.005). It was concluded that for non-elite individuals, peak power output was approximately 4% higher in a loaded jump squat exercise session structured with cluster sets as compared to an exercise session structured with traditional sets
Organ Support Therapy in the Intensive Care Unit and Return to Work in Out-of-Hospital Cardiac Arrest Survivors:a Nationwide Cohort Study
AIM: With increased survival after out-of-hospital cardiac arrest (OHCA), impact of the post-resuscitation course has become important. Among 30-day OHCA survivors, we investigated associations between organ support therapy in the Intensive Care Unit (ICU) and return to work.METHODS: This Danish nationwide cohort-study included 30-day-OHCA-survivors who were employed prior to arrest. We linked OHCA data to information on in-hospital care and return to work. For patients admitted to an ICU and based on renal replacement therapy (RRT), cardiovascular support and mechanical ventilation, we assessed the prognostic value of organ support therapies in multivariable Cox regression models.RESULTS: Of 1,087 30-day survivors, 212 (19.5%) were treated in an ICU with 0-1 types of organ support, 494 (45.4%) with support of two organs, 26 (2.4%) with support of three organs and 355 (32.7%) were not admitted to an ICU. Return to work increased with decreasing number of organs supported, from 53.8% (95% CI: 49.5-70.1%) in patients treated with both RRT, cardiovascular support and mechanical ventilation to 88.5% (95% CI: 85.1-91.8%) in non-ICU-patients. In 732 ICU-patients, ICU-patients with support of 3 organs had significantly lower adjusted hazard ratios (HR) of returning to work (0.50 [95% CI: 0.30-0.85] compared to ICU-patients with support of 0-1 organ. The corresponding HR was 0.48 [95% CI: 0.30-0.78] for RRT alone.CONCLUSIONS: In 30-day survivors of OHCA, number of organ support therapies and in particular need of RRT were associated with reduced rate of return to work, although more than half of these latter patients still returned to work.</p
Osteoprotegerin and coronary artery disease in type 2 diabetic patients with microalbuminuria
<p>Abstract</p> <p>Objective</p> <p>Plasma osteoprotegerin (P-OPG) is an independent predictor of cardiovascular disease in diabetic and other populations. OPG is a bone-related glycopeptide produced by vascular smooth muscle cells and increased P-OPG may reflect arterial damage. We investigated the correlation between P-OPG and coronary artery disease (CAD) in asymptomatic type 2 diabetic patients with microalbuminuria.</p> <p>Methods</p> <p>P-OPG was measured in 200 asymptomatic diabetic patients without known cardiac disease. Patients with P-NT-proBNP >45.2 ng/l and/or coronary calcium score (CCS) ≥400 were stratified as high risk of CAD (n = 133), and all other patients as low risk patients (n = 67). High risk patients were examined by myocardial perfusion imaging (MPI; n = 109), and/or CT-angiography (n = 20), and/or coronary angiography (CAG; n = 86). Significant CAD was defined by presence of significant myocardial perfusion defects at MPI and/or >70% coronary artery stenosis at CAG.</p> <p>Results</p> <p>Significant CAD was demonstrated in 70 of the high risk patients and of these 23 patients had >70% coronary artery stenosis at CAG. Among high risk patients, increased P-OPG was an independent predictor of significant CAD (adjusted odds ratio [CI] 3.11 [1.01-19.54] and 3.03 [1.00-9.18] for second and third tertile vs.first tertile P-OPG, respectively) and remained so after adjustments for NT-proBNP and CCS. High P-OPG was also associated with presence of >70% coronary artery stenosis(adjusted odds ratio 14.20 [1.35-148.92] for third vs. first tertile P-OPG), and 91% of patients with low (first tertile) P-OPG did not have >70% coronary artery stenosis.</p> <p>Conclusions</p> <p>Elevated P-OPG is an independent predictor of the presence of CAD in asymptomatic type 2 diabetic patients with microalbuminuria.</p
