    Vasculogenic mimicry contributes to lymph node metastasis of laryngeal squamous cell carcinoma

    <p>Abstract</p> <p>Background</p> <p>Survival of laryngeal squamous cell carcinoma (LSCC) patients has remained unchanged over recent years due to its uncontrolled recurrence and local lymph node metastasis. Vasculogenic mimicry (VM) is an alternative type of blood supplement related to more aggressive tumor biology and increased tumor-related mortality. This study aimed to investigate the unique role of VM in the progression of LSCC.</p> <p>Methods</p> <p>We reviewed clinical pathological data of 203 cases of LSCC both prospectively and retrospectively. VM and endothelium-dependent vessel (EDV) were detected by immunohistochemistry and double staining to compare their different clinical pathological significance in LSCC. Survival analyses were performed to assess their prognostic significance as well.</p> <p>Results</p> <p>Both VM and EDV existed in LSCC type of blood supply. VM is related to pTNM stage, lymph node metastasis and pathology grade. In contrust, EDV related to location, pTNM stage, T stage and distant metastasis. Univariate analysis showed VM, pTNM stage, T classification, nodal status, histopathological grade, tumor size, and radiotherapy to be related to overall survival (OS). While, VM, location, tumor size and radiotherapy were found to relate to disease free survival (DFS). Multivariate analysis indicated that VM, but not EDV, was an adverse predictor for both OS and DFS.</p> <p>Conclusions</p> <p>VM existed in LSCC. It contributed to the progression of LSCC by promoting lymph node metastasis. It is an independent predictors of a poor prognosis of LSCC.</p

    Biochemical Characterization and Phylogenetic Analysis of the Virulence Factor Lysine Decarboxylase From Vibrio vulnificus

    Cadaverine is produced in organisms from the amino acid lysine in a decarboxylation reaction catalyzed by lysine decarboxylase (EC 4.1.1.18). The inducible lysine decarboxylase CadA plays a vital role in acid stress response for enteric bacteria. Vibrio vulnificus is an extremely virulent human pathogen causing gastroenteritis when the acid conditions that prevent survival of V. vulnificus in the stomach or small intestine are overcome. A gene encoding CadA was identified from V. vulnificus. Subsequent analyses showed that CadA from V. vulnificus (VvCadA) is a decamer with a 82-kDa subunit. Homogenous VvCadA was purified from Escherichia coli and used for lysine decarboxylation with an optimal pH of 6.0 and optimal temperature of 37°C. The apparent Vmax and Km for lysine were 9.45 ± 0.24 μM/min and 0.45 ± 0.05 mM, respectively. Mutation analysis suggested that the amino-acid-binding pyridoxal phosphate, the cofactor of the enzyme, plays a vital role in the reaction. Mutation of the negatively charged residues interacting with lysine also affected the activity of the enzyme to some extent. Quantitative RT-PCR showed that expression of VvcadA was up-regulated under low pH, low salinity, and oxidative stresses. Furthermore, the concentration of cadaverine released to the cell exterior also increased under these stresses. Protein sequence similarity network (SSN) analysis indicated that lysine decarboxylases with ornithine decarboxylases and arginine decarboxylases shared a common ancestor, and that lysine decarboxylases are more conserved during evolution. Our data provide evidence for the biochemical characteristics and important roles of VvCadA under stress conditions

    Phosphorus adsorption characteristics and release risk in saline soils: a case study of Songnen Plain, China

    IntroductionThe Songnen Plain is one of the three major saline-alkali areas in China, covering a vast area, where drought and overgrazing have exacerbated the salinization trend, and will have great potential for development if utilized rationally. Phosphorus, as one of important soil nutrients, plays a crucial role in plant growth. How to minimize its loss and migration has become a current research hotspot. The objective of the present study was to elucidate the adsorption properties of phosphorus in soils affected by salinization and to establish the correlation between the potential for phosphorus release and soil properties.MethodsA batch treatment test was conducted in this study using three soils with the various salinization degrees to examine the impact of environmental factors on the adsorption properties and potential release of phosphorus.Results and discussionIt was found that the maximum phosphorus adsorption by the three salinization soils in 0-360 minutes accounted for 86.8%-90.5% of the total adsorption capacity; the equilibrium adsorption capacity was: HS&gt; MS&gt; LS. In cases where the phosphorus level in the surrounding liquid is low, the three levels of salinized soils exhibited varying levels of phosphorus discharge, with the adsorbent acting as the origin of contaminants. The Pseudo-second-order model kinetics and Langmuir equation can well describe the adsorption process, and the adsorption process is spontaneous heat absorption with entropy increase. Increasing the pH led to an increase in the adsorption of phosphorus from the three salinized soils. Additionally, the adsorption was enhanced by introducing varying concentrations of Na+, Ca2+, and Al3+ to the background solution. The phosphorus eutrophication release risk (ERI) demonstrated a gradual decline as temperature increased. Correlation analysis revealed a noteworthy positive correlation between TN, TP, and ERI, as well as a significant negative correlation between CEC, K+, and ERI. Furthermore, there was a highly significant negative correlation between coarse silt and fine silt. Considering local climatic and environmental factors is crucial for controlling the adsorption capacity of phosphorus in various salinized soils, as it can unveil the mechanism of phosphorus adsorption and impact its migration and release risk

    Dynamic chromatin organization and regulatory interactions in human endothelial cell differentiation

    Vascular endothelial cells are a mesoderm-derived lineage with many essential functions, including angiogenesis and coagulation. The gene-regulatory mechanisms underpinning endothelial specialization are largely unknown, as are the roles of chromatin organization in regulating endothelial cell transcription. To investigate the relationships between chromatin organization and gene expression, we induced endothelial cell differentiation from human pluripotent stem cells and performed Hi-C and RNA-sequencing assays at specific time points. Long-range intrachromosomal contacts increase over the course of differentiation, accompanied by widespread heteroeuchromatic compartment transitions that are tightly associated with transcription. Dynamic topologically associating domain boundaries strengthen and converge on an endothelial cell state, and function to regulate gene expression. Chromatin pairwise point interactions (DNA loops) increase in frequency during differentiation and are linked to the expression of genes essential to vascular biology. Chromatin dynamics guide transcription in endothelial cell development and promote the divergence of endothelial cells from cardiomyocytes

    Investigation of Losses in Industrial Enterprises in Liaoning Province

    Liaoning is an area where heavy industry is relatively concentrated, making up 69 percent of the provincial industrial total, while its light industry only makes up 31 percent. In the province's economic development, however, enterprise losses are a conspicuous problem. In June 1987, we investigated losses in some enterprises in a few industries, focusing on operational losses suffered by industrial enterprises included in the state budget. On the basis of this investigation and study, we present below some views and suggestions.
