    The Toxicity of Nanoparticles Depends on Multiple Molecular and Physicochemical Mechanisms

    Nanotechnology is an emerging discipline that studies matters at the nanoscale level. Eventually, the goal is to manipulate matters at the atomic level to serve mankind. One growing area in nanotechnology is biomedical applications, which involve disease management and the discovery of basic biological principles. In this review, we discuss characteristics of nanomaterials, with an emphasis on transition metal oxide nanoparticles that influence cytotoxicity. Identification of those properties may lead to the design of more efficient and safer nanosized products for various industrial purposes and provide guidance for assessment of human and environmental health risk. We then investigate biochemical and molecular mechanisms of cytotoxicity that include oxidative stress-induced cellular events and alteration of the pathways pertaining to intracellular calcium homeostasis. All the stresses lead to cell injuries and death. Furthermore, as exposure to nanoparticles results in deregulation of the cell cycle (i.e., interfering with cell proliferation), the change in cell number is a function of cell killing and the suppression of cell proliferation. Collectively, the review article provides insights into the complexity of nanotoxicology

    Giving circles are made up of individuals who pool their resources and then decide together where these should be distributed. They also include social, educational, and engagement components that seem to engage participants in their communities and increase members’ understanding of philanthropy and community issues. This study examines if and how participation in a giving circle has changed members’ behaviorrelated to giving, volunteering, and civic engagement. In addition, we asked if and how participation in a giving circle has changed members’ awareness or knowledge about philanthropy, nonprofit organizations, and community issues. Finally, we wanted to know if and how participation in a giving circle has changed members’ perceptions or attitudes about philanthropy, community issues, citizen, government and nonprofit roles and responsibilities or political and social values.     Donors in giving circles give more, give more strategically, and are more engaged in their communities, according to a new study by Dr. Angela Eikenberry and Jessica Bearman with research assistance from Melissa Brown, Hao Han, and Courtney Jensen. ever since giving circles—groups of individual donors who pool their money and other resources and decide together where these should be distributed—emerged as a philanthropic trend, we have speculated about their impact. Do donors give more or give differently because they are involved in a giving circle? Do they become more engaged and active in their communities? Are they more politically active? Until now, the evidence of giving circles’ impact on donors was mainly qualitative. Now, this new study examines, in a more comprehensive and quantitative manner, the impact of giving circles on their members’ giving and civic engagement.

    A systematic analysis of the PARP protein family identifies new functions critical for cell physiology

    The poly(ADP-ribose) polymerase (PARP) family of proteins use NAD[superscript +] as their substrate to modify acceptor proteins with ADP-ribose modifications. The function of most PARPs under physiological conditions is unknown. Here, to better understand this protein family, we systematically analyse the cell cycle localization of each PARP and of poly(ADP-ribose), a product of PARP activity, then identify the knockdown phenotype of each protein and perform secondary assays to elucidate function. We show that most PARPs are cytoplasmic, identify cell cycle differences in the ratio of nuclear to cytoplasmic poly(ADP-ribose) and identify four phenotypic classes of PARP function. These include the regulation of membrane structures, cell viability, cell division and the actin cytoskeleton. Further analysis of PARP14 shows that it is a component of focal adhesion complexes required for proper cell motility and focal adhesion function.     Curing Hepatitis C in Liver Transplant Recipients Is Associated with Changes in Immunosuppressant Use.

    Background and aimsAll-oral interferon-free antivirals are highly effective in treating recurrent hepatitis C (HCV) infection in liver transplant (LT) recipients. The aim of the study was to assess immunosuppression needs after achieving a sustained viral response (SVR).MethodsWe compared immunosuppression needs before and after achieving a SVR in adult LT recipients treated for recurrent HCV infection with all-oral direct acting agents.ResultsWe identified 52 liver LT treated recipients who achieved a SVR. The median (25th and 75th percentile interquartile range [IQR]) age was 62 years (57.75, 65). Most recipients received tacrolimus (TAC) for their immunosuppressant regimen. After achieving SVR, there was no statistically significant difference in daily dose of TAC unadjusted per weight (p > 0.05). However, there was a statistically significant decrease in daily dose of TAC adjusted per weight, serum levels of TAC, and the product of glomerular filtration rate and TAC. No statistically significant differences in cyclosporine unadjusted/adjusted per weight daily dose or serum levels were noted.ConclusionsImmunosuppression needs were increased for those patients treated with TAC but not cyclosporine. LT recipients prescribed TAC require close monitoring after treatment completion to avoid potential risk of acute rejection
