    Utjecaj uvjeta uzgoja i dodatka soli na sastav eteričnog ulja slatkog mažurana (Origanum majorana) iz Tunisa

    O. majorana shoots were investigated for their essential oil (EO) composition. Two experiments were carried out; the first on hydroponic medium in a culture chamber and the second on inert sand in a greenhouse for 20 days. Plants were cultivated for 17 days in hydroponic medium supplemented with NaCl 100 mmol L1. The results showed that the O. majorana hydroponic medium offered higher essential oil yield than that from the greenhouse. The latter increased significantly in yield (by 50 %) under saline constraint while it did not change in the culture chamber. Under greenhouse conditions and in the absence of salt treatment, the major constituents were terpinene-4-ol and trans-sabinene hydrate. However, in the culture chamber, the major volatile components were cis-sabinene hydrate and terpinene-4-ol. In the presence of NaCl, new compounds appeared, such as eicosane, spathulenol, eugenol, and phenol. In addition, in the greenhouse, with or without salt, a very important change of trans-sabinene hydrate concentration in EO occurred, whereas in the culture chamber change appeared in cis-sabinene hydrate content.U radu je opisano ispitivanje sastava eteričnog ulja izdanaka biljke O. majorana. Provedena su dva eksperimenta: prvi na hidroponom mediju u komorama za uzgoj, a drugi na inertnom pijesku u stakleniku tijekom 20 dana. Biljke su uzgajane 17 dana u hidroponom mediju u koji je dodan NaCl 100 mmol L1. Rezultati ukazuju na to da hidroponi medij O. majorana osigurava veće prinose eteričnog ulja nego staklenik. U stakleniku se prinos ulja značajno povećao dodavanjem 50 % soli dok u uzgoju u uzgojnoj komori nije bilo promjene. U uvjetima u stakleniku i u odsutnosti soli, najvažniji sastojci ulja bili su terpinen-4-ol i trans-sabinen hidrat, dok su u uvjetima uzgojne komore najvažnije hlapljive komponente bile cis-sabinen hidrat i terpinen-4-ol. U prisutnosti NaCl-a, pojavili su se novi sastojci, kao što su eikozan, spatulenol, eugenol i fenol. Dodatno je uz stakleničke uvjete, sa i bez soli, došlo do važne promjene u količini trans-sabinen hidrata u eteričnom ulju, dok se u komorama promijenio sadržaj cis-sabinen hidrata

    Lipid, fatty acid and tocol distribution of coriander fruit's different parts

    International audienceHigh amounts of neutral lipids (NL) were found (95.65% of total lipids) in whole fruit oil, while glycolipids (GLs) occurred at high levels in pericarp oil (73.21% of total lipids). Triacylglycerol was found to be the principal lipid fraction of NL which formed 93% in seed and whole fruit. However, monoacylglycerol constituted the major fraction of NL in pericarp (34.5%). At least two GLs and five phospholipids (PL) were identified. The CL composition of the pericarp is characterized by monogalactosylacylglycerol as the major fraction with a percentage of 53.39% of total lipids. The major individual PL subclasses were phosphaticlylcholine followed by phosphaticlylethanolamine in seed and whole fruit. However, the PL were not detected in pericarp, oil. The principal fatty acids identified in most lipid classes were petroselinic acid (C18:1n-12), linoleic acid (C18:2n-6), palmitic (C16:0) and oleic acid (C18:1n-9). The total tocopherol and tocotrienol (tocol) contents were 27.78 mg/100 g oil in whole fruit, 26.42 mg/100g oil in seed and 5.36mg/100g oil in pericarp. Fruit and seed oils were characterized by a high amount of gamma-tocotrienol with 19.56 mg/100 g oil. However, a-tocopherol (1.82 mg/100g oil) was found to be the tocol marker in pericarp oils. The results are important as an indication of the potentially economical utility of Coriandrum sativum L. seed oil as a new source of PL. (C) 2009 Published by Elsevier B.V

    Beneficial Effect of Jojoba Seed Extracts on Hyperglycemia-Induced Oxidative Stress in RINm5f Beta Cells

    Hyperglycemia occurs during diabetes and insulin resistance. It causes oxidative stress by increasing reactive oxygen species (ROS) levels, leading to cellular damage. Polyphenols play a central role in defense against oxidative stress. In our study, we investigated the antioxidant properties of simmondsin, a pure molecule present in jojoba seeds, and of the aqueous extract of jojoba seeds on fructose-induced oxidative stress in RINm5f beta cells. The exposure of RINm5f beta cells to fructose triggered the loss of cell viability (−48%, p < 0.001) and disruption of insulin secretion (p < 0.001) associated with of reactive oxygen species (ROS) production and a modulation of pro-oxidant and antioxidant signaling pathway. Cell pre-treatments with extracts considerably increased cell viability (+86% p < 0.001) for simmondsin and +74% (p < 0.001) for aqueous extract and insulin secretion. The extracts also markedly decreased ROS (−69% (p < 0.001) for simmondsin and −59% (p < 0.001) for aqueous extract) and caspase-3 activation and improved antioxidant defense, inhibiting p22phox and increasing nuclear factor (erythroid-derived 2)-like 2 (Nrf2) levels (+70%, p < 0.001) for aqueous extract. Simmondsin had no impact on Nrf2 levels. The richness and diversity of molecules present in jojoba seed extract makes jojoba a powerful agent to prevent the destruction of RINm5f beta cells induced by hyperglycemia

    Extraction and Characterization of Tunisian Quercus ilex Starch and Its Effect on Fermented Dairy Product Quality

    In this study, a new starch has been isolated from acorn (Quercus ilex) fruits. The chemical composition of acorn flour showed its richness in carbohydrates (64.43%), proteins (8%), and fat (10%). The extraction yield of acorn starch was about 34.5%. Thus, the composition of extracted acorn starch and its physical and functional properties were studied. Acorn starch had high purity represented by low proportions of proteins (0.92%) and lipids (0.51%) with a pH of 5.3. The swelling power was 20.76 g/g, while the solubility was about 64.22% at 90°C which suggests that acorn starch has potential for use in food industries. The FT-IR spectra of isolated native starches have shown the main bands characterizing the starch. However, X-ray diffractograms exhibited an A- and B-type diffraction pattern. Furthermore, the effect of acorn starch incorporation at different levels (0.5%, 1%, 1.5%, and 2%) on the quality parameters of a fermented dairy product was investigated at the beginning of storage. The results demonstrated that the most suitable dose of acorn starch to be incorporated in the fermented dairy product was lower than 1%. This low concentration reduced syneresis, improved functional properties, and enhanced the viscosity of the fermented dairy product

Various experimental models were used to characterize the antioxidant activities in vitro as well as on ROS-induced fluorescence using dichlorofluorescein technique from phorbol myristate acetate (PMA)-stimulated human myeloid cell line HL-60. Results showed that the antioxidant activities varied considerably with organs. Thus, flower exhibited higher DPPH-scavenging ability, reducing and chelating power than stem and fruit. Also, antioxidant capacities using ORAC method and a cell-based assay showed that fruit and stem exhibited statistically similar antioxidant activities. Moreover, F. communis contains high amounts of flavonoids with various health benefits attributed to their antioxidant potential. Likewise, to obtain biologically relevant information, the antioxidant activities of the extracts were evaluated on cellular models implicating the antioxidant activities; this test generally showed that F. communis flower extracts have the highest antioxidant capacities correlated to the highest total phenolic content. The identification of phenolic compounds in F. communis extracts using RP-HPLC revealed that resorcinol, ferulic, and syringic acids together with coumarin were the major molecules. © 2018 Società Botanica Italian
