    Regulation of AQP0 water permeability is enhanced by cooperativity.

    Aquaporin 0 (AQP0), essential for lens clarity, is a tetrameric protein composed of four identical monomers, each of which has its own water pore. The water permeability of AQP0 expressed in Xenopus laevis oocytes can be approximately doubled by changes in calcium concentration or pH. Although each monomer pore functions as a water channel, under certain conditions the pores act cooperatively. In other words, the tetramer is the functional unit. In this paper, we show that changes in external pH and calcium can induce an increase in water permeability that exhibits either a positive cooperativity switch-like increase in water permeability or an increase in water permeability in which each monomer acts independently and additively. Because the concentrations of calcium and hydrogen ions increase toward the center of the lens, a concentration signal could trigger a regulatory change in AQP0 water permeability. It thus seems plausible that the cooperative modes of water permeability regulation by AQP0 tetramers mediated by decreased pH and elevated calcium are the physiologically important ones in the living lens

    Absolute velocity along the AR7W section in the Labrador Sea

Since 1995, lowered acoustic doppler current profiler (LADCP) data have also been collected. In this work we use data from six of these sections, spanning the time period 1995 through 2008, to determine absolute velocity across AR7W and analyze the main features of the general circulation in the area. We find that absolute velocity fields are characterized by strong, nearly barotropic flows all along the section, meaning there is no “level of no motion” for geostrophic velocity calculations. There is strong variability from year to year, especially in the strength of the boundary currents at each end; nevertheless, combining data from.all 6 sections yields a well-organized velocity field resembling that presented by Pickart and Spall (2007), except that our velocities tend to be stronger: there is a cyclonic boundary current system with offshore recirculations at both ends of the line; the interior is filled with virtually uniform, top-to-bottom bands of velocity with alternating signs. At the southwestern end of the section, the LADCP data reveal a dual core of the Labrador Current at times when horizontal resolution is adequate. At the northeastern end, the location of the recirculation offshore of the boundary current is bimodal, and hence the apparent width of the boundary current is bimodal as well. In the middle of the section, we have found a bottom current carrying overflow waters along the Northwest Atlantic Mid-Ocean Channel, suggesting one of various possible fast routes for those waters to reach the central Labrador Sea. We have used the hydrographic data to compute geostrophic velocities, referenced to the LADCP profiles, as well as to compute ocean heat transport across AR7W for four of our sections. For all but one year, these fluxes are comparable to the mean air–sea heat flux that occurs between AR7W and Davis Strait from December to May (O(50–80 TW)), and much larger than the annual average values (O(10–20 TW)).This material is based upon work supported by the National Science Foundation under Grant No. OCE-0622640.     Unmanned Surface Vehicles for Freighters

    The purpose of this project is to study the technological and commercial feasibility of robotic control systems for freighters, and to develop a miniature proof of concept based on the findings. The research portion of the project involved a detailed technological and financial viability analysis of Unmanned Surface Vehicle (USV) technology for freight shipping, especially for inland waterways. The prototype-engineering portion of the project resulted in a proof of concept control system based on an open-source platform, which controls a miniature Unmanned Surface Vehicle prototype. The prototype is a 1.3-meter long twin-hulled catamaran, which navigates autonomously to ferry a miniature payload using differential thrust from two motors that are powered by two deep-cycle marine batteries. These undertakings have led to a comprehensive solution to enable zero emission inland freight shipping using mass-manufactured swarms of semi-autonomous, 100 DWT, low-draft, multi-hulled, solar ships that carry intermodal cargo containers

    Gemini-South + FLAMINGOS Demonstration Science: Near-Infrared Spectroscopy of the z=5.77 Quasar SDSS J083643.85+005453.3

    We report an infrared 1-1.8 micron (J+H-bands), low-resolution (R=450) spectrogram of the highest-redshift radio-loud quasar currently known, SDSS J083643.85+005453.3, obtained during the spectroscopic commissioning run of the FLAMINGOS multi-object, near-infrared spectrograph at the 8m Gemini-South Observatory. These data show broad emission from both CIV 1549 and CIII] 1909, with strengths comparable to lower-redshift quasar composite spectra. The implication is that there is substantial enrichment of the quasar environment, even at times less than a billion years after the Big Bang. The redshift derived from these features is z = 5.774 +/- 0.003, more accurate and slightly lower than the z = 5.82 reported in the discovery paper based on the partially-absorbed Lyman-alpha emission line. The infrared continuum is significantly redder than lower-redshift quasar composites. Fitting the spectrum from 1.0 to 1.7 microns with a power law f(nu) ~ nu^(-alpha), the derived power law index is alpha = 1.55 compared to the average continuum spectral index = 0.44 derived from the first SDSS composite quasar. Assuming an SMC-like extinction curve, we infer a color excess of E(B-V) = 0.09 +/- 0.01 at the quasar redshift. Only approximately 6% of quasars in the optically-selected Sloan Digital Sky Survey show comparable levels of dust reddening.Comment: 10 pages, 1 figure; to appear in the Astrophysical Journal Letter

    Mindfulness meditation targets transdiagnostic symptoms implicated in stress-related disorders: Understanding relationships between changes in mindfulness, sleep quality, and physical symptoms

    Mindfulness-Based Stress Reduction (MBSR) is an 8-week meditation program known to improve anxiety, depression, and psychological well-being. Other health-related effects, such as sleep quality, are less well established, as are the psychological processes associated with therapeutic change. This prospective, observational study (n=213) aimed to determine whether perseverative cognition, indicated by rumination and intrusive thoughts, and emotion regulation, measured by avoidance, thought suppression, emotion suppression, and cognitive reappraisal, partly accounted for the hypothesized relationship between changes in mindfulness and two health-related outcomes: sleep quality and stress-related physical symptoms. As expected, increased mindfulness following the MBSR program was directly correlated with decreased sleep disturbance (r=-0.21, p=0.004) and decreased stress-related physical symptoms (r=-0.38, p<0.001). Partial correlations revealed that pre-post changes in rumination, unwanted intrusive thoughts, thought suppression, experiential avoidance, emotion suppression, and cognitive reappraisal each uniquely accounted for up to 32% of the correlation between the change in mindfulness and change in sleep disturbance and up to 30% of the correlation between the change in mindfulness and change in stress-related physical symptoms. Results suggest that the stress-reducing effects of MBSR are due, in part, to improvements in perseverative cognition and emotion regulation, two “transdiagnostic” mental processes that cut across stress-related disorders
