Immunolocalization of neurokinin 1 receptor in WHO grade 4 astrocytomas, oral squamous cell and urothelial carcinoma
Neurokinin-1 receptor (NK-1R) induces inflammatory reactions in peripheral tissues but its regulatory effects in target tissues is dependent on receptor signalling. Substance P (SP) has a high affinity for the NK-1R, to which it binds preferentially. We aimed to investigate the expression of NK-1R in World Health Organization (WHO) grade 4 astrocytomas as well as in oral squamous cell carcinoma (OSCC) and urothelial carcinoma, and its association with disease progression.The study included tissue samples from 19 brain astrocytomas, 40 OSCCs and 10 urothelial carcinomas. NK-1R expression was quantitatively assessed in the tumour cells using immunohistochemistry. The relationship between NK-1R expression in astrocytomas and recurrence-free interval has been explored.The results showed that the NK-1R was intensely expressed in patients with WHO grade 4 astrocytoma, OSCC and urothelial carcinoma. However, cases clinically diagnosed as a low-grade cancer showed reduced NK-1R expression.NK-1R is overexpressed in all cases of WHO grade 4 astrocytoma, OSCC and urothelial carcinoma. The ubi-quitous presence of SP/NK-1R complex during tumour development and progression suggests a possible therapeutic key strategy to use NK-1R antagonist as an adjuvant therapy in the future
The effect of using nano agriculture wastes on microstructure and electrochemical performance of ultra-high-performance fiber reinforced self-compacting concrete under normal and acceleration conditions
This research investigates the effect of using nano agriculture waste as a partial substitute to solve increased climatic concerns in addition to producing modern ultra-high-performance fiber reinforced self-compacting concrete (UHPFC-SCC). Three types of nano materials have been incorporated, namely nano sugar cane bagasse ash (NSCBA), nano cotton stalk ash (NCSA), and nano rice straw ash (NRSA) with dosages of 1%, 2%, and 3%, respectively. Fresh concrete properties have been investigated via flow and V-funnel tests while compressive strength was evaluated up to 90 days. X-ray diffraction (XRD) analysis, Raman spectroscopy analysis and scanning electron microscopy (SEM) alongside energy dispersive spectroscopy (EDS) was performed to compliment the strength data. Finally, electrochemical measurements including linear polarization (LPR), open circuit potential (OCP) and impedance spectroscopy were investigated at normal and accelerated corrosion conditions on HSS (high strength steel Gr 60). Results demonstrate that compressive strength improved in the range of 18%â 21% between 28 and 90 days using 3% NSCBA, 3% NRSA or 1% NCSA. This is attributed to the facts that optimum dosage of nano wastes results in densification of the UHPFC-SCC matrix and strength magnitude depended on intertwining of high Ca/Si molar content in the range of 3.14â7.1 compared to 0.52 observed in the control mix (EDS analysis). Further, electrochemical measurements revealed that nano materials improved the charge transfer resistance and bulk resistance of HSS interface as well as retarded the flow of electrons between anode and cathode sites consequently limited the propagation of corrosion
Assessment of prognostic value of tissue inhibitors of metalloproteinase 3 (TIMP3) protein in ovarian cancer
Background: TIMP3 is a multifunctional proteolytic enzyme belonging to TIMPs family and acts as a potent inhibitor of matrix metalloproteinases (MMPs). TIMP3 possesses a tumor suppresive function by directly promoting tumor cell apoptosis, preventing angiogenesis and extracellular matrix remodelling. The lower expression of TIMP3 was associated with poor prognosis and overall survival in various cancer types. The aim of this study was to evaluate the association of TIMP3 protein expression with ovarian cancer (OC) clinicopathological features and survival outcomes.
Patients and Methods: One hundred forty four of OC FFPE samples were collected from King Abdulaziz University Hospital, Saudi Arabia and constructed in tissue microarray (TMA) slides. Automated Ventana immunostainer platform was used to evaluate TIMP3 protein expression patterns.
Results: The study showed that TIMP3 exhibits cytoplasmic localisation. This TIMP3 protein expres- sion was not associated with age, tumor size and the involvement of lymph nodes (p > 0.05). However, it was significantly correlated with tumor stage (p < 0.05) and borderline significant with endpoint status (p = 0.07). Interestingly, the Kaplan-Meier analysis of disease specific survival (DSS) outcomes showed a significant association (p = 0.02, log rank) between OC patients with higher TIMP3 expression compared to those with lower expression. In fact, OC patients with high TIMP3 expression had longer survivals. Multivariate Coxâs regression analysis suggests that low TIMP3 protein expression pattern is an independent poor survival marker (p = 0.025).
Conclusion: Cytoplasmic TIMP3 protein expression could be used as a good prognosticator to stratify poorly prognostic OC patients in order to personlaize their disease management
The Potential Effect of the IDH1 Mutation and MGMT Gene Promoter Methylation on the Control of Glioblastoma-Associated Epilepsy in Patients Receiving Anti-Epileptic Agents and Chemotherapies
Objectives:(a) The objective of the study was to assess the control of seizure in glioblastoma patients receiving anti-epileptic drugs and chemotherapies after total resection and its association with O-methylguanine-DNA methyltransferase (MGMT) promoter methylation and the isocitrate dehydrogenase 1 (IDH1) mutation; (b) to determine which anti-epileptic drug exerts the best effective control on glioblastoma-associated epilepsy; and (c) to identify the relationship between seizure control and anti-epileptic drugs with recurrence interval.Methods:This was a retrospective cohort study of patients with postoperative glioblastoma-associated epilepsy. The correlation between IDH1 mutation and MGMT methylation with anti-epileptic drugs, chemotherapy type, seizure control, and recurrence interval was analyzed.Results:The study included 53 patients with glioblastoma-associated epilepsy. IDH1 mutation was present in 20 patients, and MGMT methylation was present in 13 patients. 37 cases received chemoradiotherapy while 16 cases received only radiotherapy. Levetiracetam was the most prescribed anti-epileptic drug (n=36, 60%), and 36 and 16 patients had controlled and uncontrolled seizures, respectively. IDH1 mutation and unmethylated MGMT were significantly present in cases with controlled epilepsy (p<0.05). Levetiracetam showed significantly better seizure control in cases with IDH1 mutation and unmethylated MGMT promotor (p<0.05).Conclusion:(a) Glioblastoma-associated epilepsy can be better controlled in patients with the IDH1 mutation and unmethylated MGMT, (b) levetiracetam was the first-line anti-epileptic drug for controlling seizure, (c) lack of seizure control in glioblastoma patients may not be related to tumor recurrence despite 1-year treatment, and (d) better understanding of the risk factors associated with glioma-associated epilepsy are needed to improve patient quality of life
Comprehensive Analysis of Genes Associated With Sudden Infant Death Syndrome
Background: Sudden infant death syndrome (SIDS) is a tragic incident which remains a mystery even after post-mortem investigation and thorough researches. Methods: This comprehensive review is based on the genes reported in the molecular autopsy studies conducted on SIDS so far. A total of 20 original studies and 7 case reports were identified and included in this analysis. The genes identified in children or adults were not included. Most of the genes reported in these studies belonged to cardiac channel and cardiomyopathy. Cardiac channel genes in SIDS were scrutinized for further analysis. Results: After screening and removing the duplicates, 42 unique genes were extracted. When the location of these genes was assessed, it was observed that most of these belonged to Chromosomes 11, 1 and 3 in sequential manner. The pathway analysis shows that these genes are involved in the regulation of heart rate, action potential, cardiac muscle cell contraction and heart contraction. The protein-protein interaction network was also very big and highly interactive. SCN5A, CAV3, ALG10B, AKAP9 and many more were mainly found in these cases and were regulated by many transcription factors such as MYOG C2C1 and CBX3 HCT11. Micro RNA, hsa-miR-133a-3p was found to be prevalent in the targeted genes. Conclusions: Molecular and computational approaches are a step forward toward exploration of these sad demises. It is so far a new arena but seems promising to dig out the genetic cause of SIDS in the years to come
