    An investigation effect of seaweeds as shrimp -feed ingredient for growth performance of Litopenaeus vannamei

    In this project, the nutritional effects of Sargassum illicifolium Chabahar bay-Oman Sea, on growth and survival rates of whiteleg shrimp (Litopenaeus vannamei) were studied. The seaweed collected from 6 coastal area, rinsed, dried, powdered and measured the nutritional values in laboratory for surveying statistically. According to the high nutritional value of Tis coastal seaweed, this variate seaweed powder, replaced with protein resources (fish meal and Soy and Wheat) of whiteleg shrimp feed which was formulated by Havorash feed factory of Boshehr in four treatments (A: as control without any replacement) B: with 5%, C: 10 % and D: 15% seaweed replacement, each with three replicates in order to obtain isonitrogenus 33% CP., and Isocaloric (13% fat and 15% carbohydrate) feed. The weighed milled ingredients were carefully mixed using a laboratory food mixer. The mixtures were primed with 30% hot water to yield a suitable pulp. Wet diets were made into 2 mm pellet size and dried at 40 °C in a drying cabinet and maintained in standard condition which was used according to daily need shrimp, calculated by each 10 days biometry. Water stability and absorbtion capacity of the pellets in sea water were measured and compared statistically. Juvenile shrimps (Initial body weight =3 g) brought from Jask hatchery, acclimazed for one week in Chabahar hatchery condition and feeding daily 3-5% body weight. Abiotical parameters and weight and length biometrics were measured two days and 10 days, respectively. After 45 days and final biometry, FCR, CF, SGR, caracas analysis, muscle colourimetery with HPLC were done, tasted with pp Plot for determining the parametric data and statistically differences using one – way ANOVA, Duncan test of SPSS software. The Tis coastal seaweed with 9.8% CP, 2% lipid and 23% carbohydrate had higher nutritional value compared to the other gathered seaweed. Also amino acid and fatty acid profiles, vitamins and minerals were measured in all seaweed samples each, with three replications. As result, the water stability of D feed treatment in seawater (98%) and C (97%) had statistical differences with A and B (95% stability) (P0.05) but Cholestrol content of, showed differences between all, significantly (P<0.05) which was the highest (121.68±12.12) in D and the lowest in A (147.92±11.02). Feed treatment D and C performed colour changes pink partial orang and pink in shrimp muscle with no any difference compared to white and none colour in shrimp were fed B and A feed treatments . It seems this colour changing can be playing a major role in market acceptability

    Fish gene bank of the Persian Gulf and Oman Sea

    Genetic studies and gene banks preparation can identify guidelines for improving diversity and population structure and estimation, poaching and also the amount of cross breeding and provide genetic classification. In this study, sampling was performed from the important studied species habitats areas for the species such as: Dasyatis bennetti، Netuma thalassina, Netuma bilineata Carcharhinus leucas، Choerodon robustus، Pseudorhombus pentophthalmus ،Pseudorhombus arsius Thunnus tonggol ،Euthynnus affinis and Trichiurus lepturus. Total DNA extraction was performed using phenol - chloroform method which is the most common method for DNA extraction in order to achieve high quality of DNA was performed in the preparation of gene bank in this study. After relevant studies on this gene primers were designed and in use. After editing the sequences, nucleotide BLAST (Basic Local Alignment Search Tool) was performed using NCBI blast main page. The sequences obtained from each sample were aligned and corrected from any ambiguities and assembled using Bio edit program .Trees were generated using maximum parsimony (MP), a character-based algorithm and neighbor joining (NJ) a distance-based algorithm for phenetic analysis. The distance matrix option of MEGA4 was used to calculate genetic distance according to the Kimura 2-parameter model of sequence evolution. Based on the results obtained, the optical density of 260 to 280 nm in the samples was recorded between 1/8 - 2, indicating good quality DNA samples. Optimized PCR reaction to 16SrRNA gene amplification using the gradient between 48 - 60° C showed that the most suitable criteria for binding primers, 54 to 58 Celsius degrees respectively. The project objectives including the identification of the genetic structure of the species, and draw the phylogenetic trees using two genes 16SrRNA, making identification and registration of specified computer storage and regulate the structure and management of mentioned species by focus on genetic resources 10 species of majour commercial and noncommercial fishes in the Persia Gulf and Oman Sea through the creation of an integrated network of aquatic genetic resources in the region to try to identify genetic resources and aquatic gene bank. This type of analysis could be considered as an important tool to be used in broodstock selection in breeding programs. Also identify the species of fish named as Pseudorhombus pentophthalmus with a new haplotype and genetic differences 7% in compare to the samples recorded in the World gene Bank can considered as a major achievements of this research

    Background: The aim of this study was to evaluate the anticancer potential of Kelussia odoratissima. Several in vitro and in vivo biological assays were applied to explore the direct effect of an extract and bioactive compound of this plant against breast cancer cells and its possible mechanism of action. Materials and methods: K. odoratissima methanol extract (KME) was prepared, and MTT assay was used to evaluate the cytotoxicity. To identify the cytotoxic compound, a bioassay-guided investigation was performed on methanol extract. 8-Hydroxy-ar-turmerone was isolated as a bioactive compound. In vivo study was performed in the breast cancer rat model. LA7 cell line was used to induce the breast tumor. Histopathological and expression changes of PCNA, Bcl-2, Bax, p27 and p21 and caspase-3 were examined. The induction of apoptosis was tested using Annexin V-fluorescein isothiocyanate (FITC) assay. To confirm the intrinsic pathway of apoptosis, caspase-7 and caspase-9 assays were utilized. In addition, cell cycle arrest was evaluated. Results: Our results demonstrated that K. odoratissima has an obvious effect on the arrest of proliferation of cancer cells. It induced apoptosis, transduced the cell death signals, decreased the threshold of mitochondrial membrane potential (MMP), upregulated Bax and downregulated Bcl-2. Conclusion: This study demonstrated that K. odoratissima exhibits antitumor activity against breast cancer cells via cell death and cell cycle arrest

    The current study investigated the cytotoxic effect of 3-(5-chloro-2-hydroxybenzylideneamino)-2-(5-chloro-2-hydroxyphenyl)-2,3-dihydroquinazolin-41(H)-one (A) and 3-(5-nitro-2-hydroxybenzylideneamino)-2-(5-nitro-2-hydroxyphenyl)-2,3-dihydroquinazolin-4(1H)-one (B) on MCF-7, MDA-MB-231, MCF-10A and WRL-68 cells. The mechanism involved in apoptosis was assessed to evaluate the possible pathways induced by compound A and B. MTT assay results using A and B showed significant inhibition of MCF-7 cell viability, with IC 50 values of 3.27±0.171 and 4.36±0.219μg/mL, respectively, after a 72hour treatment period. Compound A and B did not demonstrate significant cytotoxic effects towards MDA-MB-231, WRL-68 and MCF-10A cells. Acute toxicity tests also revealed an absence of toxic effects on mice. Fluorescent microscopic studies confirmed distinct morphological changes (membrane blebbing and chromosome condensation) corresponding to typical apoptotic features in treated MCF-7 cells. Using Cellomics High Content Screening (HCS), we found that compound A and B could trigger the release of cytochrome c from mitochondria to the cytosol. The release of cytochrome c activated the expression of caspases-9 and then stimulated downstream executioner caspase-3/7. In addition, caspase-8 showed remarkable activity, followed by inhibition of NF-κ B activation in A-and B-treated MCF-7 cells. The results indicated that A and B could induce apoptosis via a mechanism that involves either extrinsic or intrinsic pathways

After 1 hour, the normal group was orally administered 10% Tween 20, whereas absolute alcohol was fed orally to ulcerated control, omeprazole, and experimental groups. Gastric&rsquo;s homogenate were assessed for endogenous enzymes activities. Stomachs were examined macroscopically and histologically. Grossly, the data demonstrated a significant decrease in the ulcer area of rats pretreated with plant extract in a dose-dependent manner with respect to the ulcerated group. Homogenates of the gastric tissue exhibited significantly increased endogenous enzymes activities in rats pretreated with C. barometz extract associated with the ulcerated control group. Histology of rats pretreated with C. barometz extract group using hematoxylin and eosin staining exhibited a moderate-to-mild disruption of the surface epithelium with reduction in submucosal edema and leucocyte infiltration in a dose-dependent manner. In addition, it showed heat shock protein70 protein up-expression and BCL2-associated X protein downexpression.
