On bootstrap sample size in extreme value theory

Nylon 46-polytetramethylene oxide segmented block copolymers
Block copolymers were synthesized from amine-terminated polytetramethylene oxide (PMTO) (Mw 800 and 1130) and polyamide 4,6 salt. First prepolymers were prepared at 200–210°C in the presence of a solvent (pyrrolidone). The prepolymers were postcondensed at 255°C (where possible in the solid state) to a high molecular weight. In the materials the endgroups were analysed, and the inherent viscosities and ethanol extraction values measured. The thermal properties were determined with d.s.c. and d.m.a. With ethanol, polyether could be extracted. Thus it seems that not all the polyether is present as part of the block copolymer. Melt phasing seems to have taken place and occurs even more with the higher molecular weight PTMO. With d.m.a. two glass transitions were observed; one of the polyether phase and one of the polyamide-rich phase. The melting temperatures of these polyamide 4,6 block copolymers are high (200–270°C), and the torsion modulus remained fairly constant up to these high melting temperatures
Copolymers of PBT and nylon 4T
Polybutylene terephthalate-nylon 4T copolymers (PBT-PA 4T) are synthesized from the diamide of diaminobutane and dimethyl terephthalate (DMT) with butane diol and more DMT in a concentration range of up to 50% PA 4T. The polymerization conditions were similar to those for PBT: first, a melt polymerization, followed by solid-state post-condensation. The materials were studied by differential scanning analysis (DSC) (melting and crystallization behavior) and dynamic mechanical thermal analysis (DMTA) (glass transitions and torsion moduli). The water absorptions were determined at 100% RH. By increasing the PA 4T content in the copolymers, melting temperatures increased strongly, heats of fusion decreased slightly, and glass transition temperatures increased linearly. Weak & Strong Financial Fragility
The stability of the financial system at higher loss levels is either characterized by asymptotic dependence or asymptotic independence. If asymptotically independent, the dependency, when present, eventually dies out completely at the more extreme quantiles, as in case of the multivariate normal distribution. Given that financial service firms' equity returns depend linearly on the risk drivers, we show that the marginals' distributions maximum domain of attraction determines the type of systemic (in-)stability. A scale for the amount of dependency at high loss lovels is designed. This permits a characterization of systemic risk inherent to different financial network structures. The theory also suggests the functional form of the economically relevant limit copulas

EEHV1A glycoprotein B subunit vaccine elicits humoral and cell-mediated immune responses in mice
Asian elephants are an endangered species facing many threats, including severe hemorrhagic disease (HD) caused by the elephant endotheliotropic herpesvirus (EEHV). EEHV-HD is the leading cause of death in captive juvenile Asian elephants in North America and Europe, and also affects elephants in their natural range countries. Significant challenges exist for successful treatment of EEHV-HD, which include timely recognition of disease onset and limited availability of highly effective treatment options. To address this problem, our goal is to prevent lethal disease in young elephants by developing a vaccine that elicits robust and durable humoral and cell-mediated immunity against EEHV. EEHV glycoprotein B (gB) is a major target for cellular and humoral immunity in elephants previously exposed to EEHV. Therefore, we generated a vaccine containing recombinant EEHV1A gB together with a liposome formulated TLR-4 and saponin combination adjuvant (SLA-LSQ). CD-1 mice that received one or two vaccinations with the vaccine elicited significant anti-gB antibody and polyfunctional CD4+ and CD8+ T cell responses, while no adverse effects of vaccination were observed. Overall, our findings demonstrate that an adjuvanted gB protein subunit vaccine stimulates robust humoral and cell-mediated immune responses and supports its potential use in elephants
First-in-human administration of a live-attenuated RSV vaccine lacking the G-protein assessing safety, tolerability, shedding and immunogenicity: a randomized controlled trial
Background: Human respiratory syncytial virus (RSV) is a major cause of lower respiratory tract infections in early infancy and in elderly. A pediatric vaccine against RSV would not only prevent morbidity and mortality amongst infants and young children but could also reduce transmission to elderly. The RSVDG vaccine consists of a live-attenuated RSV that lacks the G attachment protein. RSVDG is severely impaired in binding to host cells and exhibits reduced infectivity in preclinical studies. Intranasal immunization of cotton rats with RSVDG vaccine protected against replication of wildtype RSV, without inducing enhanced disease.Methods: We performed a first-in-human trial with primary objective to evaluate safety and shedding of RSV Delta G (6.5 log(10) CCID50) after intranasal administration. Healthy adults aged between 18 and 50, with RSV neutralizing serum titers below 9.6 log(2), received a single dose of either vaccine or placebo (n = 48, ratio 3:1). In addition to safety and tolerability, nasal viral load, and systemic and humoral immune responses were assessed at selected time points until 4 weeks after immunization.Results: Intranasal administration of RSV Delta G was well tolerated with no findings of clinical concern. No infectious virus was detected in nasal wash samples. Similar to other live-attenuated RSV vaccines, neutralizing antibody response following inoculation was limited in seropositive adults.Conclusions: A single dose of 6.5 log(10) CCID50 of RSV Delta G was safe and well-tolerated in seropositive healthy adults. RSV Delta G was sufficiently attenuated but there were no signs of induction of antibodies. Safety and immunogenicity can now be explored in children and eventually in seronegative infants.
