    In vitro performances of hypocotyl and cotyledon explants of tomato cultivars under sodium chloride stress

    A plant tissue culture technique is a good method for the evaluation and screening of plant genotypes for salt tolerance. In vitro evaluations of sodium chloride (NaCl) effects on two tomato cultivars (Pearl and Beril) were investigated with four NaCl levels (0, 25, 50 and 75 mM) using hypocotyl and cotyledon explants. The explants were cultured in MS media having 2.0 mg/l BAP along with different concentrations of NaCl. Sodium chloride stress negatively affected the growth traits and chlorophyll content. Significant differences were noticed between the cultivars followed by different NaCl levels, where the Beril responded superior than that of Pearl. The type of explant showed a difference in their response to shoots regeneration under NaCl stress, where the cotyledon explants achieved best results than hypocotyl explants.Key words: Cotyledons, hypocotyls, In vitro, salt stress, tomato

    Bone-specific median age of hand-wrist maturation from Sudan

    Background Maturation of bones in the hand–wrist region varies among individuals of the same age and among world groups. Although some studies from Africa report differences to other ethnic groups, the lack of detailed bone-specific maturity data prevents meaningful comparisons. Aim The aim of this study was to describe bone-specific maturity for developing hand–wrist bones in individuals in Khartoum, Sudan. Subjects and methods The sample was selected from healthy patients attending a dental hospital in Khartoum with known age and ancestry (males = 280, females = 330; aged between 3 and 25 years). Bones were assessed from radiographs of the left hand and wrist after the Greulich and Pyle Atlas (1959). Median ages of attainment for bone stages were calculated using probit analysis for each stage in males and females separately. Results Maturity data for stages of the phalanges, metacarpals, carpals and radius and ulna in males and females are presented. Median ages in females were earlier compared to males for all stages. These results are largely earlier than previously published findings or where these could be calculated. Conclusion These results of individual maturity stages of phalanges, metacarpals, carpals and the distal epiphyses of the radius and ulna are useful to assess maturity in growing individuals from Sudan

    Prognostic value of FOXP3 and TGF-b expression in both peripheral blood and lymph nodes in patients with B-Non Hodgkin’s lymphoma

    Foxp3 has been studied as a biomarker of Treg cells in many solid malignant diseases, although its role as an immunomodulator in B-NHL remain poorly understood and the effect of traditional chemotherapy on its expression remains unclear. In this study the role of circulating and intra-tumoral Treg and TGF-b in patients with B-NHL before and after chemotherapy was evaluated. Enumeration of Treg cells was carried out by flow cytometric staining of their cell surface markers CD4 and CD25 as well as by molecular analysis of its signature transcription factor FoxP3. Expression of FoxP3 was done using quantitative real-time PCR while TGF-b mRNA expression was semi-quantitatively assayed by the conventional reverse transcription-PCR. In addition, spontaneous versus mitogen-induced release of TGF-b by PBMCs was assessed by a short term cell culture followed by ELISA. This was done before and after six cycles of CHOP chemotherapy. The results were evaluated in relation to the clinicopathological data.A significant increase in mRNA transcripts of both Fox P3 and TGF-b as well as the percentage of CD4+/CD25+ in B-NHL patients before receiving the chemotherapy were recorded, when compared either to healthy controls or to patients after completion the treatment regimen. Interestingly 6 cycles of CHOP treatment caused significant reduction in all parameters under study, relative to the situation before treatment. A significant enhancement in spontaneous TGF-b release in B-NHL patient either before or after chemotherapy was obtained. These results strongly confirm the possible involvement of Treg cells and TGF-b in orienting the clinical course of the disease as well as the ability of targeting them in immunotherapeutic approaches. KEYWORDS FOXP3; TGF; Non Hodgkin’s lymphoma; NH

    Background: Inflammatory bowel disease (IBD) is a condition in which the gastrointestinal system becomes inflamed for no apparent reason. Its incidence increased during the last few decades. As a result, it's critical to understand how to validate a clinical diagnosis and monitor any progress using a simple procedure. Osteoprotegerin (OPG), which interacts with RANKL, regulates RANK/RANKL signaling. OPG is a decoy receptor for soluble receptor activator of nuclear factor-B ligand (sRANKL), which stimulates nuclear factor kappa-light-chain-enhancer of activated B cells (NF-ÎşB) and the healing trefoil factor 3 (TFF3), which could be employed as in the diagnosis and follow-up of IBD patients, biomarkers are used. Objectives: The aim of the current work was to evaluate the role of RANKL (sRANKL), OPG, NF-ÎşB , and TFF3 in in the diagnosis of active IBD. Subjects and methods: Thirty-five newly diagnosed untreated IBD patients and thirty-five healthy controls were included in this study. Patients were then given particular treatment either in the form of cortisone, mesalamin, immunotherapy or infliximab and immunotherapy for 3 months and attend for second visit. OPG, sRANKL, NF-ÎşB and TFF3 levels were estimated by ELISA method using commercial kits in patients before and after treatment and healthy control. Results: Patients with IBD had significantly increased levels of OPG, sRANKL, NF-B, and TFF3 than healthy controls (P =0.0001). After 3 months of treatment, patients' levels of NF-B and TFF3 were significantly lower (p = 0.0001 and 0.03, respectively). Conclusion: The biomarkers OPG, sRANKL, NF-ÎşB , and TFF3 show promise in the diagnosis of active IBD. The decrease in NF-ÎşB and TFF3 following treatment suggests that be used as biomarkers to track disease activity and treatment effectiveness

    Clinical features of culture-proven Mycoplasma pneumoniae infections at King Abdulaziz University Hospital, Jeddah, Saudi Arabia

    OBJECTIVE: This retrospective chart review describes the epidemiology and clinical features of 40 patients with culture-proven Mycoplasma pneumoniae infections at King Abdulaziz University Hospital, Jeddah, Saudi Arabia. METHODS: Patients with positive M. pneumoniae cultures from respiratory specimens from January 1997 through December 1998 were identified through the Microbiology records. Charts of patients were reviewed. RESULTS: 40 patients were identified, 33 (82.5%) of whom required admission. Most infections (92.5%) were community-acquired. The infection affected all age groups but was most common in infants (32.5%) and pre-school children (22.5%). It occurred year-round but was most common in the fall (35%) and spring (30%). More than three-quarters of patients (77.5%) had comorbidities. Twenty-four isolates (60%) were associated with pneumonia, 14 (35%) with upper respiratory tract infections, and 2 (5%) with bronchiolitis. Cough (82.5%), fever (75%), and malaise (58.8%) were the most common symptoms, and crepitations (60%), and wheezes (40%) were the most common signs. Most patients with pneumonia had crepitations (79.2%) but only 25% had bronchial breathing. Immunocompromised patients were more likely than non-immunocompromised patients to present with pneumonia (8/9 versus 16/31, P = 0.05). Of the 24 patients with pneumonia, 14 (58.3%) had uneventful recovery, 4 (16.7%) recovered following some complications, 3 (12.5%) died because of M pneumoniae infection, and 3 (12.5%) died due to underlying comorbidities. The 3 patients who died of M pneumoniae pneumonia had other comorbidities. CONCLUSION: our results were similar to published data except for the finding that infections were more common in infants and preschool children and that the mortality rate of pneumonia in patients with comorbidities was high

    Natural Plant Extracts and Microbial Antagonists to Control Fungal Pathogens and Improve the Productivity of Zucchini (Cucurbita pepo L.) In Vitro and in Greenhouse

Methods: The purpose of this study was to evaluate the ability of both natural plant extracts and microbial antagonists as a biotical control of some fungal pathogens, i.e., Fusarium ssp., Exserohilum ssp. and Nigrospora ssp., along with improving the growth and productivity performance of zucchini under greenhouse conditions. Eucalyptus camaldulensis leaf extract (LE), Citrus sinensis LE, Ficus benghalensis fruit extract (FE), and two microbial antagonists Pseudomonas fluorescens (accession no. MW647093) and Trichoderma viride (accession no. MW647090) were tested under in vitro and in vivo conditions. Through morphological characteristics and the internal transcribed spacer (ITS) region, Fusarium solani (accession no. MW947256), F. oxysporum (accession no. MW947254), Exserohilum rostratum (accession no. MW947255), and Nigrospora lacticolonia (accession no. MW947253) were identified. HPLC analysis was used for the identification of phenolic compounds (PCs) and flavonoid compounds (FCs) in the extracts. Results: The highest inhibition percentage of fungal growth (IPFG) against F. oxysporum was obtained with P. fluorescens, T. viride, and E. camaldulensis LE (4000 mg/L); F. solani with P. fluorescens, T. viride, and C. sinensis LE (4000 mg/L); Exserohilum rostratum with P. fluorescens, Ficus benghalensis FE (4000 mg/L) and E. camaldulensis LE (4000 mg/L), and N. lacticolonia with P. fluorescens. Using HPLC analysis, the abundant PCs in E. camaldulensis LE were pyrogallol, and caffeic acid, those in C. sinensis LE were syringic acid and ferulic acid, and those in F. benghalensis FE were gallic acid and syringic acid. In addition, the abundant FCs in E. camaldulensis LE were kaempferol, and naringin, those in C. sinensis LE were hesperidin and quercetin, and those in F. benghalensis FE were kaempferol and quercetin. Under greenhouse experiments, T. viride and E. camaldulensis LE (4000 mg/L) followed by P. fluorescens + T. viride treatments gave the best results of zucchini plants in terms of leaf area, fruits number per plant, yield per plant, and total yield (marketable and non-marketable).
