    Acetylene reduction associated with zostera novazelandica Setch. and Spartina alterniflora Loisel., in Whangateau harbour, North Island, New Zealand

    Nitrogen fixation (acetylene reduction) was investigated in Zostera novazelandica Setch. and Spartina alterniflora Loisel., in the North Island of New Zealand. Moderate rates of acetylene reduction were found in sediments in which plants were growing (means Ā± 95% confidence limit: 15.2 Ā± 2.8 |jmol C2H4 nrr2 h~l for Zostera and 24.7 Ā± 4.6 |amol C2H4 m~2 h"1 for Spartina). Activity was closely correlated with the dry weight of root; (r2 = 0.65, N = 15 for Zostera, and r2 = 0.85, A' =10 for Spartina). Sediment close to the plant beds, but without plants, exhibited only low rates of acetylene reduction (2.9 Ā± 2.2 and 4.5 Ā± l.OjLtmol C2H4 m~2 h"1, respectively). Sediments associated with Z. novazelandica and S. alterniflora in New Zealand exhibit moderate rates of nitrogenase activity compared to rates found in other countries. N fixation may contribute significantly to the nutrition of these plants in New Zealand estuaries

    In October 1987, NSF, NASA, and NOAA jointly sponsored a workshop at Columbia University to assess the experimental tools and analysis procedures in use and under development to measure and understand gas and particle fluxes across this critical air-surface boundary. Results are presented for that workshop. It is published to summarize the present understanding of the various measurement techniques that are available, identify promising new technological developments for improved measurements, and stimulate thinking about this important measurement challenge

    Nitrogenase activity associated with codium species from New Zealand marine habitats

    Nitrogenase activity, measured as acetylene reduction, was recorded at rates up to 1028 nmol.h \g * dry weight for Codium adhaerens (Cabr.) Ag. var. convolutum Dellow and Codium fragile (Sur.) Hariot subsp. tomentosoides (Van Goor) Silva collected from New Zealand habitats. In both species the ability to reduce acetylene is invariably associated with the presence of a heterocystous blue-green alga, Calothrix sp., epiphytic or embedded in the Codium thallus. A highly significant (P < 0.001) correlation between heterocyst frequency and nitrogenase activity was found. Nitrogenase and net photosynthesis of the Codium-Calothrix system have different steady-state responses to light intensity, and the kinetics of the two processes also differ in that nitrogenase is slow to respond to illumination or darkening. Glucose additions to Codium did not significantly increase nitrogenase activity. Nitrogenase is relatively insensitive to oxygen tension over the range 0-1.0 atm (0-1.033 kgf.cnT2) and still occurs at 1.5 atm (1.55 kgf.cm"2); this condition is unique in all nitrogenase systems thus far reported. Collectively these facts suggest that Calothrix is the agent primarily responsible for nitrogenase activity in these Codium species

    Nitric oxide formation in gas turbine engines: A theoretical and experimental study

    A modified Zeldovich kinetic scheme was used to predict nitric oxide formation in the burned gases. Nonuniformities in fuel-air ratio in the primary zone were accounted for by a distribution of fuel-air ratios. This was followed by one or more dilution zones in which a Monte Carlo calculation was employed to follow the mixing and dilution processes. Predictions of NOX emissions were compared with various available experimental data, and satisfactory agreement was achieved. In particular, the model is applied to the NASA swirl-can modular combustor. The operating characteristics of this combustor which can be inferred from the modeling predictions are described. Parametric studies are presented which examine the influence of the modeling parameters on the NOX emission level. A series of flow visualization experiments demonstrates the fuel droplet breakup and turbulent recirculation processes. A tracer experiment quantitatively follows the jets from the swirler as they move downstream and entrain surrounding gases. Techniques were developed for calculating both fuel-air ratio and degree of nonuniformity from measurements of CO2, CO, O2, and hydrocarbons. A burning experiment made use of these techniques to map out the flow field in terms of local equivalence ratio and mixture nonuniformity

    Purification of three catalase isozymes from facultatively alkaliphilic Bacillus firmus OF4

    AbstractCell extracts of facultatively alkaliphilic B. firmus OF4 were assayed for catalase activity and their catalase isozyme content was analyzed on native polyacrylamide gels stained for catalase activity. pH-10.5-grown cells had about twice the specific catalase activity of pH-7.5-grown cells. The higher activity, however, did not confer resistance to exogenous hydrogen peroxide challenge relative to pH-7.5-grown cells and, in fact, the pH-10.5-grown cells were much more sensitive to the challenge. Electrophoresis resolved three catalase isozymes in cell extracts. The isozymes, labeled Iā€“III in order of decreasing electrophoretic mobility, were purified and their Nterminal amino acid sequences were obtained. Isozyme III corresponded to the product of a cloned gene fragment that had been shown to possess substantial sequence similarity to the KatE (HP-II) catalase of E. coli (Quirk, P.G., Krulwich, T.A. and Hicks, D.B. (1993) Biophys. J. 64, 164A) and which had similar biochemical properties to HP-II, i.e., it was a chlorin-containing enzyme expressed only in stationary phase. Isozyme II, a protoheme enzyme, was responsible for the higher activity of alkaline-grown cells and was induced in cells treated with hydrogen peroxide or ascorbate. It showed sequence similarity to katA of Bacillus subtilis (Bol, D. and Yasbin, R. (1991) Gene 109, 31ā€“37). Isozyme I was the only isozyme that exhibited detectable levels of peroxidase activity in addition to catalase activity, resembling a catalase enzyme purified from a different alkaliphile, Bacillus YN-2000 (Yumoto, I., Fukumori, Y. and Yamanaka, T. (1990) J. Biochem. 108, 583ā€“587), to which it showed some sequence similarity
