    Role of Tyk-2 in Th9 and Th17 cells in allergic asthma.

    In a murine model of allergic asthma, we found that Tyk-2((-/-)) asthmatic mice have induced peribronchial collagen deposition, mucosal type mast cells in the lung, IRF4 and hyperproliferative lung Th2 CD4(+) effector T cells over-expressing IL-3, IL-4, IL-5, IL-10 and IL-13. We also observed increased Th9 cells expressing IL-9 and IL-10 as well as T helper cells expressing IL-6, IL-10 and IL-21 with a defect in IL-17A and IL-17F production. This T helper phenotype was accompanied by increased SOCS3 in the lung of Tyk-2 deficient asthmatic mice. Finally, in vivo treatment with rIL-17A inhibited local CD4(+)CD25(+)Foxp3(+) T regulatory cells as well as Th2 cytokines without affecting IL-9 in the lung. These results suggest a role of Tyk-2 in different subsets of T helper cells mediated by SOCS3 regulation that is relevant for the treatment of asthma, cancer and autoimmune diseases

    IL-6 plays a central role in supporting pathological TH2 and TH17 cell development and inhibiting the protective T regulatory cells in allergic asthma. TH17 cells have been demonstrated to regulate allergic asthma in general and T-bet-deficiency-induced asthma in particular. Here we found an inverse correlation between T-bet and Il-6 mRNA expression in asthmatic children. Moreover, experimental subcutaneous immunotherapy (SIT) in T-bet((-/-)) mice inhibited IL-6, IL-21R and lung TH17 cells in a setting of asthma. Finally, local delivery of an anti-IL-6R antibody in T-bet((-/-)) mice resulted in the resolution of this allergic trait. Noteworthy, BATF, crucial for the immunoglobulin-class-switch and TH2,TH17 development, was found down-regulated in the lungs of T-bet((-/-)) mice after SIT and after treatment with anti-IL-6R antibody, indicating a critical role of IL-6 in controlling BATF/IRF4 integrated functions in TH2, TH17 cells and B cells also in a T-bet independent fashion in allergic asthma

    Thermally assisted magnetization reversal in the presence of a spin-transfer torque

    We propose a generalized stochastic Landau-Lifshitz equation and its corresponding Fokker-Planck equation for the magnetization dynamics in the presence of spin transfer torques. Since the spin transfer torque can pump a magnetic energy into the magnetic system, the equilibrium temperature of the magnetic system is ill-defined. We introduce an effective temperature based on a stationary solution of the Fokker-Planck equation. In the limit of high energy barriers, the law of thermal agitation is derived. We find that the N\'{e}el-Brown relaxation formula remains valid as long as we replace the temperature by an effective one that is linearly dependent of the spin torque. We carry out the numerical integration of the stochastic Landau-Lifshitz equation to support our theory. Our results agree with existing experimental data.Comment: 5 figure

    Mind The Gap Between The Fracture Line And The Length Of The Working Area: A Finite Element Analysis Using An Extramedullary Fixation Model

    To determine the ideal working area for a simple transverse fracture line treated with a bridge plate. Methods: A 2-D finite element analysis of a hypothetical femur was performed for the quantitative evaluation of a large-fragment titanium alloy locking plate based on the principle of relative stability in a case of a simple transverse diaphyseal fracture. Two simulations (one case of strain and another case of stress distribution) were analyzed in three unique situations according to the von Mises stress theory. Load distributions were observed when the bone was subjected to a single vertical load of 1,000. N. Results: The longer the length of the implant flexion, which coincided with the working area of the plate, the greater the flexion of the implant. The highest concentrations of stress on the plate occurred in the region around the screws closest to the bone gap. The closer the screws to the fracture site, the greater the demands on the plate. Conclusion: When using a large-fragment titanium alloy locking plate to stabilize a simple transverse fracture based on the principle of relative stability (bridge plate), the distance between the proximal and distal screws closest to the fracture line must be long. The farther away this fixation is, the lower the stress on the plate and the greater the dissipation of force in the form of deflection. © 2017 Sociedade Brasileira de Ortopedia e Traumatologia

    Finite Element Analysis Of The Equivalent Stress Distribution In Schanz Screws During The Use Of A Femoral Fracture Distractor

    To evaluate the mechanical stress and elastic deformation exercised in the thread/shaft transition of Schanz screws in assemblies with different screw anchorage distances in the entrance to the bone cortex, through the distribution and location of tension in the samples.An analysis of 3. D finite elements was performed to evaluate the distribution of the equivalent stress (triple stress state) in a Schanz screw fixed bicortically and orthogonally to a tubular bone, using two mounting patterns: (1) thread/shaft transition located 20. mm from the anchorage of the Schanz screws in the entrance to the bone cortex and (2) thread/shaft transition located 3. mm from the anchorage of the Schanz screws in entrance to the bone cortex. The simulations were performed maintaining the same direction of loading and the same distance from the force vector in relation to the center of the hypothetical bone. The load applied, its direction, and the distance to the center of the bone were constant during the simulations in order to maintain the moment of flexion equally constant. The present calculations demonstrated linear behavior during the experiment. It was found that the model with a distance of 20. mm between the Schanz screws anchorage in the entrance to the bone cortex and the thread/shaft transition reduces the risk of breakage or fatigue of the material during the application of constant static loads; in this model. the maximum forces observed were higher (350. Mpa). The distance between the Schanz screws anchorage at the entrance to the bone cortex and the smooth thread/shaft transition of the screws used in a femoral distractor during acute distraction of a fracture must be farther from the entrance to the bone cortex, allowing greater degree of elastic deformation of the material, lower mechanical stress in the thread/shaft transition, and minimized breakage or fatigue.     Momentum--dependent nuclear mean fields and collective flow in heavy ion collisions

    We use the Boltzmann-Uehling-Uhlenbeck model to simulate the dynamical evolution of heavy ion collisions and to compare the effects of two parametrizations of the momentum--dependent nuclear mean field that have identical properties in cold nuclear matter. We compare with recent data on nuclear flow, as characterized by transverse momentum distributions and flow (FF) variables for symmetric and asymmetric systems. We find that the precise functional dependence of the nuclear mean field on the particle momentum is important. With our approach, we also confirm that the difference between symmetric and asymmetric systems can be used to pin down the density and momentum dependence of the nuclear self consistent one--body potential, independently. All the data can be reproduced very well with a momentum--dependent interaction with compressibility K = 210 MeV.Comment: 15 pages in ReVTeX 3.0; 12 postscript figures uuencoded; McGill/94-1

    We develop a complete and consistent description for the hadron spectra from heavy ion collisions in terms of a few collective variables, in particular temperature, longitudinal and transverse flow. To achieve a meaningful comparison with presently available data, we also include the resonance decays into our picture.     Semiclassical Trace Formulas for Noninteracting Identical Particles

    We extend the Gutzwiller trace formula to systems of noninteracting identical particles. The standard relation for isolated orbits does not apply since the energy of each particle is separately conserved causing the periodic orbits to occur in continuous families. The identical nature of the particles also introduces discrete permutational symmetries. We exploit the formalism of Creagh and Littlejohn [Phys. Rev. A 44, 836 (1991)], who have studied semiclassical dynamics in the presence of continuous symmetries, to derive many-body trace formulas for the full and symmetry-reduced densities of states.     Thermal rates for baryon and anti-baryon production

    We use a form of the fluctuation-dissipation theorem to derive formulas giving the rate of production of spin-1/2 baryons in terms of the fluctuations of either meson or quark fields. The most general formulas do not assume thermal or chemical equilibrium.
