    Effect of acute copper sulfate exposure on olfactory responses to amino acids and pheromones in goldfish (Carassius auratus)

    Exposure of olfactory epithelium to environmentally relevant concentrations of copper disrupts olfaction in fish. To examine the dynamics of recovery at both functional and morphological levels after acute copper exposure, unilateral exposure of goldfish olfactory epithelia to 100 μM CuSO4 (10 min) was followed by electro-olfactogram (EOG) recording and scanning electron microscopy. Sensitivity to amino acids (L-arginine and L-serine), generally considered food-related odorants, recovered most rapidly (three days), followed by that to catecholamines(3-O-methoxytyramine),bileacids(taurolithocholic acid) and the steroid pheromone, 17,20 -dihydroxy-4-pregnen- 3-one 20-sulfate, which took 28 days to reach full recovery. Sensitivity to the postovulatory pheromone prostaglandin F2R had not fully recovered even at 28 days. These changes in sensitivity were correlated with changes in the recovery of ciliated and microvillous receptor cell types. Microvillous cells appeared largely unaffected by CuSO4 treatment. Cilia in ciliated receptor neurones, however, appeared damaged one day post-treatment and were virtually absent after three days but had begun to recover after 14 days. Together, these results support the hypothesis that microvillous receptor neurones detect amino acids whereas ciliated receptor neurones were not functional and are responsible for detection of social stimuli (bile acidsandpheromones).Furthermore, differences in sensitivity to copper may be due to different transduction pathways in the different cell types

    Technological Change in Resource Extraction and Endogenous Growth

    We add an extractive sector to an endogenous growth model of expanding varieties and directed technological change. Extractive firms reduce the stock of non-renewable resources through extraction, but also increase the stock through R&D investment in extraction technology. Our model replicates long-term trends in non-renewable resource markets, namely stable prices and exponentially increasing extraction, for which we present data from 1792 to 2009. The model suggests that the development of new extraction technologies neutralizes the increasing demand for non-renewable resources in industrializing countries like China in the long term

    Measurement of stellar age from uranium decay

    The ages of the oldest stars in the Galaxy indicate when star formation began, and provide a minimum age for the Universe. Radioactive dating of meteoritic material and stars relies on comparing the present abundance ratios of radioactive and stable nuclear species to the theoretically predicted ratios of their production. The radioisotope 232^{232}Th (half-life 14 Gyr) has been used to date Galactic stars, but it decays by only a factor of two over the lifetime of the Universe. 238^{238}U (half-life 4.5 Gyr) is in principle a more precise age indicator, but even its strongest spectral line, from singly ionized uranium at a wavelength of 385.957 nm, has previously not been detected in stars. Here we report a measurement of this line in the very metal-poor star CS31082-001, a star which is strongly overabundant in its heavy elements. The derived uranium abundance, log(U/H) = -13.7+/-0.14+/-0.12 yields an age of 12.5+/-3 Gyr, though this is still model dependent. The observation of this cosmochronometer gives the most direct age determination of the Galaxy.     Mycorrhizal occurrence in willows in a northern freshwater wetland

    Willows growing in a north central Michigan wetland were found to be ectomycorrhizal throughout the 1978 growing season on each of two sites (one water-saturated, one relatively drier). Each site was dominated by a mix of sedges ( Carex spp.) and willows ( Salix spp.). On both sites, phosphorus was added at two levels (20 or 200 kg P ha −1 ) to simulate potential inputs from the disposal of secondarily-treated municipal wastewater.     Engineering HIV-Resistant Human CD4+ T Cells with CXCR4-Specific Zinc-Finger Nucleases

    HIV-1 entry requires the cell surface expression of CD4 and either the CCR5 or CXCR4 coreceptors on host cells. Individuals homozygous for the ccr5Δ32 polymorphism do not express CCR5 and are protected from infection by CCR5-tropic (R5) virus strains. As an approach to inactivating CCR5, we introduced CCR5-specific zinc-finger nucleases into human CD4+ T cells prior to adoptive transfer, but the need to protect cells from virus strains that use CXCR4 (X4) in place of or in addition to CCR5 (R5X4) remains. Here we describe engineering a pair of zinc finger nucleases that, when introduced into human T cells, efficiently disrupt cxcr4 by cleavage and error-prone non-homologous DNA end-joining. The resulting cells proliferated normally and were resistant to infection by X4-tropic HIV-1 strains. CXCR4 could also be inactivated in ccr5Δ32 CD4+ T cells, and we show that such cells were resistant to all strains of HIV-1 tested. Loss of CXCR4 also provided protection from X4 HIV-1 in a humanized mouse model, though this protection was lost over time due to the emergence of R5-tropic viral mutants. These data suggest that CXCR4-specific ZFNs may prove useful in establishing resistance to CXCR4-tropic HIV for autologous transplant in HIV-infected individuals

    The Ultra-Fast Outflow of the Quasar PG 1211+143 as Viewed by Time-Averaged Chandra Grating Spectroscopy

The Version of Record is available online at https://doi.org/10.3847/1538-4357/aaa427We present a detailed X-ray spectral study of the quasar PG 1211+143 based on Chandra High Energy Transmission Grating Spectrometer (HETGS) observations collected in a multi-wavelength campaign with UV data using the Hubble Space Telescope Cosmic Origins Spectrograph (HST-COS) and radio bands using the Jansky Very Large Array (VLA). We constructed a multi-wavelength ionizing spectral energy distribution using these observations and archival infrared data to create xstar photoionization models specific to the PG 1211+143 flux behavior during the epoch of our observations. Our analysis of the Chandra-HETGS spectra yields complex absorption lines from H-like and He-like ions of Ne, Mg, and Si, which confirm the presence of an ultra-fast outflow (UFO) with a velocity of approximately -17,300 km s -1 (outflow redshift z out ∼ -0.0561) in the rest frame of PG 1211+143. This absorber is well described by an ionization parameter and column density. This corresponds to a stable region of the absorber's thermal stability curve, and furthermore its implied neutral hydrogen column is broadly consistent with a broad Lyα absorption line at a mean outflow velocity of approximately -16,980 km s -1 detected by our HST-COS observations. Our findings represent the first simultaneous detection of a UFO in both X-ray and UV observations. Our VLA observations provide evidence for an active jet in PG 1211+143, which may be connected to the X-ray and UV outflows; this possibility can be evaluated using very-long-baseline interferometric observations.Peer reviewedFinal Accepted Versio

    Building Dynamic Capabilities: Innovation Driven By Individual, Firm, and Network Level Effects

    Following the dynamic capabilities perspective, we suggest that antecedents to innovation can be found at the individual, firm, and network level. Thus, we challenge two assumptions common in prior research: (1) that significant variance exists at the focal level of analysis, while other levels of analysis are assumed to be homogeneous, and (2) that the focal level of analysis is independent from other levels of analysis. Accordingly, we advance a set of hypotheses to simultaneously assess the direct effects of antecedents at the individual, firm, and network level on innovation output. We then investigate whether a firm’s antecedents to innovation lie across different levels. To accomplish this, we propose two competing interaction hypotheses. We juxtapose the hypothesis that the individual, firm, and network level antecedents to innovation are substitutes versus the proposition that these innovation mechanisms are complements. We test our multi-level theoretical model using an unusually comprehensive and detailed panel dataset that documents the innovation attempts of global pharmaceutical companies within biotechnology over a 22-year time period (1980-2001). We combine these data with direct field observations conducted prior to, during, and after the completion of the study. We find evidence that the antecedents to innovation lie across different levels of analysis and can have compensating or reinforcing effects on firm-level innovative output

    Therapeutic efficacy of TBC3711 in monocrotaline-induced pulmonary hypertension

    Background: Endothelin-1 signalling plays an important role in pathogenesis of pulmonary hypertension. Although different endothelin-A receptor antagonists are developed, a novel therapeutic option to cure the disease is still needed. This study aims to investigate the therapeutic efficacy of the selective endothelin-A receptor antagonist TBC3711 in monocrotaline-induced pulmonary hypertension in rats. Methods: Monocrotaline-injected male Sprague-Dawley rats were randomized and treated orally from day 21 to 35 either with TBC3711 (Dose: 30 mg/kg body weight/day) or placebo. Echocardiographic measurements of different hemodynamic and right-heart hypertrophy parameters were performed. After day 35, rats were sacrificed for invasive hemodynamic and right-heart hypertrophy measurements. Additionally, histologic assessment of pulmonary vascular and right-heart remodelling was performed. Results: The novel endothelin-A receptor antagonist TBC3711 significantly attenuated monocrotaline-induced pulmonary hypertension, as evident from improved hemodynamics and right-heart hypertrophy in comparison with placebo group. In addition, muscularization and medial wall thickness of distal pulmonary vessels were ameliorated. The histologic evaluation of the right ventricle showed a significant reduction in fibrosis and cardiomyocyte size, suggesting an improvement in right-heart remodelling. Conclusion: The results of this study suggest that the selective endothelin-A receptor antagonist TBC3711 demonstrates therapeutic benefit in rats with established pulmonary hypertension, thus representing a useful therapeutic approach for treatment of pulmonary hypertension
